BACKGROUND/OBJECTIVES: The accumulation of amyloid-β (Aβ) in the brain is a hallmark of Alzheimer’s disease (AD) and plays a key role in cognitive dysfunction. Perilla frutescens var. japonica extract (PFE) and its major compound, rosmarinic acid (RA), have shown antioxidant and anti-inflammatory activities. We investigated whether administration of PFE and RA contributes to cognitive improvement in an Aβ25-35-injected mouse model. MATERIALS/METHODS: Male ICR mice were intracerebroventricularly injected with aggregated Aβ25-35 to induce AD. Aβ25-35-injected mice were fed PFE (50 mg/kg/day) or RA (0.25 mg/kg/day) for 14 days and examined for learning and memory ability through the T-maze, object recognition, and Morris water maze test. RESULTS: Our present study demonstrated that PFE and RA administration significantly enhanced cognition function and object discrimination, which were impaired by Aβ25-35, in the T-maze and object recognition tests, respectively. In addition, oral administration of PFE and RA decreased the time to reach the platform and increased the number of crossings over the removed platform when compared with the Aβ25-35-induced control group in the Morris water maze test. Furthermore, PFE and RA significantly decreased the levels of nitric oxide (NO) and malondialdehyde (MDA) in the brain, kidney, and liver. In particular, PFE markedly attenuated oxidative stress by inhibiting production of NO and MDA in the Aβ25-35-injected mouse brain. CONCLUSIONS: These results suggest that PFE and its active compound RA have beneficial effects on cognitive improvement and may help prevent AD induced by Aβ.
Emerging Japanese Encephalitis Virus Genotype V in Republic of Korea

Japanese encephalitis (JE) is a vaccine-preventable mosquito-borne disease caused by infection with the Japanese encephalitis virus (JEV). JEV has five genotypes, including genotype V (GV), which is considered ancestral to the other genotypes. The first GV strain, GV Muar, was isolated from a Malayan patient in 1952 and GV did not reappear for 57 years until GV XZ0934 was isolated from a mosquito sample in China. Since 2010, 21 GV strains have been identified in Republic of Korea (ROK). Both GV Muar and GV XZ0934 are more pathogenic than other GI/GIII strains and are serologically distinct. However, because the ROK's GV strains have not been experimentally tested, their characteristics are not known. Characterization of the ROK's isolates is needed to enable development of effective GV strain-based vaccines to protect against GV infections.
Learning, memory deficits, and impaired neuronal maturation attributed to acrylamide

<P>Acrylamide (ACR) is a neurotoxin known to produce neurotoxicity characterized by ataxia, skeletal muscle weakness, cognitive impairment, and numbness of the extremities. Previously, investigators reported that high-dose (50mg/kg) ACR impaired hippocampal neurogenesis and increased neural progenitor cell death; however, the influence of subchronic environmentally relevant low dose-(2, 20, or 200g/kg) ACRs have not been examined in adult neurogenesis or cognitive function in mice. Accordingly, the aim of the present study was to investigate whether low-dose ACR adversely affected mouse hippocampal neurogenesis and neurocognitive functions. Male C57BL/6 mice were orally administered vehicle or ACR at 2, 20, or 200g/kg/day for 4weeks. ACR did not significantly alter the number of newly generated cells or produce neuroinflammation or neuronal loss in hippocampi. However, behavioral studies revealed that 200g/kg ACR produced learning and memory impairment. Furthermore, incubation of ACR with primary cultured neurons during the developmental stage was found to delay neuronal maturation without affecting cell viability indicating the presence of developmental neurotoxicity. These findings indicate that although exposure to in vivo low-dose ACR daily for 4weeks exerted no apparent marked effect on hippocampal neurogenesis, in vitro observations in primary cultured neurons noted adverse effects on learning and memory impairment suggestive of neurotoxic actions.</P>
Characterization of physiologic 18F-FDG uptake with PET-CT in dogs.

<P>We evaluated the whole body distribution of 2-deoxy-2-[18F]fluoro-D-glucose (18F-FDG) in seven beagle dogs using positron emission tomography/computed tomography. The mean and maximum standard uptake values (SUV) for various tissues were computed. The SUV of the aortic blood pool was 0.65 +/- 0.19. Moderate uptake was present in brain (3.40 +/- 1.01). Mild uptake was present in orbital muscles, soft palate, laryngeal and pharyngeal region, mandibular salivary gland, myocardium, liver, pancreas, kidney, and intestine. 18F-FDG uptake would be normally higher in these tissues because of normal physiologic activity. Mean and maximum SUV values of the eye, skeletal muscle, bone tissue, spleen, adrenal gland, stomach, tongue, gall bladder, and lung were similar to or lower than that of the aortic blood pool. These data provide a normal baseline for comparing pathologic 18F-FDG uptake.</P>
Physicochemical Composition and Antioxidative Effects of Yacon (Polymnia Sonchifolia)

