      • Comprehensive Bioinformation Analysis of the MRNA Profile of Fascin Knockdown in Esophageal Squamous Cell Carcinoma

        Background: Fascin, an actin-bundling protein forming actin bundles including filopodia and stress fibers, is overexpressed in multiple human epithelial cancers including esophageal squamous cell carcinoma (ESCC). Previously we conducted a microarray experiment to analyze fascin knockdown by RNAi in ESCC. Method: In this study, the differentially expressed genes from mRNA expression profilomg of fascin knockdown were analyzed by multiple bioinformatics methods for a comprehensive understanding of the role of fascin. Results: Gene Ontology enrichment found terms associated with cytoskeleton organization, including cell adhesion, actin filament binding and actin cytoskeleton, which might be related to fascin function. Except GO categories, the differentially expressed genes were annotated by 45 functional categories from the Functional Annotation Chart of DAVID. Subpathway analysis showed thirty-nine pathways were disturbed by the differentially expressed genes, providing more detailed information than traditional pathway enrichment analysis. Two subpathways derivated from regulation of the actin cytoskeleton were shown. Promoter analysis results indicated distinguishing sequence patterns and transcription factors in response to the co-expression of downregulated or upregulated differentially expressed genes. MNB1A, c-ETS, GATA2 and Prrx2 potentially regulate the transcription of the downregulated gene set, while Arnt-Ahr, ZNF42, Ubx and TCF11-MafG might co-regulate the upregulated genes. Conclusions: This multiple bioinformatic analysis helps provide a comprehensive understanding of the roles of fascin after its knockdown in ESCC.

      • Research on Life Prediction of Train Control Center Based on Random and Fuzzy Theory

        The health state assessment and remaining life prediction of Train Control Center (TCC) is an important premise to carry out state repair of equipment, which can effectively protect the safe operation of the system. The paper aims to assess the health state of the equipment by adopting the health index model of TCC based on fuzzy comprehensive evaluation. As to various uncertain factors the TCC has such as the operating environment, the quality of the equipment and maintenance, the life prediction model of TCC is established based on the evaluative result, namely health index HI. Further variation that the equipment health index changes with time is established to realize the life prediction of TCC. The example shows that the life of TCC can be predicted effectively by means of the health state assessment for the equipment. The railway department can obtain the corresponding data, which can provide the basis of the maintenance and update for the equipment.

      • MSP58 Knockdown Inhibits the Proliferation of Esophageal Squamous Cell Carcinoma in Vitro and in Vivo

        Esophageal carcinoma (EC) is one of the most aggressive cancers with a poor prognosis. Understanding the molecular mechanisms underlying esophageal cancer progression is a high priority for improved EC diagnosis and prognosis. Recently, MSP58 was shown to behave as an oncogene in colorectal carcinomas and gliomas. However, little is known about its function in esophageal carcinomas. We therefore examined the effects of MSP58 knockdown on the growth of esophageal squamous cell carcinoma (ESCC) cells in vitro and in vivo in order to gain a better understanding of its potential as a tumor therapeutic target. We employed lentiviral-mediated small hairpin RNA (shRNA) to knock down the expression of MSP58 in the ESCC cell lines Eca-109 and EC9706 and demonstrated inhibition of ESCC cell proliferation and colony formation in vitro. Furthermore, flow cytometry and western blot analyses revealed that MSP58 depletion induced cell cycle arrest by regulating the expression of P21, CDK4 and cyclin D1. Notably, the downregulation of MSP58 significantly inhibited the growth of ESCC xenografts in nude mice. Our results suggest that MSP58 may play an important role in ESCC progression.

        Ca2+ entry via alpha1G and TRPV4 channels differentially regulates surface expression of P-selectin and barrier integrity in pulmonary capillary endothelium.

