:Watermelon (Citrullus lanatus) is an important vegetable crop in the world and contains various health promoting compounds including many antioxidants such as lycopene and beta-carotene. Genetic diversity of cultivated watermelon is very narrow, which is one of major factors limiting the improvement of watermelon breeding program. Gummy stem blight (Didymella bryoniae (Auersw.)) is one of the most destructive diseases and causes crown blight, defoliation and fruit rot, resulting in severe production loss in watermelon. Single nucleotide polymorphisms (SNPs), important molecular markers for genetic studies are frequent sequence variation in the plant genome. This study was performed to develop SNP markers related to gummy stem blight resistance from three watermelon lines, one susceptible and two resistant to gummy stem blight. Of them, 22 SNP primers which were sequence information on SNPs derived from comparing a resistant and susceptible line was used for high resolution melt analysis. A total of 14 candidate SNP primers were detected melt temperature difference. These SNP primers can be useful for developing disease resistance-related markers and, therefore, beneficial to watermelon breeding program.

Tribbles homolog 2 (TRIB2) is implicated in tumorigenesis and drug resistance in various types of cancers. However, the role of TRIB2 in the regulation of tumorigenesis and drug resistance of cancer stem cells (CSCs) is still elusive. In the present study, we showed increased expression of TRIB2 in spheroid-forming and aldehyde dehydrogenase-positive CSC populations of A2780 epithelial ovarian cancer cells. Short hairpin RNA-mediated silencing of TRIB2 expression attenuates the spheroid-forming, migratory, tumorigenic, and drug-resistant properties of A2780 cells, whereas overexpression of TRIB2 increases the CSC-like characteristics. TRIB2 overexpression induced GSK3β inactivation by augmenting AKT-dependent phosphorylation of GSK3β at Ser9, followed by increasing β-catenin level via reducing the GSK3β-mediated phosphorylation of β-catenin. Treatment of TRIB2-ovexpressed A2780 cells with the phosphoinositide3-kinase inhibitor LY294002 abrogated TRIB2-stimulated proliferation, migration, drug resistance of A2780 cells. These results suggest a critical role for TRIB2 in the regulation of CSC-like properties by increasing the stability of β-catenin protein via the AKT-GSK3β-dependent pathways.
MAB SNP marker development to accelerate the breeding of Chinese cabbage

The goal of marker-assisted backcrossing is to reduce the number of generations significantly by using genome-based molecular markers. Among other types of molecular markers, SNP (single nucleotide polymorphism) is mostly used in genetic diversity analysis due to its abundance. To develop high-throughput SNP marker for MAB system, we selected 20 Chinese cabbage lines each represent traits as inner leaf color, disease resistance, head type and maturity etc. Then, we sequenced the transcriptomes of 20 lines by using Illumina Hiseq2000. The average transcriptome size was 1.37 Gbase, and the average of short reads mapping rate was about 62.15% (30xcoverage). We identified 13,976 SSR markers and 380,198 SNPs by aligning contigs of 20 Chinese cabbage lines. To develop SNP marker set, we chose 409 SNPs that covers the whole Brassica rapa transcriptome. The filtering criteria were depth, polymorphism, segregation ratio, lack of adjacent SNP and copy number. We positioned the selected SNP markers to the Chinese cabbage linkage map. Clustering dendrogram was produced using SNP marker and three different clusters were generated. The result showed that the genotyping data is partially linked to the phenotyping data. We assume that the developed SNP marker set can be applied in the Chinese cabbage MAB system soon.

Watermelon (Citrullus lanatus) is one of the most economically important cucurbitaceous crop over the world. Screening of proper reference genes was needed to reverse transcription quantitative real-time PCR (qRT-PCR), and it is bausic step of many researches including gene expression analysis. However, the reference genes on watermelon has not yet been reported systematically. Therefore, eight candidates of reference genes were selected with reference to Arabidopsis or cucumber papers. They are β-Actin, elongation factor 1-α, glyceraldehy-3-phosphate-dehydrogenase, NADP-isocitrate dehydrogenase, leunig, polypyrimidine tract-binding protein1, ubiquitin-conjugating eznyme E2, and 18S ribosomal RNA. The expression levels of genes were evaluated by qRT-PCR under biotic stress (Colletotrichum orbiculare treatment), plant hormone treatment (100 μM ABA), and abiotic stresses such as drought, cold (4℃), salt (250 mM NaCl) stresses. We founded appropriate reference genes which did not induce or reduce gene expression levels under broad spectrum of stresses by qRT-PCR analysis. These results may provide proper information for the use of appropriate reference genes for gene expression studies in watermelon qRT-PCR analysis.