야콘의 생리활성 기능과 이용 가능성에 관한 연구의 일환으로 야콘 분말의 영양성분 및 항산화 활성을 측정한 결과는 다음과 같다. 야콘 분말의 일반성분은 건물량(dry matter basis)을 기준으로 수분 함량은 3.53%, 조단백질 1.13%, 조지방 0.40%, 조회분 0.79%, 식이섬유소 1.63% 및 탄수화물 92.52%를 함유하였다. 구성당은 fructose가 1320.84 ㎎/l로 가장 많았고 glucose가 1283.70 ㎎/l 검출되어 총 2종이 검출되었다. 아미노산은 glutamic acid 함량이 123.12 ㎎%로 가장 높았고, 다음으로 aspartic acid, proline, arginine, alanine, valine, histidine, lysine순이었다. 구성 지방산 중 포화지방산인 palmitic acid와 lauric acid 2종류의 지방산만 각각 51.72%와 48.28%로 검출되었으며, 불포화지방산은 검출되지 않았다. 유기산은 총 3종의 유기산이 검출되었으며, 이 중 oxalic acid가 2519.29 ㎎/l로 가장 많았고, 다음으로 benzoic acid 1895.17 ㎎/l, formic acid 247.84 ㎎/l 순이었다. 비타민 A, C 및 E의 함량은 각각 0.057 ㎎%, 0.670 ㎎% 및 0.001 ㎎%로 검출되었으며, 무기질은 총 5종의 무기질 성분이 검출되었고, 이 중 K 함량이 가장 많았으며, 다음으로 Na, Ca, Mg 순이었고 Zn의 함량은 미량이었다. 야콘 에탄올 추출물을 n-hexane, chloroform, ethylacetate, n-butanol, water로 계통 분획하여 항산화 활성을 측정한 결과 Rancimat으로 측정한 항산화지수는 pH 1.2 조건에서 분획 중 ethylacetate 분획이 1.45로 가장 높게 나타났으나 양성대조군으로 사용한 BHT 1.83에 비해서는 낮은 활성을 나타내었다. 아질산염 소거능에서도 ethylacetate 분획이 49.82%로 분획 중 가장 높은 아질산염 소거능을 보였으나, 양성대조군인 BHT의 75.29%보다는 낮게 나타났다. 이상의 결과 야콘은 체내 신진대사와 생리활성을 증진시킬 수 있는 영양소들이 다량 함유하고 있으며 항산화 효과를 나타내는 물질을 함유하고 있는 것으로 판단되어 야콘의 기능성 식품으로써의 이용화가치가 한층 더 높아질 것으로 기대해 본다. This study was carried out to investigate the physicochemical and functional properties of Yacon (Polymnia Sonchifolia) powder. The proximate composition of Yacon powder as a dry matter basis was 3.53% moisture content, 1.13% crude protein, 0.40% crude fat, 0.79% crude ash, 1.63% dietary fiber and 92.52% carbohydrate. The major free sugars were identified as fructose and glucose. Analysing total amino acids, 18 kinds of components were isolated from Yacon powder. The essential amino acid contained in Yacon powder accounted for 28.40% of total amino acid, while the non-essential amino acid accounted for 73.61%. Analysing total fatty acids, only 2 kinds - palmitic acid and lauric acid - were detected. Oxalic acid was the major organic acid. The contents of vitamin A, vitamin C and vitamin E were 0.057 mg%, 0.670 mg% and 0.001 mg%, respectively. The mineral contents of Yacon powder were in the order of Zn<Mg<Ca<Na<K. Antioxidant index by Rancimat test and nitrite scavenging ability of ethylacetate fraction of Yacon ethanol extract were the highest among fractions and were less than that of BHT. These results suggest that Yacon is recommendable as a vegetable of high nutritional quality and possesses potential antioxidative capacity.

Neurodegenerative diseases are often associated with oxidative damage in neuronal cells. This study was conducted to investigate the neuro-protective effect of methanolic (MeOH) extract of Perilla frutescens var. japonica and its one of the major compounds, rosmarinic acid, under oxidative stress induced by hydrogen peroxide (H2O2) in C6 glial cells. Exposure of C6 glial cells to H2O2 enhanced oxidative damage as measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and thiobarbituric acid-reactive substance assays. The MeOH extract and rosmarinic acid prevented oxidative stress by increasing cell viability and inhibiting cellular lipid peroxidation. In addition, the MeOH extract and rosmarinic acid reduced H2O2-induced expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) at the transcriptional level. Moreover, iNOS and COX-2 protein expression was down-regulated in H2O2-indcued C6 glial cells treated with the MeOH extract and rosmarinic acid. These findings suggest that P. frutescens var. japonica and rosmarinic acid could prevent the progression of neurodegenerative diseases through attenuation of neuronal oxidative stress.
Effects of anesthetic protocol on normal canine brain uptake of 18F-FDG assessed by PET/CT.

<P>The purpose of this study was to assess the effects of four anesthetic protocols on normal canine brain uptake of 2-deoxy-2-[18F]fluoro-D-glucose (FDG) using positron emission tomography/computed tomography (PET/CT). Five clinically normal beagle dogs were anesthetized with (1) propofol/isoflurane, (2) medetomidine/pentobarbital, (3) xylazine/ketamine, and (4) medetomidine/tiletamine-zolazepam in a randomized cross-over design. The standard uptake value (SUV) of FDG was obtained in the frontal, parietal, temporal and occipital lobes, cerebellum, brainstem and whole brain, and compared within and between anesthetic protocols using the Friedman test with significance set at P < 0.05. Significant differences in SUVs were observed in various part of the brain associated with each anesthetic protocol. The SUV for the frontal and occipital lobes was significantly higher than in the brainstem in all dogs. Dogs receiving medetomidine/tiletamine-zolazepam also had significantly higher whole brain SUVs than the propofol/isoflurane group. We concluded that each anesthetic protocol exerted a different regional brain glucose uptake pattern. As a result, when comparing brain glucose uptake using PET/CT, one should consider the effects of anesthetic protocols on different regions of the glucose uptake in the dog's brain.</P>