        <P>Pulmonary vascular endothelial cells express a variety of ion channels that mediate Ca(2+) influx in response to diverse environmental stimuli. However, it is not clear whether Ca(2+) influx from discrete ion channels is functionally coupled to specific outcomes. Thus we conducted a systematic study in mouse lung to address whether the alpha(1G) T-type Ca(2+) channel and the transient receptor potential channel TRPV4 have discrete functional roles in pulmonary capillary endothelium. We used real-time fluorescence imaging for endothelial cytosolic Ca(2+), immunohistochemistry to probe for surface expression of P-selectin, and the filtration coefficient to specifically measure lung endothelial permeability. We demonstrate that membrane depolarization via exposure of pulmonary vascular endothelium to a high-K(+) perfusate induces Ca(2+) entry into alveolar septal endothelial cells and exclusively leads to the surface expression of P-selectin. In contrast, Ca(2+) entry in septal endothelium evoked by the selective TRPV4 activator 4alpha-phorbol-12,13-didecanoate (4alpha-PDD) specifically increases lung endothelial permeability without effect on P-selectin expression. Pharmacological blockade or knockout of alpha(1G) abolishes depolarization-induced Ca(2+) entry and surface expression of P-selectin but does not prevent 4alpha-PDD-activated Ca(2+) entry and the resultant increase in permeability. Conversely, blockade or knockout of TRPV4 specifically abolishes 4alpha-PDD-activated Ca(2+) entry and the increase in permeability, while not impacting depolarization-induced Ca(2+) entry and surface expression of P-selectin. We conclude that in alveolar septal capillaries Ca(2+) entry through alpha(1G) and TRPV4 channels differentially and specifically regulates the transition of endothelial procoagulant phenotype and barrier integrity, respectively.</P>

        Phenotypes and Prognostic Factors of Syringomyelia in Single-Center Patients With Chiari I Malformation: Moniliform Type as a Special Configuration

        Objective: The specific association between morphometric characteristics of the syrinx and the prognosis of Chiari malformation type I (CM-I) with syringomyelia following surgical procedure seems to have not been fully elaborated. This study focused on the preoperative clinical and radiologic parameters in CM-I patients with syringomyelia to find out the relationship between the patients’ clinical status and the phenotypes of the syrinx with surgical outcome. Methods: A continuous series of pediatric and adult patients with CM-I and syringomyelia from a prospectively maintained database in a single center were included, and we explored the related factors affecting the prognosis following decompression surgery through retrospective analysis of clinical presentations, imaging characteristics, and the morphological features of syringomyelia, to provide a clinical reference for the treatment of syringomyelia. Results: There were 28 pediatric patients (13.8%), and 174 adults (86.2%) included in our study. The average Chicago Chiari Outcome Scale score was 14.56 ± 1.78. The overall prognosis after surgery was good in our series, among them 152 cases (75.25%) with a favorable prognosis, and syrinx was resolved effectively in 172 cases (85.15%). According to the univariate and multivariate analyses, the preoperative symptom duration, observation time, and with/without moniliform type were independent factors affecting the prognosis in adults. The most obvious difference between moniliform type and nonmoniliform type lies in the preoperative symptom duration, ventral subarachnoid space at the foramen magnum, and with/without straightened cervical physio-curve. Conclusion: Timely decompression surgery could achieve a better outcome in CM-I patients with syringomyelia. Moniliform syringomyelia may suggest a relatively better prognosis.

        Hypoglycemic Effects of Ganoderma lucidum Polysaccharides in Type 2 Diabetic Mice

        Our aims were to investigate the hypoglycemic effects and mechanisms of action of Ganoderma lucidum polysaccharides (GLPs) administered for 7 days in type 2 diabetic mice. The mice were randomly divided into four groups (8 mice/group): normal control group, diabetic control group, low-dose GLP-treated diabetic group (50 mg/kg/d), and high-dose GLP-treated diabetic group (100 mg/kg/d). Diabetes was induced by streptozotocin injection and high-fat dietary feeding. At the end of the study, fasting serum glucose, insulin, body weight (BW) and epididymal white adipose tissue weight were measured. The hepatic mRNA levels of glycogen phosphorylase (GP), fructose-1,6-bisphosphatase (FBPase), phosphoenolpyruvate carboxykinase (PEPCK) and glucose-6-phosphatase (G6Pase) genes were determined by real-time polymerase chain reaction. Both doses of GLPs significantly decreased fasting serum glucose, insulin and epididymal fat/BW ratio compared with the diabetic control group (p < 0.05). The hepatic mRNA levels of GP, FBPase, PEPCK and G6Pase were significantly lower in both GLP-treated groups compared with the diabetic control group. Taken together, GLPs significantly decrease fasting serum glucose levels in type 2 diabetic mice in a dose-dependent manner. The decreases in fasting serum glucose levels may be associated with decreased mRNA expression levels of several key enzymes involved in gluconeogenesis and/or glycogenolysis.

