        소화기 ; 대전 지역에서 급성 A형 간염의 유전자형에 따른 임상 특성 고찰

        목적: B형 간염이나 C형 간염 등은 유전자형에 따른 차이가 알려져 있지만, 국내에서 증가하는 IIIA 유전자형과 기존의 IA 유전자형에 의한 간염의 차이가 거의 알려진 바 없다. 따라서 대전 지역에서 발생한 급성 A형 간염의 유전자형에 따른 차이를 알고자 한다. 방법: 2009년 11월부터 2010년 6월까지 급성 A형 간염으로 진단되면서 유전자형이 확인된 81명의 환자를 대상으로 임상적 소견들과 검사실 소견들을 전향적으로 연구하였다. 결과: 평균 나이는 32.6±7.4세이며, 평균 입원 기간은 7.7±2.4일이었다. 직업은 서비스직, 사무직 그리고 교육직이 가장 많았다. 유전자형 간의 급성 간염에 따른 증상들의 종류는 별다른 차이가 없었다. 유전자형에 따른 증상의 빈도는 IIIA 유전자형에서 구토와 설사가, IA 유전자형에서는 복통과 피부 반점이 상대적으로 많았다. 검사실 소견에서는 IIIA 유전자형에서 헤모글로빈의 평균, CRP 평균, AST 최대 수치 평균은 상대적으로 유의하게 높았다. 그리고 AST와 ALT 및 총 빌리루빈 최대 수치들과 INR의 수치가 IIIA 유전자형에서 상대적으로 높게 분포하는 경향을 보였다. 결론: 대전 지역에서 유행하는 급성 A형 간염에서 IIIA 유전자형에 따른 급성 간염은 기존의 IA 유전자형과 차이가 있을 것으로 보이며, 다기관 연구를 통한 심도 깊은 연구가 필요하겠다. Background/Aims: Acute viral hepatitis A is a major health problem in Korea and the influx of genotype IIIA is thought to be one reason. We examined the differences in the clinical characteristics and laboratory findings of genotypes IA and IIIA in Daejeon. Methods: From November 2009 to June 2010, 81 patients positive for IgM anti-HAV were enrolled prospectively. The hepatitis A was genotyped using real-time polymerase chain reaction. The clinical characteristics and laboratory results were compared on the Results: The mean patient age was 32.6±7.4 years. The mean hospitalization was 7.7±2.4 days. The patient occupation varied. Clinically, vomiting and diarrhea were relatively more prevalent in genotype IIIA than in IA. Abdominal pain and skin spots were relatively more prevalent in genotype IA than in IIIA. The hemoglobin, peak aspartate aminotransferase (AST) level, and C-reactive protein were statistically higher in genotype IIIA than in IA. The distributions of the peak AST, alanine aminotransferase (ALT) and total bilirubin values tended to be perched in genotype IIIA than in IA. The international normalized ratio (INR) tended to be slightly prolonged in genotype IIIA than in IA. Conclusions: Recently, genotype IIIA of acute viral hepatitis A has become prevalent in Daejeon. Hepatitis A genotype IIIA probably causes worse laboratory abnormalities than genotype IA.

      • A rationally designed small molecule for identifying an <i>in vivo</i> link between metal–amyloid-β complexes and the pathogenesis of Alzheimer's disease

        <▼1><P>An <I>in vivo</I> chemical tool designed to target metal–Aβ complexes and modulate their activity was applied to the 5XFAD mouse model of Alzheimer’s disease (AD) demonstrating the involvement of metal–Aβ in AD pathology.</P></▼1><▼2><P>Multiple factors, including amyloid-β (Aβ), metals, and reactive oxygen species (ROS), are involved in the development of Alzheimer's disease (AD). Metal ions can interact with Aβ species generating toxic oligomers and ROS <I>in vitro</I>; however, the involvement of metal–Aβ complexes in AD pathology <I>in vivo</I> remains unclear. To solve this uncertainty, we have developed a chemical tool (<B>L2-b</B>) that specifically targets metal–Aβ complexes and modulates their reactivity (<I>i.e.</I>, metal–Aβ aggregation, toxic oligomer formation, and ROS production). Through the studies presented herein, we demonstrate that <B>L2-b</B> is able to specifically interact with metal–Aβ complexes over metal-free Aβ analogues, redirect metal–Aβ aggregation into off-pathway, nontoxic less structured Aβ aggregates, and diminish metal–Aβ-induced ROS production, overall mitigating metal–Aβ-triggered toxicity, confirmed by multidisciplinary approaches. <B>L2-b</B> is also verified to enter the brain <I>in vivo</I> with relative metabolic stability. Most importantly, upon treatment of 5XFAD AD mice with <B>L2-b</B>, (i) metal–Aβ complexes are targeted and modulated in the brain; (ii) amyloid pathology is reduced; and (iii) cognition deficits are significantly improved. To the best of our knowledge, by employing an <I>in vivo</I> chemical tool specifically prepared for investigating metal–Aβ complexes, we report for the first time experimental evidence that metal–Aβ complexes are related directly to AD pathogenesis.</P></▼2>

