        Research Articles : The Influence of Challenge on Cathepsin B and D Expression Patterns in the Silkworm Bombyx mori L

        Cathepsins are well-characterized proteases that are ubiquitously expressed in lysosomes. Previous work revealed that Bombyx mori cathepsins B and D are expressed in the fat body and undergo decomposition during larval-pupal metamorphosis. Quantitative RTPCR was performed to detect cathepsin gene expression at the transcription level when challenged by B. mori nuclear polyhedrosis virus (BmNPV), temperature and hormones (20-hydroxyecdysone (20E) and juvenile hormone analogue (JHA)). mRNAs encoding cathepsins B and D were significantly enhanced after the larvae were infected with BmNPV, and the peak of the induction appeared at 1 day before spinning. This attenuated the inducing effect on cathepsin expression caused by infection. Temperature shock induced cathepsin expression at the later stage of the 5th instar, and transcription levels varied with development stage and temperature. Cathepsin B and D mRNA expression in the fat body were significantly induced by JHA at the day before spinning, and with 20E, the expression reached a peak at the last day of the 5th instar. Cathepsin B and D mRNA expression exhibited detectable changes post-treatment, without significant differences between or among the hormone concentrations.

        Therapeutic Drug Monitoring of Biologics for Patients with Inflammatory Bowel Diseases: How, When, and for Whom?

        During the past decade, we have entered an era of biologics for the treatment of Crohn’s disease and ulcerative colitis. The therapeutic goal of inflammatory bowel disease (IBD) management has evolved from symptom control and clinical remission to mucosal healing or even deep remission. Histological remission for ulcerative colitis and transmural healing of Crohn’s disease are potential future goals. With the adoption of the treat-to-target concept, and given the need for tight control of IBD activity, therapeutic drug monitoring (TDM) is an important element of precision medicine. TDM involves the measurement of serum biologics and anti-drug antibodies levels, to confirm whether the right drug with the right dosage was prescribed to reach the right serum levels. TDM may help clinicians adjust biologics based on objective biomarkers instead of using empirical dosage escalation or making symptom-based therapeutic adjustments. Wellestablished reactive TDM algorithms have been proposed, and emerging evidence supports the clinical application of a proactive TDM strategy to enhance the duration of effective biologics and improve clinical outcomes. Recently, the proactive TDM strategy was shown to avoid the secondary loss of response to biologics, and improve long-term clinical outcomes in IBD patients. This review summarizes data from trials, and practice guidelines, on the clinical application of proactive and reactive TDM strategies for the daily care of biologic-treated IBD patients.

        Novel AgCl/Ag2SO3 Hybrids as a Visible-Light-driven Photocatalyst: Preparation, Characterization, and Degradation of Rhodamine-B and Methyl Orange

        The novel AgCl/Ag2SO3 hybrids as an efficient photocatalyst had been fabricated by an in situ synthetic method. The correlations between the structure and the photocatalytic properties of the as-fabricated hybrids were analyzed. Experimental results exhibited that with increasing the amount of Ag2SO3, the degradation rate of the as-obtained samples was firstly increased and then decreased under the visible light irradiation. When the mass ratio of AgCl to Ag2SO3 was 1:2, in 30?min, it displayed the highest degradation rate of 99.2% for rhodamine-B, which was obviously higher than 46.1, 60.5, and 14.6% of pure AgCl, Ag2SO3, and TiO2 (P25), respectively. Similar results could be found in degradation of methyl orange. It had the maximum of 97.4% in 90?min, which was higher than 55.2, 48.7, and 12.7% of pure AgCl, Ag2SO3, and P25, respectively. Moreover, the as-prepared hybrids possessed the enhanced separation and transfer of photo-generated electron?hole pairs compared to the pure samples. In addition, the holes and superoxide radicals played the dominant role and the hydroxyl radicals played the secondary role during the process of photocatalytic degradation.

        Relationship between Fractional Exhaled Nitric Oxide Level and Efficacy of Inhaled Corticosteroid in Asthma-COPD Overlap Syndrome Patients with Different Disease Severity

        This study explored the relationship between the fractional exhaled nitric oxide (FeNO) level and the efficacy of inhaled corticosteroid (ICS) in asthma-chronic obstructive pulmonary disease (COPD) overlap syndrome (ACOS) patients with different disease severity. A total of 127 ACOS patients with ACOS (case group) and 131 healthy people (control group) were enrolled in this study. Based on the severity of COPD, the ACOS patients were divided into: mild ACOS; moderate ACOS; severe ACOS; and extremely severe ACOS groups. We compared FeNO levels, pulmonary function parameters including percentage of forced expiratory volume in 1 second (FEV1) to predicted value (FEV1%pred), ratio of FEV1 to forced vital capacity (FEV1/FVC), inspiratory capacity to total lung capacity (IC/TLC) and residual volume to total lung capacity (RV/TLC), arterial blood gas parameters, including PH, arterial partial pressure of oxygen (PaO2) and arterial partial pressure of carbon dioxide (PaCO2), total serum immunoglobulin E (IgE), induced sputum eosinophil (EOS), plasma surfactant protein A (SP-A), plasma soluble receptor for advanced glycation end products (sRAGE), sputum myeloperoxidase (MPO), sputum neutrophil gelatinase-associated lipocalin (NGAL) and Asthma Control Test (ACT) scores, and COPD Assessment Test (CAT) scores. Compared with pre-treatment parameters, the FeNO levels, RV/TLC, PaCO2, total serum IgE, induced sputum EOS, plasma SP-A, sputum MPO, sputum NGAL, and CAT scores were significantly decreased after 6 months of ICS treatment, while FEV1%pred, FEV1/FVC, IC/TLC, PH, PaO2, plasma sRAGE, and ACT scores were significantly increased in ACOS patients with different disease severity after 6 months of ICS treatment. This finding suggests that the FeNO level may accurately predict the efficacy of ICS in the treatment of ACOS patients.

