        Aeromicrobium endophyticum sp. nov., an endophytic actinobacterium isolated from reed (Phragmites australis)

        A Gram-staining-positive, motile and short-rod-shaped actinobacterium designated 9W16Y-2T was isolated from surface- sterilized leaves of reed (Phragmites australis) collected from Taklamakan Desert in Xinjiang Uygur Autonomous Region, China. Colonies were pale greenish yellow, circular, smooth, and convex. The 16S rRNA gene sequence of strain 9W16Y-2T exhibited highest sequence similarities with Aeromicrobium camelliae CGMCC 1.12942T (99.0%) and Aeromicrobium erythreum NRRL B-3381T (97.2%). Phylogenetic analyses based on 16S rRNA gene sequences and single-copy phylogenetic marker genes (pMGs) showed that strain 9W16Y- 2T belonged to the genus Aeromicrobium and formed a monophyletic clade with Aeromicrobium camelliae CGMCC 1.12942T. Furthermore, average nucleotide identity (ANI) and DNA-DNA hybridization (DDH) clearly separated strain 9W16Y-2T from the other species of the genus Aeromicrobium with values below the thresholds for species delineation. The G+C content of the genomic DNA is 68.9 mol%. The diagnostic diamino acid of the cell-wall peptidoglycan was LLdiaminopimelic acid. The predominant menaquinone was MK-9(H4). The major fatty acids (> 10% of the total fatty acids) were C18:0 10-methyl (TBSA) (28.2%), C16:0 (21.0%), C16:0 2-OH (20.8%) and C18:1 ω9c (12.8%). The polar lipid profile comprised diphosphatidylglycerol, phosphatidylglycerol, phosphatidylcholine, phosphatidylinositol, an unidentified aminophospholipid and an unidentified lipid. Based on the phylogenic, phenotypic and chemotaxonomic features, strain 9W16Y-2T represents a novel species of the genus Aeromicrobium, for which the name Aeromicrobium endophyticum sp. nov. is proposed. The type strain is 9W16Y-2T (= CGMCC 1.13876T = JCM 33141T).

        ACOX1 destabilizes p73 to suppress intrinsic apoptosis pathway and regulates sensitivity to doxorubicin in lymphoma cells

        Lymphoma is one of the most curable types of cancer. However, drug resistance is the main challenge faced in lymphoma treatment. Peroxisomal acyl-CoA oxidase 1 (ACOX1) is the rate-limiting enzyme in fatty acid β-oxidation. Deregulation of ACOX1 has been linked to peroxisomal disorders and carcinogenesis in the liver. Currently, there is no information about the function of ACOX1 in lymphoma. In this study, we found that upregulation of ACOX1 promoted proliferation in lymphoma cells, while downregulation of ACOX1 inhibited proliferation and induced apoptosis. Additionally, overexpression of ACOX1 increased resistance to doxorubicin, while suppression of ACOX1 expression markedly potentiated doxorubicin-induced apoptosis. Interestingly, downregulation of ACOX1 promoted mitochondrial location of Bad, reduced mitochondrial membrane potential and provoked apoptosis by activating caspase-9 and caspase-3 related apoptotic pathway. Overexpression of ACOX1 alleviated doxorubicin-induced activation of caspase-9 and caspase-3 and decrease of mitochondrial membrane potential. Importantly, downregulation of ACOX1 increased p73, but not p53, expression. p73 expression was critical for apoptosis induction induced by ACOX1 downregulation. Also, overexpression of ACOX1 significantly reduced stability of p73 protein thereby reducing p73 expression. Thus, our study indicated that suppression of ACOX1 could be a novel and effective approach for treatment of lymphoma. [BMB Reports 2019; 52(9): 566-571]

      • Prognostic Role of Hypoxic Inducible Factor Expression in Non-small Cell Lung Cancer: A Meta-analysis

