The fruits of Poncirus trifoliata (L.) are widely used in Oriental medicine as a remedy for allergic inflammation. As a part of our program to screen medicinal plants for potential anti-inflammatory compounds, 21α-methylmelianodiol (21α-MMD) and 21β-methylmelianodiol (21β-MMD), which are two isomers of 21-methylmelianodiol isolated from the fruits of P. trifoliata for the first time, were found to inhibit nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. 21α-MMD and 21β-MMD attenuated LPS-induced inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 protein expressions as well as the mRNA levels of iNOS, COX-2, tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β). To investigate the mechanism involved, we examined the effect of 21α-MMD and 21β-MMD on LPS-induced nuclear factor-κB (NF-κB) activation. Both 21α-MMD and 21β-MMD significantly inhibited LPS-induced NF-κB transcriptional activity in RAW 264.7 macrophages. Moreover, the in vivo anti-inflammatory effect of 21α-MMD was examined in two mouse models of acute inflammation. In the carrageenan-induced paw edema model, administration of 21α-MMD (20 and 100 mg/kg, i.p.) dose-dependently reduced paw swelling. In addition, 21α-MMD significantly inhibited the dye leakage in an acetic acid-induced vascular permeability assay. Taken together, our data indicate that 21-methylmelianodiol is an important constituent of the fruit of P. trifoliata, and that the inhibition of iNOS and COX-2 expression by 21α-MMD and 21β-MMD might be one of the mechanisms responsible for their anti-inflammatory effects.

<P>A Fredholm integral equation of the first kind has been developed for analyzing the performance of the toroidal high-voltage direct current (HVDC) grounding system buried in the horizontal multilayer soil. The method of moment is used to solve the equations. The formulation of potential in the earth due to a circular current is proposed in closed form by means of conception of the complex resistivity and depth, which leads to the saving of the computational time greatly. The numerical results show the good agreements with those in other papers and the measured data</P>

<P>X-linked chronic granulomatous disease (X-CGD) is an immune deficiency resulting from defective production of microbicidal reactive oxygen species (ROS) by phagocytes. Causative mutations occur throughout the CYBB gene, resulting in absent or defective gp91(phox) protein expression. To correct CYBB exon 5 mutations while retaining normal gene regulation, we utilized TALEN or Cas9 for exon 5 replacement in induced pluripotent stem cells (iPSCs) from patients, which restored gp91phox expression and ROS production in iPSC derived granulocytes. Alternate approaches for correcting the majority of X-CGD mutations were assessed, involving TALEN- or Cas9-mediated insertion of CYBB minigenes at exon 1 or 2 of the CYBB locus. Targeted insertion of an exon 1-13 minigene into CYBB exon 1 resulted in no detectable gp91phox expression or ROS activity in iPSC-derived granulocytes. In contrast, targeted insertion of an exon 2-13 minigene into exon 2 restored both gp91phox and ROS activity. This demonstrates the efficacy of two correction strategies: seamless repair of specific CYBB mutations by exon replacement or targeted insertion of an exon 2-13 minigene to CYBB exon 2 while retaining exon/intron 1. Furthermore, it highlights a key issue for targeted insertion strategies for expression from an endogenous promoter: retention of intronic elements can be necessary for expression.</P>

The experiment was conducted to investigate the effects of dihydropyridine on laying performance and fat metabolism of laying hens. Five hundred and forty laying hens, 40 weeks old, were randomly allotted to three groups, each of which included four replicates of 45 hens. The groups were given a basal corn-soybean meal diet supplemented with 0, 150 mg/kg and 300 mg/kg dihydropyridine. Results showed that compared with the control group (0 mg/kg dihydropyridine), supplements of 150 and 300 mg/kg dihydropyridine increased egg production rate by 9.39% (p<0.01) and 12.97% (p<0.01), increased mean egg weight by 3% (p>0.05) and 4.8% (p>0.05), and improved feed efficiency by 9.54% (p<0.05) and 7.25% (p<0.05), respectively; The addition of 150 and 300 mg/kg dihydropyridine decreased percentage of abdominal fat by 35.4% (p<0.05) and 46.9% (p<0.05), decreased liver fat content by 32.4% (p<0.05) and 10.5% (p<0.05), increased HSL activity of abdominal fat by 39.64% (p<0.05) and 48.48% (p<0.05), increased HSL activity of liver by 9.4% (p>0.05) and 47.34% (p<0.05) and increased the content of cAMP in adenohypophysis by 14.67% (p<0.05) and 10.91% (p<0.05), respectively; The inclusion of 150 mg/kg dihydropyridine increased liver superoxide dismutase activity by 69.61% (p<0.05), and increased hepatic apoB concentration by 53.96% (p<0.05); The supplementation of 150 or 300 mg/kg dihydropyridine decreased malondialdehyde concentration of hepatic mitochondria by 30.90% (p<0.01) and 10.39% (p<0.05), respectively; Supplemented dihydropyridine had no significant effects on TG, Ch HDL-C and VLDL-C concentrations in serum; addition of 150 or 300 mg/kg dihydropyridine increased T3 levels in serum by 15.34% (p<0.05) and 11.88% (p<0.05) and decreased insulin concentration by 40.44% (p<0.05) and 54.37% (p<0.05), respectively. The results demonstrated that adding dihydropyridine had the tendency of improving very low density lipoprotein receptor (VLDLR) content in the ovary. It was concluded that dihydropyridine could improve laying performance and regulate the fat metabolism of laying hens and that 150 mg/kg dihydropyridine is the optimum dose for laying birds in practical conditions.
Magnetic Study of Heavily Mn-doped CuO Thin Films

Cu0.7Mn0.3O films were deposited by Plasma Enhanced Chemical Vapor Deposition and annealed in different atmospheres. The XRD diffraction results indicate that samples annealed in air and N2 both show pure CuO phases, while the film annealed in O2 has a secondary phase CuMn2O4. The content of Mn was confirmed by Energy dispersive X-ray spectroscopy. The M-T and M-H curves show that the average magnetic moment of the films prepared in O2 is larger than those prepared in air and in N2 atmosphere. Ferromagnetic segments Mn-O-Cu-O-Mn can be used to understand the ferromagnetism of CuO under heavily Mn-doping. And the magnetic moment per Mn ions calculated on the base of this chain model is consistent with the experimental result.

Thermal evaporation was carried out in a horizontal quartz tube under an oxygen flow of about 50 ml/min and the influence of reaction time and temperature on the microstructure of the CuO nanowires (CNWs) is examined. The magnetic susceptibilities of the as-synthesized CNWs in the 5e300 K range were studied. It is interesting to note that the CNWs with a much larger diameter than 10 nm exhibit anomalous ferromagnetic behavior which has never been reported previously, demonstrating the effect of their peculiar morphology. The saturation magnetization (MS) and coercive field (Hc) of CNWs grown at 700 ℃ are 2.39 × 10-4 emu and 48 Oe, respectively. We fabricated gas sensors based on p-type single CNWs and demonstrate that CuO nanowires could be a promising candidate for a gas sensor with good performance. The reaction between the reducing gas and O leads to a decrease of the hole density in the surface charge layer and an increase of the CuO resistance.