        Agglomeration of particles during coal combustion in multistage spouted fluidized tower

        An experimental platform of spray agglomeration has been designed and built for removing small fly ash particles (PM10) from coal combustion. Systematic experiments were conducted in a multistage spouted tower using kinds of agglomerant solutions. The particle concentration increases greatly from the first stage to the second stage of the tower. With the increase of flue gas flow rate the oscillation of impulse signal response curves increases and the internal circulation of the tower intensifies. The influencing factors such as the surfactant, PH value, flow rate of the agglomerant solutions and inlet flue gas temperature were analyzed. SEM was used to analyze the microstructure of the particles. Final results indicate that the special shape of a multistage spouted fluidized tower has significant influences on the effect of agglomeration. The findings from this work will be helpful to form the basis, and provide guidance for, further studies on the control of fine particles such as PM2.5 or even smaller.

      • Wind tunnel investigation on wind characteristics of flat and mountainous terrain

        Wind tunnel test is often adopted to assess the site-specific wind characteristics for the design of bridges as suggested by current design standards. To investigate the wind characteristics of flat and mountainous terrain, two topographic models are tested in a boundary layer wind tunnel. The wind characteristics, including the vertical and horizontal mean wind speed distributions, the turbulence intensity, and the wind power spectra, are presented. They are investigated intensively in present study with the discussions on the effect of wind direction and the effect of topography. It is indicated that for flat terrain, the wind direction has negligible effect on the wind characteristics, however, the assumption of a homogenous wind field for the mountainous terrain is not applicable. Further, the non-homogeneous wind field can be defined based on a proposed approach if the wind tunnel test or on-site measurement is performed. The calculated turbulence intensities and wind power spectra by using the measured wind speeds are also given. It is shown that for the mountainous terrain, engineers should take into account the variability of the wind characteristics for design considerations

        A putative plastidial adenine nucleotide transporter, BRITTLE1-3, plays an essential role in regulating chloroplast development in rice (Oryza sativa L.)

        Differentiation from proplastids into chloroplasts isa light- and energy-dependent process. How this process isregulated is still poorly understood at the molecular level. We herein report a new putative plastidial adenine nucleotidetransporter, BRITTLE1-3 (referred to as OsBT1-3), encoded bythe rice (Oryza sativa) White Stripe Leaf 2 (WSL2) gene. Loss of OsBT1-3 function results in defective chloroplastbiogenesis, severely reduced photosynthetic efficiency, andfinally a white stripe leaf phenotype in the first four leaves. The expression levels of genes related to chlorophyllbiosynthesis and photosynthesis are drastically reduced,accompanied with over accumulation of reactive oxygenspecies (ROS) in the wsl2 mutant. OsBT1-3 is targeted tothe chloroplasts and it expresses in almost all tissues inplants, especially in young leaves. OsBT1-3 consists of 419amino acids and exhibits features of all mitochondrialcarrier proteins, including a typical transmembrane-spanningdomain and a highly conserved sequence motif designatedas the ‘mitochondrial energy transfer signatures’. Phylogeneticanalysis shows that OsBT1-3 is a putative plastidialadenine nucleotide transporter and is most closely relatedto ZmBT1-2. Together, these observations suggest that thenew putative adenine nucleotide transporter, OsBT1-3,plays an essential role in regulating chloroplast biogenesisand maintenance of ROS homeostasis during rice seedlingde-etiolation.

        Highly efficient and ultra-rapid adsorption of malachite green by recyclable crab shell biochar

        In this study, waste crab shell was hydrothermally carbonized and then activated to prepare an effectiveand recyclable adsorbent. The adsorption performance of the adsorbent for different dyes was investigatedthrough adsorption experiments, and the results showed that it had the largest adsorption performancefor malachite green. Furthermore, the adsorption performance and mechanism of malachite greenwere investigated by static and dynamic adsorption method. The crab shell biochar could remove6142.5 mg g1 of malachite green within 3 minutes, indicating that the crab shell biochar could ultrarapidand highly efficient adsorb malachite green. In addition, the malachite green adsorbed on the crabshell biochar could be removed by calcination, which achieved the entire regeneration of the crab shellbiochar. The adsorption of malachite green by crab shell biochar adsorbent conformed to the pseudosecond-order kinetic model, the Dubinin-Radushkevich and Langmuir isotherm adsorption model. Physical and chemical characterization indicated that the adsorption mechanism of malachite greenincluded alkaline fading effect, hydrogen bonding and p-p interaction.