        Introduction: Reported prognostic roles of hypoxic inducible factor (HIF) expression in non-small cell lung cancer (NSCLC) have varied. This meta-analysis aimed to examine the relationship between HIF expression and clinical outcome in NSCLC patients. Methods: PubMed were used to identify relevant literature with the last report up to December $20^{th}$, 2012. After careful review, survival data were collected from eligible studies. We completed the meta-analysis using Stata statistical software (Version 11) and combined hazard ratio (HR) for overall survival (OS). Subgroup specificity, heterogeneity and publication bias were also assessed. All of the results were verified by two persons to ensure accuracy. Results: Eight studies were finally stepped into this meta-analysis in which seven had available data for HIF-$1{\alpha}$ and three for HIF-$2{\alpha}$. Combined HRs suggested that higher expression of $HIF1{\alpha}$ had a negative impact on NSCLC patient survival (HR=1.50; 95%CI=1.07-2.10; p=0.019). The expression of HIF-$2{\alpha}$ was also relative to a poorer survival (HR=2.02; 95%CI=1.47-2.77; p=0.000). No bias existed in either of the two groups. Conclusion: This study suggests that elevations of HIF-$1{\alpha}$ and HIF-$2{\alpha}$ expression are both associated with poor outcome for patients with NSCLC. The data support further and high quality investigation of HIF expression for predicting poor outcome in patients with NSCLC.

      • Theoretical and Numerical Analysis of 1 : 1 Main Parametric Resonance of Stayed Cable Considering Cable-Beam Coupling

        <P>For the 1 : 1 main parametric resonances problems of cable-bridge coupling vibration, a main parametric resonances model considering cable-beam coupling is developed and dimensionless parametric resonances differential equations are derived. The main parametric resonances characteristics are discussed by means of multiscale approximation solution methods. Using an actual cable of cable-stayed bridge project for research object, numerical simulation analysis under a variety of conditions is illustrated. The results show that when the coupling system causes 1 : 1 parametric resonance, nonlinear main parametric resonances in response are unrelated to initial displacement of the cable, but with the increase of deck beam end vertical initial displacement increases, accompanied with a considerable “beat” vibration. When the vertical initial displacement of deck beam end is 10−6 m order of magnitude or even smaller, “beat” vibration phenomenon of cable and beam appears. Displacement amplitude of the cable is small and considerable amplitude vibration may not occur at this time, only making a slight stable “beat” vibration in the vicinity of the equilibrium position, which is different from 2 : 1 parametric resonance condition of cable-bridge coupling system. Therefore, it is necessary to limit the initial displacement excitation amplitude of beam end and prevent the occurrence of amplitude main parametric excitation resonances.</P>

      • Synergistic Increase of Oxidative Stress and Tumor Markers in PAH-Exposed Workers

        In this study, we investigated oxidative stress and tumor marker levels of polycyclic aromatic hydrocarbons (PAHs) in 136 coke oven workers and in 60 control subjects, and evaluated the correlation between oxidative stress and tumor marker levels. Questionnaires on basic demographic information were also administered. Significant differences in employment time and percentages of alcohol drinkers were observed between the control and exposed groups. PAH exposure was assessed using urinary 1-hydroxy-pyrene (1-OHP) levels and was found to be significantly higher in workers than in the controls. Significant differences (P<0.001) of MDA, GST, LDH, NSE, Cyfra21-1, and of SCC and TNF-a (P<0.0001 and P<0.05, P<0.001, respectively) levels were observed among controls and coke-oven workers, except for bottom coke oven workers. Associations between age and risk of increased TNF-a, smoking and increased GST activities, and drinking with increased MDA concentrations, were marginal (P=0.055, P=0.048, P=0.057, respectively). The association between smoking with MDA (P=0.004), NSE (P=0.005), SCC (P=0.004) andTNF-a (P<0.001), and drinking with TNF-a levels was significant (P=0.012). In addition, a significant positive correlation between oxidative stress and tumor markers was found in the present study. These results suggest that a synergistic increase of oxidative stress and tumor markers induced by PAHs may play a role in toxic responses for PAHs in coke oven workers.

        Background Bladder cancer is the 10th common cancer worldwide. Osteopontin has been found to enhance cell proliferation, metastasis and invasion in various human tumors. Objective To investigate the roles of osteopontin in bladder cancer. Methods The RNA interference and overexpression of osteopontin were performed in bladder cancer cell lines (T24 and SCaBER). Cell proliferation and apoptosis were measured using CCK-8 assay and flow cytometry, respectively. Cell invasion was determined using transwell assay. Results Osteopontin was highly expressed in bladder cancer tissues in comparison with the adjacent normal tissues. Its high expression significantly correlated with high histologic grade, high TNM stage (III and IV) and poor prognosis. For T24 cells with osteopontin interference and SCaBER cells with osteopontin overexpression, cell proliferation was significantly inhibited (3.58-fold vs. 5.62-fold) and enhanced (7.81-fold vs. 5.29-fold), respectively. The apoptosis portion of T24 cells significantly increased from 4.48 to 10.75%, and that of SCaBER cells significantly declined from 7.33 to 4.01%. The invaded T24 and SCaBER cells significantly decreased to 52.0% and increased to 2.0-fold, respectively. Osteopontin overexpression enhanced the expression (1.54-fold and 2.39-fold; 2.33-fold and 2.05-fold) and activation (1.80-fold and 1.96-fold; 2.00-fold and 2.59-fold) of JAK1 and STAT1 in two cell lines of bladder cancer. Conclusion Osteopontin might enhance proliferation, inhibit apoptosis and accelerate invasion and thus promote the development and metastasis of bladder cancer, and osteopontin’s functions might be mediated by activating JAK1/STAT1 signaling pathway.

