<P>Patterns of salivary HCO3<SUP>−</SUP>secretion vary and depend on species and gland types. However, the identities of the transporters involved in HCO3<SUP>−</SUP>transport and the underlying mechanism of intracellular pH (pHi) regulation in salivary glands still remain unclear. In this study, we examined the expression of the Na<SUP>+</SUP>-HCO3<SUP>−</SUP>cotransporter (NBC) and its role in pHiregulation in guinea pig salivary glands, which can serve as an experimental model to study HCO3<SUP>−</SUP>transport in human salivary glands. RT-PCR, immunohistochemistry, and pHimeasurements from BCECF-AM-loaded cells were performed. The amiloride-sensitive Na<SUP>+</SUP>/H<SUP>+</SUP>exchanger (NHE) played a putative role in pHiregulation in salivary acinar cells and also appeared to be involved in regulation in salivary ducts. In addition to NHE, NBC also played a role in pHiregulation in both acini and ducts. In the parotid gland, NBC1 was functionally expressed in the basolateral membrane (BLM) of acinar cells and the luminal membrane (LM) of ducts. In the submandibular gland, NBC1 was expressed only in the BLM of ducts. NBC1 expressed in these two types of salivary glands takes up HCO3<SUP>−</SUP>and is involved in pHiregulation. Although NBC3 immunoreactivity was also detected in submandibular gland acinar cells and in the ducts of both glands, it is unlikely that NBC3 plays any role in pHiregulation. We conclude that NBC1 is functionally expressed and plays a role in pHiregulation in guinea pig salivary glands but that its localization and role are different depending on the type of salivary glands.</P>
Purification and Characterization of an Extracellular Protease from Bacillus pumilus CN8

The protease produced by a Bacillus pumilus CN8 strain was purified by DEAE-Cellulose-52 ion exchange. It has a molecular weight of approximately 96,920 Dalton. In the present study, this protease showed strong activity over a broad range of pH (6.5-9.5) and temperature from 40℃ to 60℃, and the protease performed the maximal activity at pH 7.3 at 42℃. The effect of metal ions on protease activity showed that K+ could slightly increase the protease activity, and other ions such as Zn²+, Fe²+, Na+, Ca²+, Mg²+ had no significant activation or inhibition to the protease (P > 0.05), and the more important is that Cu²+, Mn²+, Sn²+, Cd²+ had a strong inhibitory effect on the protease activity.

<P><B>Abstract</B></P> <P>In this work, we developed the perovskite photodiodes based on the dimensionality-reduced quasi two-dimensional (Q-2D) photoactive layer structure by incorporating phenylethylammonium iodide (PEAI) into methylammonium lead iodide (MAPbI<SUB>3</SUB>), which effectively enhanced both the crystalline phase and the ambient stability of the perovskite. The Q-2D perovskite photodiode exhibited a dark current of 1.76 × 10<SUP>−7</SUP> A/cm<SUP>2</SUP>, resulting in the detectivity (D*) of 2.20 × 10<SUP>12</SUP> J and responsivity of 0.53 A/W, which is among the highest performance levels without the voltage bias (0 V) due to the systematically optimized perovskite phase resulting in the reduced leakage current. In addition, the current density of Q-2D perovskite photodiode maintained 76% of the initial level current density even after 80 days in the ambient condition, compared to 15% of 3D perovskite photodiode control sample. Such superior performance and stability were mainly attributed to the enhanced degree of crystallization of the Q-2D perovskites, which was confirmed by X-ray diffraction and grazing incidence wide-angle X-ray scattering (GIWAXS) measurement. Also, the improved stability of Q-2D perovskite films was confirmed by both lifetime test and atomic force microscopy studies where the negligible number of pinholes was observed in the quasi-2D perovskite films while considerable deformations were found in the 3D perovskites photodiode. Our study suggests a simple and robust protocol for the development of stable and high-performance perovskite photodetectors via dimensional and constitutional optimization of conventional perovskites for the practical usage of perovskite in the photodiode applications.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The Q-2D perovskite photodiode exhibited the D* of 2.20 × 10<SUP>12</SUP> J and R of 0.53 A/W without the voltage bias (0 V). </LI> <LI> The current density of Q-2D perovskite photodiode maintained 76 % of the initial level while 15 % for the 3D one. </LI> <LI> Grazing incidence wide-angle X-ray scattering (GIWAXS) analysis revealed the origin of the stability improvement. </LI> <LI> Quasi-2D perovskite materials can be promising candidates for stable, tunable and flexible optoelectronic applications. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>Dimensionality-controlled perovskite photodiodes with improved stability were systematically fabricated while retaining the comparable electrical performance of conventional three-dimensional perovskites. <P><B>Abstract</B></P> <P>Developing perovskite solar cells (PSCs) with high-efficiency and a low-temperature process has great potential for the realization of the scalable, economic, and roll-to-roll based renewable energy conversion devices. ZnO has been recognized as the promising electron transport layer (ETL) that may overcome the limitations of the conventional TiO<SUB>2</SUB>. However, methylammonium lead iodide (MAPbI<SUB>3</SUB>) perovskite is prone to decomposition at the ZnO surface, which hinders the development of simple one-step deposition of perovskite, resulting in the limited photovoltaic performance. Herein, we report interlayer mediated efficient ETLs for viable flexible PSCs. The utilization of polyethyleneimine ethoxylated layer on ZnO prevents the direct contact between the perovskite and ETLs, avoiding the photoactive layer decomposition. Thus, interlayer-mediated PSCs show higher efficiency with enhanced stability by decreasing the electron transport barrier. As a result, the PSC employing tailor-designed ETL interfaces exhibited average power conversion efficiency (PCE) of 15.8%, which was superior by 25.4% to that of a control device (12.6%). With our strategy, we further demonstrated PSCs on flexible substrates which exhibit an average PCE of 11.9% under low-temperature fabrication. Background/AimsWe evaluated the clinical significance and prognostic power of functional luminal imaging probe (FLIP) panometry in patients with achalasia treated with peroral endoscopic myotomy (POEM), and examined the clinical parameters associated with symptomatic improvement and the presence of contractility (POC) following POEM. MethodsWe reviewed the electronic medical records of patients with achalasia treated with FLIP panometry and POEM at a tertiary teaching hospital in Seoul, Republic of Korea. Follow-up examination was composed of esophageal manometry and questionnaires on symptoms. We analyzed the FLIP data by interpolating using the cubic spline method in MATLAB. ResultsWe retrospectively analyzed 33 men and 35 women (mean age: 52 ± 17 years), of whom 14, 39, and 15 patients were diagnosed with achalasia types I, II, and III, respectively. The FLIP panometry diagnoses were reduced esophagogastric junction opening (REO) with a retrograde contractile response (n = 43); REO with an absent contractile response (n = 5); REO with a normal contractile response (n = 11); and a retrograde contractile response (n = 9). Overall, the patients showed improvements in Eckardt scores following POEM from 6.48 ± 2.20 to 1.16 ± 1.15 (P < 0.01). Post-POEM symptomatic improvement was not significantly associated with any of the clinical parameters, including panometry diagnosis. Conversely, post-POEM POC was significantly associated with the presence of repetitive antegrade contractions and achalasia subtypes (both P < 0.01). ConclusionWhile FLIP panometry was not significantly associated with the clinical course of achalasia, FLIP panometry was associated with POC following POEM and may complement manometry in the functional evaluation of esophageal motility disorders.