        Base-pair opening dynamics of the microRNA precursor pri-miR156a affect temperature-responsive flowering in Arabidopsis

        <P><B>Abstract</B></P> <P>Internal and environmental cues, including ambient temperature changes, regulate the timing of flowering in plants. Arabidopsis miR156 represses flowering and plays an important role in the regulation of temperature-responsive flowering. However, the molecular basis of miR156 processing at lower temperatures remains largely unknown. Here, we performed nuclear magnetic resonance studies to investigate the base-pair opening dynamics of model RNAs at 16 °C and investigated the <I>in vivo</I> effects of the mutant RNAs on temperature-responsive flowering. The A9C and A10CG mutations in the B5 bulge of the lower stem of pri-miR156a stabilized the C15∙G98 and U16∙A97 base-pairs at the cleavage site of pri-miR156a at 16 °C. Consistent with this, production of mature miR156 was severely affected in plants overexpressing the A9C and A10CG constructs and these plants exhibited almost no delay in flowering at 16 °C. The A10G and A9AC mutations did not strongly affect C15∙G98 and U16∙A97 base-pairs at 16 °C, and plants overexpressing A10G and A9AC mutants of miR156 produced more mature miR156 than plants overexpressing the A9C and A10CG mutants and showed a strong delay in flowering at 16 °C. Interestingly, the A9AC mutation had distinct effects on the opening dynamics of the C15∙G98 and U16∙A97 base-pairs between 16 °C and 23 °C, and plants expressing the A9AC mutant miR156 showed only a moderate delay in flowering at 16 °C. Based on these results, we propose that fine-tuning of the base-pair stability at the cleavage site is essential for efficient processing of pri-miR156a at a low temperature and for reduced flowering sensitivity to ambient temperature changes.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A mutation in pri-miR156a B5 affected base-pairing stability at the cleavage site. </LI> <LI> Mutations in the B5 bulge increased base-pair stabilities and decreased miR156. </LI> <LI> The A9AC mutation affected opening dynamics between 16 and 23 °C. </LI> <LI> Cleavage site base-pair stabilities reflected miR156 levels and flowering time. </LI> </UL> </P>

        <P>A(3) adenosine receptor (AR) ligands including A(3) AR agonist, N-6-(3-iodobenzyl)adenosine-5'-N-methyluronamide (1a, IB-MEGA) were examined for adiponectin production in human bone marrow mesenchymal stem cells (hBM-MSCs). In this model, 1a significantly increased adiponectin production, which is associated with improved insulin sensitivity. However, A(3) AR antagonists also promoted adiponectin production in hBM-MSCs, indicating that the A(3) AR pathway may not be directly involved in the adiponectin promoting activity. In a target deconvolution study, their adiponectin-promoting activity was significantly correlated to their binding activity to both peroxisome proliferator activated receptor (PPAR) gamma and PPAR delta. They functioned as both PPAR gamma partial agonists and PPAR delta antagonists. In the diabetic mouse model, la and its structural analogues A(3) AR antagonists significantly decreased the serum levels of glucose and triglyceride, supporting their antidiabetic potential. These findings indicate that the polypharmacophore of these compounds may provide therapeutic insight into their multipotent efficacy against various human diseases.</P>

        Comparison of Proteome Components of Helicobacter pylori Before and After Mouse Passage

