Runx3 is required for the differentiation of lung epithelial cells and suppression of lung cancer

Human lung adenocarcinoma, the most prevalent form of lung cancer, is characterized by many molecular abnormalities. K-ras mutations are associated with the initiation of lung adenocarcinomas, but K-ras-independent mechanisms may also initiate lung tumors. Here, we find that the runt-related transcription factor Runx3 is essential for normal murine lung development and is a tumor suppressor that prevents lung adenocarcinoma. Runx3−/− mice, which die soon after birth, exhibit alveolar hyperplasia. Importantly, Runx3−/− bronchioli exhibit impaired differentiation, as evidenced by the accumulation of epithelial cells containing specific markers for both alveolar (that is SP-B) and bronchiolar (that is CC10) lineages. Runx3−/− epithelial cells also express Bmi1, which supports self-renewal of stem cells. Lung adenomas spontaneously develop in aging Runx3+/− mice (∼18 months after birth) and invariably exhibit reduced levels of Runx3. As K-ras mutations are very rare in these adenomas, Runx3+/− mice provide an animal model for lung tumorigenesis that recapitulates the preneoplastic stage of human lung adenocarcinoma development, which is independent of K-Ras mutation. We conclude that Runx3 is essential for lung epithelial cell differentiation, and that downregulation of Runx3 is causally linked to the preneoplastic stage of lung adenocarcinoma.

The determination of optimum cutting condition is difficult since the metal removal capability of any machine tool will depend upon factors such as speed, feed, depth of cut, work-piece material, machine temperature, type of work piece mounting, etc, and wide variations are evident in practice between the combinations of cutting conditions employed by different users. In this study, the static and dynamic components of cutting resistance were measured by tool dynamometer when S45C, Al alloy and FC 20 were machining on the lathe under some variable cutting conditions. The results obtained are as follows; 1) The dynamic component of these cutting resistances were increased in accordance with the increase in cutting area. 2) The proportion of the dynamic component to the static component was almost constant under the same material and cutting conditions. 3) There were much differences between the dynamic component of cutting resistance and cutting velocity in accordance with the materials of specimens.

We have succeeded in preparing single crystalline lms of BaFeO3, which may contain unusual Fe4+ ions, on (100) SrTiO3 substrates by pulsed laser beam deposition. The RHEED patterns for the lms on the (100) substrates apparently show a C4 symmetry, suggesting that the sample had a pseudo-cubic perovskite structure rather than hexagonal unit cell. The (100) epitaxially grown lms were found to be highly insulating, up to the leakage current density lower than 1 A/cm2 at the bias voltage of ..1:5 V. In addition, the lms showed a high value of dielectric constant of " = 59, and relatively small dissipation factor of about 0.07 at 100 kHz. The samples were also revealed to have a remanent magnetization at 300 K.

<P><B>Abstract</B></P><P>Mg<SUP>2+</SUP> ion stimulates the DNA strand exchange reaction catalyzed by RecA, a key step in homologous recombination. To elucidate the molecular mechanisms underlying the role of Mg<SUP>2+</SUP> and the strand exchange reaction itself, we investigated the interaction of RecA with Mg<SUP>2+</SUP> and sought to determine which step of the reaction is affected. Thermal stability, intrinsic fluorescence, and native mass spectrometric analyses of RecA revealed that RecA binds at least two Mg<SUP>2+</SUP> ions with K<SUB>D</SUB> ≈ 2 mM and 5 mM. Deletion of the C-terminal acidic tail of RecA made its thermal stability and fluorescence characteristics insensitive to Mg<SUP>2+</SUP> and similar to those of full-length RecA in the presence of saturating Mg<SUP>2+</SUP>. These observations, together with the results of a molecular dynamics simulation, support the idea that the acidic tail hampers the strand exchange reaction by interacting with other parts of RecA, and that binding of Mg<SUP>2+</SUP> to the tail prevents these interactions and releases RecA from inhibition. We observed that binding of the first Mg<SUP>2+</SUP> stimulated joint molecule formation, whereas binding of the second stimulated progression of the reaction. Thus, RecA is actively involved in the strand exchange step as well as bringing the two DNAs close to each other.</P>

<P></P> <P>The CCQM-K97 key comparison was organized by the inorganic analysis working group (IAWG) of CCQM as a follow-up to completed pilot study CCQM-P96 and P96.1 to test the abilities of the national metrology institutes to accurately quantitate the mass fraction of arsenobetaine (AsB) in standard solution and in fish tissue. A pilot study CCQM-P133 was parallelized with this key comparison. National Institute of Metrology (NIM), China and National Metrology Institute of Japan (NMIJ) acted as the coordinating laboratories.</P> <P>Six NMIs participated in CCQM-K97 and two institutes participated in CCQM-P133, and all of them submitted the results. Some NMIs submitted more than one results by different methods. The results were in excellent agreement with each other, and obviously better than those of previous P96 and P96.1. Therefore the calibrant which each NMI used was comparable. Various effects of geometry on the magnetic properties at microwave frequencies ( 20 GHz)are systematically studied for micro- and nano-structured ferromagnetic thin films with fourfoldin-plane magnetic anisotropy by using micromagnetic simulations. Obtained numerical results onthe dynamics of the micro- and the nano-structured magnetic thin film are useful for design considerationsfor various high-frequency devices, including thin-film inductors, microwave filters andabsorbers. The permeability and the resonance frequency are found to be well scaled with a structuralparameter of w/t (w = lateral pattern size and t = film thickness) for pattern sizes that differby an order of magnitude. An exceptional resonance behavior of the smallest pattern with w =0.5 µm was reasoned to be significant in suppressing the precession in the vicinity of the patternedge. We also demonstrated that stable magnetization directions orthogonal to each other could beswitched with external fields, enabling local control of the permeability in the structured pattern.
Runx3 expression in gastrointestinal tract epithelium: resolving the controversy

We reported earlier that RUNX3 is expressed in human and mouse gastrointestinal tract (GIT) epithelium and that it functions as a tumor suppressor in gastric and colorectal tissues. However, there have been conflicting reports describing the absence of Runx3 in GIT epithelial cells. A part of the controversy may be derived from the use of a specific antibody by other groups (referred to as G-poly). Here, we show further evidence to support our earlier observations and provide a possible explanation for this apparent controversy. We generated multiple anti-RUNX3 monoclonal antibodies and found that RUNX3 antibodies recognizing the RUNX3 N-terminal region (residues 1–234) react with RUNX3 in gastric epithelial cells, whereas those recognizing the C-terminal region (beyond residue 234) did not. G-poly primarily recognizes the region beyond 234 and hence, is unable to detect Runx3 in this tissue.Oncogene (2009) 28, 1379–1384; doi:10.1038/onc.2008.496; published online 26 January 2009