The cytotoxic mechanism of protein-bound polysaccharide isolated from Phellinus linteus (PL, Mesim_(??)) has been investigated. PL inhibited the proliferation and colony formation of SW480 human colon cancer cells. Flow cytometry analysis showed that PL increased the populations of both apoptotic sub-G_(1) and G_(2)/M phase. The result obtained from TUNEL assay corroborated apoptosis which was shown in flow cytometry. Western blot analysis suggested that PL-induced apoptosis and growth inhibition were associated with decrease in Bcl-2, increase of the release of cytochrome c, and reduced expression of cyclin B1. These results suggest that PL has a direct antitumor effect through apoptosis and cell cycle blockade in certain cancer cells.

요독성 심낭염은 만성 선부전 환자에서 나타날 수 있는 합병증으로 투석치료의 도입이후 발생률이 감소하고 있으나 말기 산부전 환자의 투석시작 전후로 혈역학적 문제가 발생할 수 있는 심각한 합병증 중 하나이며 드물게 오랜 기간 투석 치료를 받은 환자에서도 발생할 수 있다. 요독성 심낭염이 진행되어 심낭삼출이 발생할 경우 기저 질환에 대한 치료와 요독의 효과적인 제거를 위한 고효율의 투석을 시행하지만 혈역학적으로 불안정 할 경우 심낭천자 및 심막절제술의 침습적인 치료가 필요하다. 저자들은 오랜 기간 동안 혈액투석을 시행받은 환자에서 발생한 만성 요독성 교착성 심낭염을 경험하여 이를 보고하려고 한다. 63세 남자 환자가 혈액 투석 중에 발생한 저혈압과 호흡 곤란으로 내원하였다. 2달 전 부터 투석 중 흉부 압박감이 있었고, 1 주 일 전부터는 투석 중 저혈압이 발생하고 호흡곤란이 동반되었다고 한다. 고혈압에 의한 말기신부전으로 9년 전부터 혈액 투석을 받고 있었다. 흉부 방사선 단순촬영에서 섬비대가 있었고, 심장 초음파와 흉부 컴퓨터 단층 촬영에서 교착성 심낭염을 발견하였다. 이후 섬막절제술을 시행하였고, 병리학적 소견으로 보아 만성 요독성 교착성 심낭염을 진단할 수 있었다. 환자는 수술 후 혈압은 정상이었고, 투석 중에도 저혈압은 발생하지 않았다. 현재 외래에서 추적관찰 중이다.
