The aim of this study was to investigate the use and the satisfaction of fresh convenience foods, specifically fresh-cut fruits and vegetables according to customer health consciousness. The survey included a total of 1,132 subjects (568 men and 564 women). Subjects were classified into two groups: health conscious consumers (HCC)(n=865) and non-health conscious consumers (NHCC)(n=267). When choosing convenience foods, the HCC group had a higher percentage of respondents who considered eco-labels(p<0.05), while showing a significantly lower proportion of those who considered quantity/bundle size (p<0.05) than the NHCC group did. Overall satisfaction with fresh cut fruits and vegetables was 3.75 points in the HCC group, and this was significantly higher than 3.61 points in the NHCC group (p=0.006). The study results indicate when the customers are health conscious, they are more likely to consider health promoting factors when purchasing fresh convenience foods, resulting in higher level of satisfaction. This study evaluated the quality properties of naturally cured sausages with pre-converted vegetable solution (PCS) including two types of starter cultures. The PCS consisted of Chinese cabbage powder, starter culture, and water. As starter cultures, Staphylococcus hominis (SH) and Staphylococcus carnosus (SC) were used. Samples were included: negative control (NC, no NaNO₂); positive control (PC, 70 ppm NaNO₂); TRT 1 (6.29% PCS-SH, 35 ppm NaNO₂); TRT 2 (5.98% PCS-SC, 35 ppm NaNO₂); TRT 3 (12.50% PCS-SH, 70 ppm NaNO₂); and TRT 4 (11.89% PCS-SC, 70 ppm NaNO₂). TRTs 3 and 4 had the highest (P<0.05) cooking loss, probably due to lower (P<0.05) pH. However, All TRTs showed similar (P>0.05) CIE a* to PC. Although TRTs 3 and 4 contained the same concentration of NaNO₂ as PC, residual nitrite in PC was the highest (P<0.05), but they showed higher (P<0.05) nitrosyl hemochrome and curing efficiency than PC. Hardness, gumminess, and chewiness were increased (P<0.05) with the level of PCS. Consequently, the PCS levels may affect the cured color developments of naturally cured sausages, whereas types of starter cultures had limited effects.
Quercetin prevents adipogenesis by regulation of transcriptional factors and lipases in OP9 cells

<P>With the industrialization of society, the increase in the prevalence of obesity and metabolic disorders has become an important health concern in a number of countries. Quercetin (3,30,40,5,7-pentahydroxyflavone) is well known as a bioactive flavonoid in a variety of biological resources. The aim of the present study was to explore the machanisms responsible for the anti-adipogenic activity of quercetin and its effects on the lipolysis in OP9 mouse stromal cells which rapidly differentiate into adipocytes. The differentiation of OP9 cells into adipocytes was evaluated by the measurement of lipid accumulation by Oil Red O (ORO) staining; lipid accumulation was significantly impaired by treatment with quercetin. Reverse transcription-polymerase chain reaction (RT-PCR) and western blot analysis were used to measure the expression levels of CCAAT/enhancer binding protein α (C/EBPα), proliferator-activated receptor γ (PPARγ), sterol regulatory element-binding protein-1 (SREBP-1) and fatty acid synthase (FAS). The mRNA expression levels of lipases, such as adipose triglyceride lipase (ATGL), hormone sensitive lipase (HSL) and lipoprotein lipase (LPL) were also measured by RT-PCR. Quercetin significantly decreased the expression of transcription factors, including C/EBPα, PPARγ and SREBP-1c both at the protein and mRNA level. The results from the present study demonstrate that quercetin prevents adipogenesis by upregulating ATGL and HSL expression and downregulating FAS, LPL and adipocyte fatty acid-binding protein (aP2) expression, as well as the expression of transcription factors. Our data suggest that quercetin has therapeutic potential by regulating the expression of transcriptional factors and enzymes associated with adipogenesis.</P>

Objective: SIRT6 involves in various cellular signaling including tumorigenesis in association with -catenin. However, the role of SIRT6 in tumorigenesis was reported controversially and the reports for the role of SIRT6 in ovarian cancers is limited. Therefore, we evaluated expression and roles of SIRT6 and -catenin in ovarian carcinomas and ovarian cancer cells. Methods: In OVCAR3 and OVCAR5 ovarian cancer cells, knock-down of SIRT6 did not affect for the proliferation of cells. However, knock-down of SIRT6 significantly inhibited migration and invasion activity of OVCAR3 and OVCAR5 cells. SIRT6-mediated invasiveness of ovarian cancer cells were related with the expressions of epithelial-to-mesenchymal transition related signaling such as snail, vimentin, N-cadherin, E-cadherin, and activated -catenin. Especially, SIRT6 overexpression mediated increase of invasiveness and activation of epithelial-to-mesenchymal transition signaling was attenuated by knock-down of - catenin. Results: In 104 human ovarian carcinomas, nuclear expression of SIRT6 (P = 0.010) and activated -catenin (P = 0.013) were significantly associated with shorter overall survival in multivariate analysis. In 63 high-grade serous carcinomas, nuclear expression of SIRT6 (P = 0.040) and active -catenin (P = 0.005) were independent indicators of shorter survival of patients. Conclusion: In conclusion, this study suggests that SIRT6--catenin signaling is involved in the epithelial-to-mesenchymal transition of ovarian cancer cells, and the expression of SIRT6 and active -catenin might be used as poor prognostic indicators of ovarian carcinoma patients. In addition, our results suggest that SIRT6--catenin signaling might be a new therapeutic target of ovarian carcinomas.
온실 지붕면의 적설 하중 수치해석을 위한 CFD-DEM 결합 모델 개발