        Enhancement of Haloacetate Dehalogenase Production by Strain Mutation and Condition Optimization

        Enhancement of the activity of an inducible chloroacetate dehalogenase was carried out by efficient and safe mutation with UV and microwave irradiation along with optimization of culture conditions. First, a stable mutant of Pseudomonas sp. CGMCC 3267-MW6 with chloroacetate dehalogenase activity of 2.77 U/mL (3-fold higher activity than the wild strain) was produced by mutation. The maximum activity of this inducible enzyme was measured as 29.41 U/mL when Pseudomonas sp. CGMCC 3267-MW6 was cultured with 4 g/L 3-hydroxybutyrate for 12 h followed by 40 mM 3-chlorobutyrate for an additional 20 h. Production of the enzyme was found to be associated with growth of the bacterium. According to these results, we determined that the optimum inducer of chloroacetate dehalogenase activity would be a hard degradable substrate. The optimum auxiliary carbon source would be the primary metabolite of the substrate or the precursor of the metabolite. The optimum time of inducer supplementation would be during the middle stage of exponential phase. The optimum concentration of substrate would be sufficient but would not induce inhibition. Finally, the optimum collection time would be at the later stage of exponential phase. This work provides further knowledge of chloroacetate dehalogenase and the optimization of inducible enzyme production.

        Fault Detection for Interconnected Systems Subject to Packet Dropouts via Switching Scheme

        The design of fault detection filter for interconnected systems with packet dropouts occurring in measurement channels is proposed in this paper. Taking into account arbitrary disconnected interconnection among subsystems, switching scheme is introduced to simplify the analysis of the problem. By modeling these situations as independent subsystems, the novel systems based on arbitrary switched systems are remodeled. The design of the filter makes the augmented system exponentially stable and satisfies the disturbance attenuation performance. A feasible linear matrix inequality for the existence of filter is derived by switched Lyapunov method based on stochastic function. Finally, two examples are shown to illustrate the method and its effectiveness.

        Molecular and cytological evidences for the natural wheatgrass hybrids occurrence and origin in west China

        Interspecies hybridization has been frequently observed in the tribe Triticeae. Natural hybridization between Kengyilia and Roegneria or Elymus species has not been reported as yet. Several sterile wheatgrass individuals exhibiting intermediately morphological traits between Kengyilia and Roegneria or Elymus species were identified in the meadow of Sichuan and Gansu provinces in China, suggesting their natural hybrid origin. The putative hybrids were analyzed by using the sequences of ITS and trnH-psbA together with cytological observation in order to assess the origin of hybrids. Both ITS and cytological data revealed the evidence of allopolyploid origin and confirmed the presence of StStYYP and StStYYHP genomes in the putative natural hybrids. The data suggest that the StStYYP hybrid originated from hybridization between Kengyilia and Roegneria and the hybrid with StStYYHP originated from hybridization between Kengyilia and Elymus. Chloroplast sequence data demonstrated that K. rigidula and K. melanthera were the likely maternal donors in the hybridization events.

        Analysis of the low velocity impact response of functionally graded carbon nanotubes reinforced composite spherical shells

        The low velocity impact analysis of functionally graded carbon nanotubes reinforced composite (FG-CNTRC) spherical shells including the contact force and indentation of the shell surface center was presented. The material properties were set according to the rule of mixture. The fibers and polymeric matrix were temperature-dependent. Timoshenko-Midlin assumption was used to establish the dimensionless nonlinear governing equations of the axis-symmetric transverse isotropy spherical shells. The Hertz contact theory was established to obtain the force and displacement between the spherical shell and impactor. Seven species of comparison analyses including grading profiles and volume fraction were mentioned to investigate the influence on the low velocity impact response of the FG-CNTRC spherical shells. The results revealed that the contact force was determined by the material properties of both contact surface and noncontact surface collectively. Furthermore, the increase of volume fraction can result in the larger contact force and shorter contact time.