충청남도 기초자치단체 교육경비 보조사업 제한의 유효성 분석

본 연구는 충청남도 기초자치단체 교육경비 보조 사업 제한의 유효성을 검토하기 위해 수행되었다. 교육경비 보조사업의 제한은 ‘지방자치단체 중 지방세와 세외수입으로 소속공무원의 인건비를 충당하지 못하는 경우에 교육경비 보조를 할 수 없게 한’ 제도이다. 이 제도에 따라 중앙정부는 인건비를 충당하지 못하는 지방자치단체에 교육경비 지원을 못 하도록 하였고, 지방자치단체는 직접 지원하는 교육경비 보조금을 축소하였으나 장학회를 통해 우회적으로 지원하는 경비를 늘려 왔다. 충청남도 인건비 미충당 기초자치단체인 청양과 서천이 장학회를 통해 학교의 교육경비를 보조하고 있으며, 서천군의 경우는 이의 지원 규모가 직접 지원 규모를 능가하고 있다. 교육경비 보조사업의 제한 제도가 기능을 발휘하지 못하고 있는 사례이다. 충청남도 인건비 미충당 2개 자치단체의 교육경비 보조 제한의 유효성을 분석한 결과 유효성이 없음이 드러났다. 유효하지 않은 법령을 고집하면서 교육경비 보조를 못하게 하는 것은 기초자치단체에도, 학교에도 바람직하지 않다. 어려운 여건에 있는 기초자치단체에도 지역의 특성에 맞게 학교와 협력할 수 있게 교육경비 보조 사업 제한 규정을 수정할 필요가 있다. This study was conducted to examine the validity of the restriction on educational expenses subsidized by local governments in Chungcheongnam-do. The restriction of the education expenses subsidy project is a regulation that prohibits local governments from subsidizing education expenses if they cannot cover the labor costs of their public officials due to local taxes and non-tax revenues. The local governments suggested to the Ministry of Legislation, the Anti-Corruption and Civil Rights Commission and the Ministry of Education that restrictions on educational expenses are unfair to create areas that return to develop regional education, but no solution has been reached. Rather, the Ministry of Public Administration and Security strongly controlled local governments not to subsidize educational expenses in order to ensure sound financial management of local governments that could not solve labor costs. In 2013, the Act on the Collection of Non-Tax Revenue was enacted to reduce the size of local governments’ own income by excluding surplus, carryover, deposit, and loan principal income. Under the central government’s strong administrative guidance, local governments that do not have sufficient labor costs have reduced subsidies directly to schools, but instead are increasing expenses provided through scholarship associations. Cheongyang and Seocheon, the basic autonomous governments that do not cover labor costs in Chungcheongnam-do, are subsidizing educational expenses of schools through scholarships, and in the case of Seocheon-gun, the amount of support exceeds the amount of direct support. This is an example of proving that the restrictions on educational expense assistance projects are not functioning properly. Support for educational expenses subsidies through scholarship associations cannot be considered a reasonable process. Since the scholarship association is a corporation operated by the fund, it can be run laxly and can be executed by the heads of local governments without deliberation and resolution. The two local governments in Chungcheongnam-do have analyzed the validity of the restriction on educational expenses and found that it is not valid. It is not desirable for local governments and education to waste administrative power while insisting on invalid laws. Even in the case of basic autonomous governments in difficult conditions by revising the statutes, it is necessary to revise the regulations on the subsidy of educational expenses so that education can be developed in cooperation with schools according to local characteristics.
Identification of multiple nuclear localization signals in murine Elf3, an ETS transcription factor

<P><B>Abstract</B></P><P>We investigated nuclear localization signal (NLS) determinants within the AT-hook and ETS DNA-binding domains of murine Elf3 (mElf3), a member of the subfamily of epithelium-specific ETS transcription factors. Deletion mutants containing the AT-hook, ETS domain or both localized strictly in the nucleus, suggesting that these individual domains contain independent NLS motif(s). Within the AT-hook domain, four basic residues (<SUP>244</SUP>KRKR<SUP>247</SUP>) were critical for strong NLS activity, and two potent bipartite NLS motifs (236–252 and 249–267) were sufficient for nuclear import of mElf3, although less efficient than the full domain. In addition, one stretch of basic residues (<SUP>318</SUP>KKK<SUP>320</SUP>) within the ETS domain appears to be essential for mElf3 nuclear localization. Taken together, mElf3 contains multiple NLS motifs, which may function cooperatively to effect efficient nuclear transport.</P>
