Hyperactivated ${\alpha}2$-6-sialylation on N-glycans due to overexpression of the Golgi enzyme ${\beta}$-galactoside: ${\alpha}2$-6-sialyltransferase (ST6Gal-I) often correlates with cancer progression, metastasis, and poor prognosis. This study was aimed to determine the association between ST6Gal-I expression and the risk of recurrence and survival of patients with localized clear-cell renal cell carcinoma (ccRCC) following surgery. We retrospectively enrolled 391 patients (265 in training cohort and 126 in validation cohort) with localized ccRCC underwent nephrectomy at a single center. Tissue microarrays were constructed for immunostaining of ST6Gal-I. Prognostic value and clinical outcomes were evaluated. High ST6Gal-I expression was associated with Fuhrman grade (p<0.001 and p=0.016, respectively) and the University of California Los-Angeles Integrated Staging System (UISS) score (p=0.004 and p=0.017, respectively) in both cohorts. Patients with high ST6Gal-I expression had significantly worse overall survival (OS) (p<0.001 and p<0.001, respectively) and recurrence free survival (RFS) (p<0.001 and p=0.002, respectively) than those with low expression in both cohorts. On multivariate analysis, ST6Gal-I expression remained associated with OS and RFS even after adjusting for the UISS score. Stratified analysis suggested that the association is more pronounced among patients with low and intermediate-risk disease defined by the UISS score. High ST6Gal-I expression is a potential independent adverse predictor of survival and recurrence in ccRCC patients, and the prognostic value is most prominent in those with low and intermediate-risk disease defined by the UISS score.

<P><B>Abstract</B></P> <P>The Archean tectonic framework of the North China Craton (NCC) involved the assembly of several micro-continents along zones of ocean closure during the late Neoarchean, followed by high-grade metamorphism associated with the final assembly and cratonization of continental blocks during Late Paleoproterozoic. Here we investigate a metamorphosed igneous suite comprising pyroxenite, gabbro and trondhjemite in association with charnockite from the margin of the Qianhuai micro-block with a view to understand the magmatic and metamorphic history and crustal evolution in the NCC during late Neoarchean. We present new data from field investigations, petrology, geochemistry and zircon U-Pb, REE and Lu-Hf analyses. Mineral phase equilibria modeling shows that the pyroxenite formed at P-T conditions of 1.5–1.6 GPa and 900–1000 °C, possibly representing underplating of high temperature basaltic magma at the base of the continental arc, with the subsequent metamorphic conditions estimated as 1.2–1.4 GPa and 840–870 °C from the metagabbros. Both metagabbro and pyroxenite show an anticlockwise P-T path characterized by initial heating followed by moderate to high-pressure metamorphism and cooling. The base of a thickened arc is a viable tectonic settings to account for this scenario. Geochemical data suggest that the pyroxenite-gabbro-trondhjemite association represent the fractionated sequence from a common mafic magma, whereas the charnockite corresponds to anhydrous lower crustal melts. The high degree of hydration of the mafic and ultramafic units suggest retrograde metamorphism under hydrous conditions associated with slab subduction. The mafic-ultramafic rocks fall within the field of island arc tholeiite and the trondhjemite falls in the field of oceanic arc basalt. Geochemical data indicate arc signature of the rocks and the subduction processes involved in the generation of precursor basaltic melts that underplated the overriding arc crust and triggered lower crustal melting.</P> <P>The U-Pb data on magmatic zircon grains from the trondhjemite, metagabbro, pyroxenite and charnockite yield emplacement ages of 2481 Ma, 2449 Ma and 2464 and 2480 Ma and suggest subduction-related magmatism during Neoarchean-early Paleoproterozoic transition, followed by high grade metamorphism at ca. 1.94 Ga as indicated by the lower intercept ages. Zircon REE data confirm the magmatic crystallization history. The zircon Lu-Hf data show consistently positive εHf(t) values (0.5–11.7) and plot between CHUR and new crust line suggesting that depleted mantle sources contributed significantly to arc building and crustal growth. The computed model ages suggest that the magma source involved Neoarchean-Mesoarchean juvenile components. Chitin deacetylase (CDA) is an insect chitin degradation enzyme that catalyzes the deacetylation of chitin to form chitosan. In this study, combination of rapid-amplification of cDNA ends (RACE) technology with Cnaphalocrocis medinalis transcriptome database analysis revealed the presence of at least five C. medinalis CDAs (CmCDAs), which were CmCDA1, CmCDA2, CmCDA4, CmCDA5, and CmCDA6. The cDNA sequences of CmCDA1, CmCDA2, and CmCDA4 hadwhole open reading frame (ORF) for further analysis. Phylogenetic analysis indicated that insect CDAs could be categorized into five groups. CmCDAs' structural domain analysis revealed that all three CDAs contained the chitin deacetylase-like catalytic domain. CmCDA1 and CmCDA2 belong to Group I because they both contain the chitin-binding peritrophin-A domain (ChBD), low-density lipoprotein receptor class A domain (LDLa), and chitin deacetylase-like catalytic domain. CmCDA4 only contains ChBD and chitin deacetylase-like catalytic domain thus belongs to Group III. Tissue and developmental stage expression analysis showed that the expression levels of CmCDA1, CmCDA2, and CmCDA4 are significantly higher in the head than other tissues and also significantly higher in adults than in larvae. CmCDA5 had significantly higher expression in the integument than other tissues, suggesting potential roles in the process of degradation of chitin. In contrast, CmCDA5 showed relatively high expression in larvae. In conclusion, this study analyzed the cDNA sequences of three CDA genes and determined their expression and molecular characteristics, which provided a new sequence resource and improved the development of bio-pesticides and the biological pest control and contributed to management of this important agricultural pest.
Serum Carbohydrate Antigen 19-9 as an Indicator of Liver Metastasis in Colorectal Carcinoma Cases

