Roles of Immunohistochemical Staining in Diagnosing Pulmonary Squamous Cell Carcinoma

Background: Differentiating morphologic features based on hematoxylin-eosin (HE) staining is the most common method to classify pathological subtypes of non-small-cell lung cancer (NSCLC). However, its accuracy and inter-observer reproducibility in pathological diagnosis of poorly differentiated NSCLC remained to be improved. Materials and Methods: We attempted to explore the role of immunohistochemistry (IHC) staining in diagnosing pulmonary squamous cell carcinoma (SQCC) with poorly differentiated features by HE staining or with elevated serum adenocarcinoma-specific tumor markers (AD-TMs). We also compared the difference of epidermal growth factor receptor (EGFR) mutation rate between patients with confirmed SQCC and those with revised pathological subtype. Logistic regression analyses were used to test the association between different factors and diagnostic accuracy. Results: A total of 132 patients who met the eligible criteria and had adequate specimens for IHC confirmation were included. Pathological revised cases in poor differentiated subgroup, biopsy samples and high-level AD-TMs cases were more than those with high/moderate differentiation, surgical specimens and normal-level AD-TMs. Moreover, biopsy sample was a significant factor decreasing diagnostic accuracy of pathological subtype (OR, 4.037; 95% CI 1.446-11.267, p=0.008). Additionally, EGFR mutation rate was higher in patients with pathological diagnostic changes than those with confirmed SQCC (16.7% vs 4.4%, p=0.157). Conclusions: Diagnosis based on HE staining only might cause pathological misinterpretation in NSCLC patients with poor differentiation or high-level AD-TMs, especially those with biopsy samples. HE staining and IHC should be combined as pathological diagnostic standard. The occurrence of EGFR mutations in pulmonary SQCC might be overestimated.

Load forecasting has always been essential to the operation and planning of power systems in deregulated electricity markets. Various methods have been proposed for load forecasting, and the neural network is one of the most widely accepted and used techniques. However, to obtain more accurate results, more information is needed as input variables, resulting in huge computational costs in the learning process. In this paper, to reduce training time in multi-layer perceptron-based short-term load forecasting, a graphics processing unit (GPU)-based computing method is introduced. The proposed approach is tested using the Korea electricity market historical demand data set. Results show that GPU-based computing greatly reduces computational costs.

Background/Aims To determine the value of salivary pepsin in discriminating sub-types of gastroesophageal reflux disease (GERD) and GERD-related disorders. Methods Overall, 322 patients with different sub-types of GERD and 45 healthy controls (HC) were studied. All patients took Gastroesophageal Reflux Disease Questionnaire (GerdQ) and underwent endoscopy and 24-hour esophageal pH monitoring and manometry. Salivary pepsin concentration (SPC) was detected by using colloidal gold double-antibody immunological sandwich assay. Oral esomeprazole treatment was administrated in the patients with non-erosive reflux disease (NERD) and extra-esophageal symptoms (EES). Results Compared to HC, patients with erosive esophagitis, NERD, EES, EES plus typical GERD symptoms, or Barrett’s esophagus had a higher prevalence of saliva and SPC (all P < 0.001). There was no significant difference in the positive rate for pepsin in patients with functional heartburn or GERD with anxiety and depression, compared to HC. After esomeprazole treatment, the positive rate and SPC were significantly reduced in NERD (both P < 0.001) and in EES (P = 0.001 and P = 0.002, respectively). Of the 64 NERD patients, 71.9% (n = 46) were positive for salivary pepsin, which was significantly higher than the rate (43.8%, n = 28) of pathological acid reflux as detected by 24-hour esophageal pH monitoring (P = 0.002). Conclusions Salivary pepsin has an important significance for the diagnosis of GERD and GERD-related disorders. Salivary pepsin and 24-hour esophageal pH monitoring may complement with each other to improve the diagnostic efficiency.
The dynamic stability analysis of guyed masts under random wind loads

On the basis of the first Lyapunov stability theory, this paper develops a dynamic stability criterion for elastic structural systems under arbitrary dynamic loads, and shows the stability criterion using energy variation. After the dynamic stability criterion is validated through a classic example, it is used for the dynamic stability investigation of practical guyed masts under random wind loads. The criterion is reliable, simple and of advantage for structures with large number of elements and nodes. The slack guys and internal resonance between guys and mast are two main factors which induces the dynamic instability of guyed masts, at the same time, some dynamic stability characteristics of guyed masts are found.
Improved Longitudinal Displacement Profile and Initial Support for Tunnel Excavation

