<P><B>Abstract</B></P> <P>Severe fever with thrombocytopenia syndrome is a tick-borne infectious disease. The present study investigated the prevalence of severe fever with thrombocytopenia syndrome virus (SFTSV) in shelter dogs and cats in the Republic of Korea (ROK). Blood samples were collected from 426 dogs and 215 cats in animal shelters throughout the ROK in 2016. Of the tested samples, one (0.2%) dog and one (0.5%) cat were positive for SFTSV. Phylogenetic analysis of the sequences obtained in the present study showed that the viruses belonged to the J3 clade, which is considered the dominant clade in the ROK. This study reports the first molecular detection of SFTSV in shelter dogs and cats in the ROK.</P>

<P><B>Abstract</B></P> <P>The purpose of this study was to investigate the seroprevalence of antibodies against severe fever with thrombocytopenia syndrome virus (SFTSV) in shelter dogs in the Republic of Korea (ROK) using an indirect immunofluorescence assay and virus neutralization test. Sera were collected from 426 dogs in 12 animal shelters throughout the ROK from March 2016 to November 2016. Overall, 59 of 426 (13.9%) samples were seropositive for antibodies against SFTSV. A significant difference was observed in accordance with the sampling region (<I>p</I> <0.001), but not according to the sex (<I>p</I> =0.279) or breed (<I>p</I> =0.729) of the dogs. The seroprevalence of SFTSV showed an inversely proportional trend to the latitude of the sampling regions: the highest rate was observed in the southern region followed by the Jeju-do region. This is the first report on the nationwide prevalence of antibodies against SFTSV in companion dogs in animal shelters throughout the ROK.</P>

<P>Oligodendrocytes are myelin-forming cells in the central nervous system. Research into the effects of aging on oligodendrocyte protein expression remains limited. The present study aimed to determine the alterations in oligodendrocyte-specific protein (OSP) expression in the gerbil hippocampus at 1, 2, 3, 4, 6 and 24 months of age with western blot and immunohistochemistry analyses. OSP expression levels in the hippocampus were highest at 6 months of age. OSP immunoreactivity was identified in numerous cell bodies at 1 month, although the number of OSP immunoreactive cells was different according to hippocampal subregion. The number of OSP immunoreactive cells significantly decreased at 2 months and, thereafter, numbers decreased gradually. The detection of OSP immunoreactive fibers was negligible in all layers in the hippocampal subregions until 4 months. OSP immunoreactive fibers were abundant at 6 and 24 months, although the fiber distribution patterns in the CA1-3 areas and dentate gyrus were different. The results demonstrated that OSP expression in the gerbil hippocampus was age-dependent. The detection of OSP immunoreactive cell bodies and fibers was significantly different according to the layers of hippocampal subregions, indicating that myelination may be continuously altered in the hippocampus during normal aging.</P>

<P>Insulin-like growth factor-I (IGF-I) is a multifunctional protein present in the central nervous system. A number of previous studies have revealed alterations in IGF-I and its receptor (IGF-IR) expression in various regions of the brain. However, there are few reports on age-dependent alterations in IGF-I and IGF-IR expressions in the olfactory bulb, which contains the secondary neurons of the olfactory system. The present study examined the cellular morphology in the olfactory bulb by using cresyl violet (CV) staining at postnatal month (PM) 3 in the young group, PM 6 in the adult group and PM 24 in the aged group in gerbils. In addition, detailed examinations were performed of the protein levels and immunoreactivities of IGF-I and IGF-IR in the olfactory bulb in each group. There were no significant changes in the cellular morphology between the three groups. The protein levels and immunoreactivities of the IGF-I and IGF-IR were the highest in the young group and they decreased with age. He protein levels and immunoreactivities of the IGF-I and IGF-IR were the lowest in the aged group. In brief, our results indicate that IGF-I and IGF-IR expressions are strong in young olfactory bulbs and significantly reduced in aged olfactory bulbs. In conclusion, subsequent decreases in IGF-I and IGF-IR expression with age may be associated with olfactory decline. Further studies are required to investigate the roles of IFG-I and IGF-IR in disorders of the olfactory system.</P>
Efficacy of Intracytoplasmic Sperm Injection for Leukocytospermia

White blood cells (WBCs) are present in most human ejaculates, but abnormally high concentration of seminal leukocytes may reflect an underlying pathological condition. The World Health Organization (WHO) has defined leukocytospermia as status of more than 10? WBC/mL of semen. The purpose of this study was firstly, to compare the outcomes between conventional IVF and ICSI in leukocytospermia, and secondly, to investigate whether ICSI may be an alternation treatment for patients with leukocytospermia. Total 346 cycles of conventional IVF and ICSI candidates underwent IVF cycles at Eulji Hospital Infertility Clinic. Semen Parameters including concentration, motility, morphology of spermatozoa and concentration of leukocytes were assessed from the raw ejaculates. There was no difference in sperm concentration, motility and morphology. The rates of fertilization and good embryo development from ICSI were significantly higher than those from conventional IVF in leukocytospermia (60.4% & 32.5%, respectively for ICSI group and 44.4% & 28.5%, respectively for IVF group, P<0.001). The pregnancy rate after ICSI was also higher than that from conventional IVF (34.0% vs 29.1%, P<0.05). These results indicate that the presence of seminal leukocyes (>1×10? WBC/mL of semen) is adversely related with fertilization, embryo development and pregnancy rate. Therfore the measurement of seminal leukocytes in routine semen analysis appears to be of prognostic value with regard to male fertilizing potential. In conclusion, it is suggested that ICSI is an alternative choice of treatment for patients with leukocytospermia.

By both in vitro hydroxylamine mutagenesis of the wild type 3-phosphoglycerate kinase gene (PGK) promoter DNA and insertion of the leu2-d gene, we have created yeast expression vectors potentially useful for production of eukaryotic genes in yeast. The guanine (G) to adenine (A) change at the -3 position from the ATG start codon of the PGK promoter-based vector rendered a 6∼7 times elevated expression of the adjacent eukaryotic gene, and insertion of the leu2-d gene in the vector containing the mutated PGK promoter further enhanced the expression of the gene. When expression of the AIDS virus HIV1-gagP17 gene in a IacZ fusion form was examined with this new vector, a 15 times higher level of expression than that from the original PGK promoter was observed. Northern and Southern analysis showed that this elevated expression is due to the production of a high copy number of mRNA by leu2-d gene functioning and by efficient translation of the produced mRNA. Thus, the vector that contained the A at the -3 position from the ATG start codon in the promoter region and the leu2-d gene shows increased expression capability and will be potentially useful for production of eukaryotic genes in yeast.
