        Conduction Mechanism in (La0.7Sr0.3MnO3)n(BiFeO3)n Multilayered Thin Films

        (La0.7Sr0.3MnO3)n(BiFeO3)n multilayered thin films were deposited on (0 0 1) SrTiO3 substrates by using the RF magnetron sputtering method, and their conduction mechanisms in the temperature range between 150 K and 300 K were investigated using several common dielectric conduction models. The results indicate the current-voltage characterization of the as-fabricated thin films obey Ohm’s law at 300 K, but the space-charge-limited conduction mechanism becomes dominant in the as-fabricated thin films as the temperature is decreased.

        Identification and heterologous reconstitution of a 5-alk(en)ylresorcinol synthase from endophytic fungus Shiraia sp. Slf14

        A new type III polyketide synthase gene (Ssars) was discovered from the genome of Shiraia sp. Slf14, an endophytic fungal strain from Huperzia serrata. The intron-free gene was cloned from the cDNA and ligated to two expression vectors pET28a and YEpADH2p-URA3 for expression in Escherichia coli BL21(DE3) and Saccharomyces cerevisiae BJ5464, respectively. SsARS was efficiently expressed in E. coli BL21(DE3), leading to the synthesis of a series of polyketide products. Six major products were isolated from the engineered E. coli and characterized as 1,3-dihydroxyphenyl- 5-undecane, 1,3-dihydroxyphenyl-5-cis-6 -tridecene,1,3-dihydroxyphenyl- 5-tridecane, 1,3-dihydroxyphenyl-5-cis-8 - pentadecene, 1,3-dihydroxyphenyl-5-pentadecane, and 1,3- dihydroxyphenyl-5-cis-10 -heptadecene, respectively, based on the spectral data and biosynthetic origin. Expression of SsARS in the yeast also led to the synthesis of the same polyketide products, indicating that this enzyme can be reconstituted in both heterologous hosts. Supplementation of soybean oil into the culture of E. coli BL21(DE3)/SsARS increased the production titers of 1–6 and led to the synthesis of an additional product, which was identified as 5-(8 Z,11 Z-heptadecadienyl) resorcinol. This work thus allowed the identification of SsARS as a 5-alk(en)ylresorcinol synthase with flexible substrate specificity toward endogenous and exogenous fatty acids. Desired resorcinol derivatives may be synthesized by supplying corresponding fatty acids into the culture medium.

        Effect of acidic properties of hierarchical HZSM-5 on the product distribution in methanol conversion to gasoline

        Hierarchical ZSM-5 zeolites with different SiO2/Al2O3 ratio but similar crystal size were directly synthesized by a single-template hydrothermal method, and the intrinsic effect of acidic properties on their catalytic performance in methanol to gasoline (MTG) reaction was comprehensively investigated. The physicochemical properties of HZSM-5 zeolites were characterized by XRD, N2 adsorption-desorption, SEM, NH3-TPD, FTIR, and TGA techniques. The results show good linear correlations between the yields of gasoline components and the relative content of Brønsted acid sites, and the hierarchical HZSM-5 zeolite with SiO2/Al2O3 molar ratio of 200 was firstly found to exhibit high reactivity, excellent product distribution and superior stability in MTG reaction, which can be attributed to its appropriate acid distribution with moderate Brønsted acid sites and proper B/L ratio, predominantly promoting gasoline range hydrocarbons production and inhibiting side reactions.

        Wnt pathway is involved in 5-FU drug resistance of colorectal cancer cells

        Colorectal cancer (CRC) is one of the leading causes of cancer-related death worldwide. 5-Fluorouracil (5-FU) is widely used in the treatment of cancers, but its antineoplastic activity is limited in drug-resistant cancer cells. To investigate the detailed mechanism of 5-FU resistance, we developed a model of 5-FU-resistant cells from HCT-8 cells, a wellestablished colorectal cancer cell line. We found that the drug-resistant cells demonstrated high expression of TCF4 and β-catenin, indicating an upregulated Wnt pathway. A microarray analysis revealed that the suppression of the checkpoint kinase 1 (CHK1) pathway explained the resistance to 5-FU, especially in p53 wild-type cancer cells such as HCT-8. Our data also demonstrated that the CHK1 pathway is suppressed by the Wnt pathway in 5-FU-resistant cells. In summary, we have discovered a novel mechanism for 5-FU resistance mediated by histone deacetylation, which also revealed the crosstalk between the Wnt pathway and CHK1 pathway.