The ground support system has a significant impact on tunnel safety while the installation time of initial support is commonly designed by empirical method. The three dimensional numerical model, based on the engineering practice, is analyzed by FLAC3D. In this paper, an improved LDP (longitudinal displacement profile) equation is proposed, which is more accurate in describing the convergence of tunnel before excavation face reaching. Then, a methodology is presented to determine the installation location of initial support in a more efficient and effective way. By simulating the process of excavation load releasing, the appropriate time for installing initial support is when the evolution of surrounding rock displacement has a tendency to increase rapidly, namely that the stability of rock mass arrives at the critical state. The mathematical relationship between the convergence of tunnel and the relative location to tunnel face can be established by fitting monitoring data with the improved LDP equation. Substituting the critical tunnel convergence into the LDP relationship, the corresponding installation location of initial support will be calculated. Meanwhile, several schemes with diverse geological conditions are carried out to validate the improved LDP and the methodology.
A Variational Framework for Single Image Dehazing Based on Restoration

The single image dehazing algorithm in existence can satisfy the demand only for improving either the effectiveness or efficiency. In order to solve the problem, a novel variational framework for single image dehazing based on restoration is proposed. Firstly, the initial atmospheric scattering model is transformed to meet the kimmel`s Retinex variational model. Then, the green light component of image is considered as an input of the variational framework, which is generated by the sensitivity of green wavelength. Finally, the atmospheric transmission map is achieved by multi-resolution pyramid reduction to improve the visual effect of the results. Experimental results demonstrate that the proposed method can remove haze effectively with less memory consumption.

Background/Aims High-resolution anorectal manometry (HRAM) has been considered a first-line diagnostic tool for functional defecation disorder. However, clinical studies on HRAM used in constipation patients are very limited and few studies have reported the characteristics of anorectal pressure in Chinese patients. The aim of this study is to investigate the characteristics of motility data in a cohort of Chinese patients with functional constipation. Methods A total of 82 consecutive patients with functional constipation who underwent a standardized HRAM were retrospectively enrolled in this study. The functional defecation disorder was classified into Rao’s types. Results The mean age of 82 patients was 51 years (range, 16–83 years). Indications for anorectal manometry were functional constipation for all patients. The mean resting pressure was 69.2 ± 21.2 mmHg (range, 24.5–126.9 mmHg). The mean maximum squeezing pressure was 198.4 ± 75.6 mmHg (range, 54.2–476.9 mmHg). The mean length of the anal high pressure zone was 3.4 ± 1.0 cm (range, 0.6–4.9 cm). Sixty (73.2%) patients were diagnosed as functional defecation disorder. In attempted defecation, type I was most common (n = 24), followed by type II (n = 12), type III (n = 11), and type IV (n = 13) that were present on HRAM according to Rao’s classification. In all 60 patients with functional defecation disorder, 37 were women and 23 were men. Men were significantly more likely than women to have functional defecation disorder (92.0% vs 64.9%, P = 0.014). Conclusion HRAM could be used as a test for the diagnosis of functional defecation disorder and functional defecation disorder is common in Chinese patients with functional constipation.

Xanthomonas oryzae pv. oryzae (Xoo) strains are plant pathogenic bacteria that can cause serious blight of rice, and their virulence towards plant host is complex, making it difficult to be elucidated. Caenorhabditis elegans has been used as a powerful model organism to simplify the host and pathogen system. However, whether the C. elegans is feasible for studying plant pathogens such as Xoo has not been explored. In the present work, we report that Xoo strains PXO99 and JXOIII reduce the lifespan of worms not through acute toxicity, but in an infectious manner; pathogens proliferate and persist in the intestinal lumen to cause marked anterior intestine distension. In addition, Xoo triggers (i) the p38 MAPK signal pathway to upregulate its downstream C17H12.8 expression, and (ii) the DAF-2/DAF-16 pathway to upregulate its downstream gene expressions of mtl-1 and sod-3 under the condition of daf-2 mutation. Our findings suggest that C. elegans can be used as a model to evaluate the virulence of Xoo phytopathogens to host.
DEPDC1 is a novel cell cycle related gene that regulates mitotic progression

DEPDC1 is a recently identified novel tumor-related gene that is upregulated in several types of cancer and contributes to tumorigenesis. In this study, we have investigated the expression pattern and functional implications of DEPDC1 during cell cycle progression. Expression studies using synchronized cells demonstrated that DEPDC1 is highly expressed in the mitotic phase of the cell cycle. Immunofluorescence assays showed that DEPDC1 is predominantly localized in the nucleus during interphase and is redistributed into the whole cell upon nuclear membrane breakdown in metaphase. Subsequently, siRNA-mediated knockdown of DEPDC1 caused a significant mitotic arrest. Moreover, knockdown of DEPDC1 resulted in remarkable mitotic defects such as abnormal multiple nuclei and multipolar spindle structures accompanied by the upregulation of the A20 gene as well as several cell cycle-related genes such as CCNB1 and CCNB2. Taken together, our current observations strongly suggest that this novel cancerous gene, DEPDC1, plays a pivotal role in the regulation of proper mitotic progression.