        The mouse model is alleged to be a useful tool for understanding of pathophysiological roles of Helicobacter pylori in the development of gastric disorders. However, it has been observed that H. pylori strains significantly differed in their fitness in mice and even mouse strains differed in their susceptibilities to a H. pylori strain. Bacterial components of H. pylori which could affect on its fitness in mice have to be elucidated for the establishment of the mouse model for H. pylori infections. In the comparison of colonization ability between two H. pylori Korean isolates, 51 (isolated from a patient with duodenal ulcer) and 52 (isolated from a patient with gastric cancer), 52 could colonize better than 51 on the gastric mucosa of mouse. Proteome components of H. pylori 52, as a good colonizer and H. pylori 51, as a poor one were quantitatively compared each other. Five bacterial proteins including catalase, urease subunit alpha/beta, enolase and ferritin, were up-regulated in 52. In addition, the respective proteome components of the two strains were also compared with their mouse-passaged homologous strains. Seven and five proteins, which included catalase, flagellin A/B in common, were up-regulated in mouse-adapted 51 and 52, respectively. Among the fourteen identified proteins, urease subunit alpha/beta, flagellin A/B, catalase, ferritin, superoxide dismutase and neutrophil-activation protein have been previously known to be necessary to gastric colonization of H. pylori in animal models. The other up-regulated proteins including enolase, elongation factor Tu and fructose-bisphosphate aldolase have been reported to be associated with acid tolerance of H. pylori. These data provide confirmatory evidence for the importance of those proteins in the development of H. pylori-associated gastric disorders.

      • Toxicity of Conventional Pesticides Used in Cucumber to lleis koebelei Timberlake (Coleoptera: Coccinellidae: Halyziini)

        This study is the first report about the toxicity of pesticides to the mycophagous predator, I. koebelei, of powdery mildew of agricultural crops. Pesticides we tested are composed of synthetic and environmental-friendly products and being used conventionally for the control of insect or microbial pests on cucumber in Korea. our study was conducted to determine the relative toxicities of several pesticides used in Korea cucumber production to mycophagous natural enemy, I. koebelei and to provide a background for implementation of integrated powdery mildew management programs. Based on IOBC classification, three insecticides, bifenthrin + imidacloprid, acetamiprid + indoxacarb, acetamiprid + etopheprox are classified as having a Class 4 (harmful). Spiromesifen showed the low toxicity to the survival and the fecundity of I. koebelei when this chemical had been exposed to 3rd larva or newly emerged adult via feeding with cucumber powdery mildew. However, pyriproxyfen not only decreased the fecundity of female adult but also strongly prohibited from pupation. Many commercial biological or botanical pesticides can restrict the population of I. koebelei. However, Q pact (a.i. Ampelomyces quisqualis 94013), Top seed (a.i. Paenibacillus polymyxa AC-1), BT one (Bacillus thuringiensis) and Solbitchae (insecticidal microorganism) had no toxicity to I. koebelei when this chemical had been exposed to 3rd larva or newly emerged adult feeding with cucumber powdery mildew.

        The black cutworm, Agrotis ipsilon (Hufnagel, 1766) is a cosmopolitan pest that poses an economic threat to many crops such as pepper, corn, perilla, etc. A. ipsilon can be mass reared by providing Chinese cabbage, Brassica rapa under 60~70%, 16L:8D and 25±1℃ in laboratory. This study presents the mass rearing methods and the morphological characteristics of the immature stages of A. ipsilon including descriptions of the each immature stages.

        Synthesis, structure and electrochemical performance of double perovskite oxide Sr<sub>2</sub>Fe<sub>1-x</sub>Ti<sub>x</sub>NbO<sub>6-δ</sub> as SOFC electrode