        Recent progresses and perspectives of VN-based materials in the application of electrochemical energy storage

        Developing well-behaved electrode materials is crucial for manufacturing high-performance batteries,SCs and electrocatalysis, and it is also one of the main driving forces for the development of EES devices. In consideration of the high robustness and electrical conductivity during the electrochemical reactionprocess, VN-based materials exhibit good performance as electrode materials (or catalytic materials)for batteries, supercapacitors, and excellent catalytic activity for electrocatalysts. Especially, the VNbasedmaterials decorated into other active compounds with various morphologies elaborately presentexcellent performances due to the abundant active sites and fully synergistic effect. This review presentsthe structure-performance relation by designing different-dimensional nanostructures and various applicationsof VN-based materials in electrochemical energy storage (EES) applications. Finally, the perspectiveson future challenges and progress have been discussed. This review can offer a specificunderstanding for the optimization strategies of VN-based materials, thus booming the rapid developmentand practical applications for EES devices in the future.

        P-Selectin-mediated Acute Inflammation Can Be Blocked by Chemically Modified Heparin, RO-Heparin

        Selectins are carbohydrate-binding cell adhesion molecules that play a major role in the initiation of inflammatory responses. Heparin can bind to P-selectin, and its anti-inflammatory property is mainly due to inhibition of P-selectin. However, the strong anticoagulant activity of heparin limits its clinical use. We prepared periodate-oxidized, borohydride-reduced heparin (ROheparin) by chemical modification and tested its anticoagulant and anti-inflammatory activities. Activated partial thromboplastin time (aPTT) assays showed that, compared with heparin, RO-heparin had greatly reduced anticoagulant activity. Intravenous administration of this compound led to reduction in the peritonealinfiltration of neutrophils in a mouse acute inflammation model. In vitro cell adhesion experiments demonstrated that the effect of RO-heparin on inflammatory responses was mainly due to inhibiting the interaction of P-selectin with its ligands. These results indicate that RO-heparin may be a safer treatment for inflammation than heparin, especially when selectin is targeted.

        An integrated strategy for production of four anthocyanin compounds from Ribes nigrum L. by deep eutectic solvents and flash chromatography

        This study explored an integrated strategy for extraction and preparative separation of four anthocyaninsfrom Ribes nigrum L. using DES-based microwave assisted extraction andflash chromatography. Theextraction yield of anthocyanins reached 2.03 mg/g after optimization. An efficient preparativeseparation process of four main anthocyanins by macroporous resin andflash chromatography wasestablished afterwards. The yields of D3G, D3R, C3G and C3R reached 0.32 mg/g, 0.47 mg/g, 0.18 mg/g and0.43 mg/g with the purities of more than 95%, respectively. The developed procedure represents anefficient and convenient alternative for the systematic utilization of natural products from plantmaterials.

        ONE-POT SYNTHESIS OF INTERESTING 3D γ-MnOOH NETWORKS

        Three-dimensional (3D) γ-MnOOH networks are successfully prepared by one-pot solvothermalmethod without using any catalyst. The samples are characterized by X-ray diffraction (XRD),scanning electron microscopy (SEM) and high-resolution transmission electron microscopy(HRTEM). It is found that the amounts of urea and H2O2 added, reaction temperature and timehave important infuences on the samples. It is interesting that the 3D networks are formed fromthe oriented attachment (OA) of Mn3O4 octahedrons; and that the phase transformation from Mn3O4 to γ-MnOOH occurs via the protonation of Mn3O4. This study is expected to offer a facileapproach to the syntheses of new, intricate nanostructures.