온실은 일반 건축물에 비해 안전율이 낮고 대부분이 바람과 적설 하중에 취약한 경량구조로 이루어져 있다. 최근 이상 기후에 의한 폭설의 빈도가 증가함에 따라 온실에 발생하는 피해가 증가하고 있고, 이는 경제손실, 작물생산 중단 등 농민에게 2차적인 피해를 발생시킨다. 폭설에 의한 온실의 피해를 줄이기 위해 눈의 특성과 온실 지붕면의 적설 분포를 파악하는 것이 중요하다. 따라서 본 연구에서는 눈의 특성을 고려한 온실 지붕면의 적설 분포를 DEM 모델을 이용하여 분석하였다. 또한 CFD-DEM 결합 모델을 개발하여 풍 환경에 따른 온실 지붕면의 적설 분포와 이에 따른 압력 분포를 분석하였다. 모형실험과 수치해석기법을 이용하여 실험 환경을 제어하고, 다양한 데이터 측정 지점을 설정하여 적설분포를 모 의하였다. 경사판 (15°, 45°)과 1/25로 축소된 온실모형 (양지붕형, 복숭아형)으로 모형실험을 진행하였으며 DEM을 기반으로 한 수치해석 모델을 개발하였다. 모형실험은 농촌진흥청 농업공학부에 위치한 적설 모의실험 시설에서 눈의 밀도 및 모형에 적설 되는 눈의 분포 특성을 파악하였고, 획득한 데이터를 통해 개발된 수치해석모델을 검증하였다. 검증 결과, 15° 경사판은 0.9631, 45° 경사판은 0.9746, 양지붕형 온실모형은 0.9922, 복숭아형 온실모형은 0.9962의 R<sup>2</sup>을 나타내어 설계한 DEM 모델을 통해 적설 분포 예측이 가능할 것으로 판단되었다. 검증된 모델을 통해 풍 환경, 온실의 폭, 지붕면의 경사각, 연동 수를 고려한 실제 온실사이즈에 대한 수치해석을 진행하였다. 분석 결과, 평균 적설 하중은 양지붕형 연동온실에서 59.77Pa로 가장 높았고, 최대 하중은 복숭아형 연동온실에서 193.62Pa로 나타났다. 또한, 풍 하측의 적설 하중이 풍 상측 적설 하중보다 증가함을 나타냈다. 그러나 풍 하측의 적설하중은 풍속이 증가함에 따라 감소함을 보였다. 이러한 시뮬레이션 결과는 온실 설계 기준에 대한 참고 자료로 활용될 수 있을 것으로 판단된다.
GC-MS/MS를 이용한 농산물 중 273종 다성분 분석법 검증

본 연구는 농산물 중 273종 농약의 안전관리를 위한 잔류실태조사를 진행하기 전, 다성분 분석법을 검증하고자 수행하였다. 시료는 대표농산물 중 현미, 감귤을 선정하여 아세토니트릴로 추출한 후 d-SPE(dispersive-solid phase extraction)로 정제하였다. 시험용액을 가스크로마토그래프-질량분석기(GC-MS/MS)를 이용하여 아크리나트린을 포함한 273종 농약에 대한 분석을 실시하였다. 다성분 분석법의 유효성 검증을 위해 무처리 시료용액을 이용하여 0.005-0.2 mg/kg 범위에서 6농도로 검량선을 작성하였다. 모든 검체에서 GC-MS/MS 대상 농약의 상관계수(r 2)는 0.98 이상이었고, 정량한계(LOQ)는 0.01 mg/kg 이하로 나타났다. 정량한계를 포함한 3수준 농도의 273종 표준물질을 처리하여 회수율을 검증한 결과, 대부분의 농약에서 60∼120%의 회수율을 보였으며 상대 표준편차는 32% 이내로 나타났다. 본 연구에서 검증한 결과 273종 다성분 분석법이 농산물 중 잔류농약에 대한 잔류실태조사에 적합함을 확인하였다.