Purpose: The liver is the organ to which colorectal carcinomas (CRCs) most commonly metastasize, and surgical resection has been established as the most effective and potentially curative treatment for CRC with liver metastasis (LM). Therefore, surveillance of LM is vital for improvement of prognosis of CRC patients. In this study, we aimed to explore the potential value of carbohydrate antigen 19-9 (CA 19-9), carcinoembryonic antigen (CEA), and marker enzymes in indicating LM with CRC. Methods: Three groups of eligible patients with metastatic cancers were retrospectively included: CRC patients with LM (CRC-LM) or without LM (CRC-NLM), and non-CRC patients with LM (NCRC-LM). All metastatic lesions were identified by CT or MRI. Data on characteristics of the patients, the primary site, the locations of metastasis, CA 19-9, CEA, and biochemical parameters were collected for analysis. Results: A total of 493 patients were retrospectively included. More alcohol consumption was found in CRC-LM than CRC-NLM. Some biochemical enzymes were found to be significantly higher in groups with LM than without (CRC-LM or NCRC-LM v.s CRC-NLM). Both CEA and CA 19-9 were much higher in CRC-LM than CRC-NLM or NCRC-LM. For CRC patients, CA 19-9, ${\gamma}$-glutamyl transpeptidase, CEA and alcohol consumption were identified as independent factors associated with LM. Conclusion: Our analysis suggested the CA 19-9 might be a potential valuable indicator for LM of CRC in the clinic.

1-Deoxynojirimycin (DNJ) as a kind of alkaloid has been confirmed that could modulate glycometabolism andhas toxicity for the eri-silkworm in our previous research. On the contrary, what is the potentially defensivemechanism when the DNJ enters the eri-silkworm. Based on comparative transcriptome sequencing, we foundthat ATP-binding cassette (ABC) transporter genes could be induced significantly by DNJ. In this study, a total of16 putative ABC transporter genes were identified, which can be classified into seven subfamilies, namely oneABCA, four ABCBs, three ABCCs, two ABCDs, one ABCE, three ABCFs, and two ABCGs. Phylogenetic analysisrevealed that ScABCs had strong conservation with Bombyx mori. Reverse transcription quantitative PCR (RTqPCR)suggested that 6 ABC transporters had a strong positive correlation between RT-qPCR and transcriptomedata. Additionally, S. c. ricini ABC transporter C-subfamily 4 (ScABCC4), S. c. ricini ABC transporter G-subfamily4 (ScABCG4), S. c. ricini ABC transporter A-subfamily 3 (ScABCA3) and S. c. ricini ABC transporter C-subfamily10 (ScABCC10) showed different expression pattern in two feed dose (1% and 2% DNJ) and three time points(6h, 12 h, 48 h). This study provides the first study on identification, characterization and expression patterns ofABC transporter genes in S. c. ricini response to DNJ, and lays a foundation for further understanding of theirphysiological roles response to the alkaloid.
Apoptosis of Colorectal Cancer UTC116 Cells Induced by Cantharidinate

Effects of Cantharidinate on apoptosis of human colorectal cancer UTC-116 cells were investigated by means of 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay, H and E staining, flow cytometry, and Raman Spectra analysis. The results showed Cantharidinate to exert inhibitory action on proliferation of human colorectal cancer UTC-116 cells, inducing apoptosis, arresting cells in G1 phase, with decline of S and G2 phases. In addition, the results of Raman spectrum showed significant changes in the UTC-116 cells chemical structure with stretching after the application of Cantharidinate. Taken together, these results suggest that the treatment of human colorectal cancer with Cantharidinate may be associated with multiple molecular mechanisms for apoptosis. Furthermore, similar to fluorouracil, Cantharidinate should be considered as novel assistant drug for controlling the growth of human colorectal cancer UTC-116 cells.