        <P><B>Abstract</B></P> <P>A new series of double perovskite oxides Sr<SUB>2</SUB>Fe<SUB>1-x</SUB>Ti<SUB>x</SUB>NbO<SUB>6-δ</SUB> (x = 0, 0.05 & 0.1) were prepared by traditional solid state reaction method and characterized by X-ray diffraction, scanning electron microscope (SEM), energy dispersive x-ray (EDX), ac impedance and dc four point using Van Der Pauw method. The samples were sintered at 1200 °C for 12 h in air to get single phase. Rietveld refinement shows that all composition crystallizes in tetragonal symmetry in the space group <I>I</I>4/<I>m.</I> The lattice parameter were related to the ideal single perovskite a<SUB>p</SUB> as a ≈ b ≈ √2a<SUB>p</SUB>, and c ≈ 2a<SUB>p</SUB>. Scanning electron microscope shows increase in porosity with increase of Ti content. Sr<SUB>2</SUB>Fe<SUB>1-x</SUB>Ti<SUB>x</SUB>NbO<SUB>6-δ</SUB>/YSZ shows a good interface with no lamination problem between the two materials. EDX shows that the element contents of the materials synthesized in this study close to the nominal stoichiometry. Chemical compatibility studies with YSZ electrolyte shows no reaction with the anode. A symmetrical cell with the configuration of Sr<SUB>2</SUB>Fe<SUB>1-x</SUB>Ti<SUB>x</SUB>NbO<SUB>6-δ</SUB>|YSZ|Sr<SUB>2</SUB>Fe<SUB>1-x</SUB>Ti<SUB>x</SUB>NbO<SUB>6-δ</SUB> (x = 0.05 & 0.1) shows the lowest polarization resistance of 49.80 Ω cm<SUP>2</SUP> and the highest ac conductivity of 3.57 × 10<SUP>−3</SUP> S cm<SUP>−1</SUP> in dry H<SUB>2</SUB>/Ar for x = 0.1. The activation energy was found to be 1.20 eV which is almost similar to Sr<SUB>2</SUB>FeNbO<SUB>6-δ</SUB> as reported in literature. The dc conductivity measurement was highest for SFTN0.1 at 750 °C in dry 5% H<SUB>2</SUB>/Ar with a value of 1.17 S cm<SUP>−1</SUP>.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Titanium was substituted for Iron in the mixed conducting perovskite oxide SFN. </LI> <LI> A single double perovskite phase was obtained in air. </LI> <LI> Titanium substitution improves porosity, a.c conductivity and area specific resistance. </LI> <LI> The enhancement has been explained by a simple defect chemistry model. </LI> <LI> Electrochemical performance are interesting for applicability as an SOFC anode material. </LI> </UL> </P>

        Tight junction protein 1 is regulated by transforming growth factor-β and contributes to cell motility in NSCLC cells

        Tight junction protein 1 (TJP1), a component of tight junction, has been reported to play a role in protein networks as an adaptor protein, and TJP1 expression is altered during tumor development. Here, we found that TJP1 expression was increased at the RNA and protein levels in TGF-β-stimulated lung cancer cells, A549. SB431542, a type-I TGF-β receptor inhibitor, as well as SB203580, a p38 kinase inhibitor, significantly abrogated the effect of TGF-β on TJP1 expression. Diphenyleneiodonium, an NADPH oxidase inhibitor, also attenuated TJP1 expression in response to TGF-β in lung cancer cells. When TJP1 expression was reduced by shRNA lentiviral particles in A549 cells (A549-sh TJP1), wound healing was much lower than in cells infected with control viral particles. Taken together, these data suggest that TGF-β enhances TJP1 expression, which may play a role beyond structural support in tight junctions during cancer development.       • Polyphenolic compounds from Korean <i>Lonicera japonica</i> Thunb. induces apoptosis via AKT and caspase cascade activation in A549 cells

        <P><I>Lonicera japonica</I> Thunb. (<I>L. japonica</I> T.) has historically been used in Korean herbal medicine due to its anticancer and protective effects on the respiratory system. In the present study, the polyphenolic compounds in <I>L. japonica</I> T. were investigated using high-performance liquid chromatography coupled with tandem mass spectrometry, and its anticancer effects on A549 non-small-cell lung cancer cells were studied. Polyphenolic compounds potentially inhibit A549 cells in a dose-dependent manner. Flow cytometry and western blot analysis demonstrated that polyphenolic compounds induce apoptosis by regulating the protein expression levels of caspases, poly-(ADP-ribose) polymerase and the B-cell lymphoma-2-associated X-protein/B-cell lymphoma-extra large ratio. Furthermore, polyphenolic compounds inhibited mitochondrial membrane potential activity. Caspase-3 activity was increased in a dose-dependent manner and polyphenolic compounds inhibited the activation of protein kinase B by dephosphorylation. These results suggest that polyphenolic compounds in A549 cells indicate the anticancer activity through the induction of apoptosis.</P>
