MiR‑15a attenuates peripheral nerve injury‑induced neuropathic pain by targeting AKT3 to regulate autophagy

        Longxue Cai,Xianfa Liu,Qicai Guo,Qi huang,Qiong Zhang,Zuohong Cao 한국유전학회 2020 Genes & Genomics Vol.42 No.1

Objective Aim of this study was to detect the expression of miR-15a in rats following chronic constriction injury (CCI) and to investigate the regulatory functions of miR-15a during neuropathic pain (NP) development. Methods CCI was performed in adult Sprague–Dawley rats to set up the rat model of neuropathic pain. MiR-15a agomir and scrambled control were delivered into the implanted catheter of rats. The mechanical allodynia and thermal hyperalgesia were assessed in both CCI- and sham-operated groups. Rat lumbar spinal cord tissues were harvested for mRNA and protein analyses. The primary spinal microglia were isolated from adult Sprague–Dawley rats and transfected with miR-15a mimics, scramble miRNA, miR-15a inhibitor or its corresponding negative control. Cell lysates were collected for mRNA and protein analyses. Results Compared to sham-operated group, the expression of miR-15a in CCI rats was significantly reduced, whereas neuroinflammation in spinal cord tissues was increased. Intrathecal administration of miR-15a agomir significantly attenuated CCI-induced NP and the levels of proinflammatory cytokines, including IL-6, IL-1β, and TNF-α. AKT3 was predicted and confirmed as a miR-15a-regulated gene. We further demonstrated that miR-15a overexpression downregulated the level of AKT3 in primary rat microglia and rat CCI model. Moreover, the upregulation of miR-15a induced the expressions of autophagy-associated proteins, suggesting that the regulation mechanism of miR-15a in NP development involves AKT3mediated autophagy via inhibiting the expression of AKT3. Conclusion Our findings indicated that miR-15a might serve as a promising therapeutic target for the management of NP through the stimulation of autophagic process.

Spatial target path following and coordinated control of multiple UUVs

Qi, Xue,Xiang, Peng,Cai, Zhi-jun The Society of Naval Architects of Korea 2020 International Journal of Naval Architecture and Oc Vol.12 No.1

The coordination control of multiple Underactuated Underwater Vehicles (UUVs) moving in three dimensional space is investigated in this paper. The coordinated path following control task is decomposed into two sub tasks, that is, path following control and coordination control. In the spatial curve path following control task, path following error dynamics is build in the Serret-Frenet coordinate frame. The virtual reference object can be chosen freely on the desired spatial path. Considering the speed of the UUV, the line-of-sight navigation is introduced to help the path following errors quickly converge to zero. In the coordination control sub task, the communication topology of multiple UUVs is described by the graph theory. The speed of each UUV is adjusted to achieve the coordination. The path following system and the coordination control system are viewed as the feedback connection system. Input-to-state stable of the coordinated path following system can be proved by small gain theorem. The simulation experiments can further demonstrate the good performance of the control method.

An Adaptively Speculative Execution Strategy Based on Real-Time Resource Awareness in a Multi-Job Heterogeneous Environment

( Qi Liu ),( Weidong Cai ),( Qiang Liu ),( Jian Shen ),( Zhangjie Fu ),( Xiaodong Liu ),( Nigel Linge ) 한국인터넷정보학회 2017 KSII Transactions on Internet and Information Syst Vol.11 No.2

MapReduce (MRV1), a popular programming model, proposed by Google, has been well used to process large datasets in Hadoop, an open source cloud platform. Its new version MapReduce 2.0 (MRV2) developed along with the emerging of Yarn has achieved obvious improvement over MRV1. However, MRV2 suffers from long finishing time on certain types of jobs. Speculative Execution (SE) has been presented as an approach to the problem above by backing up those delayed jobs from low-performance machines to higher ones. In this paper, an adaptive SE strategy (ASE) is presented in Hadoop-2.6.0. Experiment results have depicted that the ASE duplicates tasks according to real-time resources usage among work nodes in a cloud. In addition, the performance of MRV2 is largely improved using the ASE strategy on job execution time and resource consumption, whether in a multi-job environment.

      • Probe spectrum measurements of Eu3+ ions as a relevant tool for monitoring in vitro hydroxyapatite formation in a new borate biomaterial

        Qi, Shuyun,Huang, Yanlin,Li, Yadong,Cai, Peiqing,Kim, Sun Il,Seo, Hyo Jin The Royal Society of Chemistry 2014 Journal of Materials Chemistry B Vol.2 No.37

<P>CaB2O4 powders and ceramics were prepared by the conventional solid-state reaction. <I>In vitro</I> hydroxyapatite (HA) mineralization was investigated by soaking the samples in simulated body fluid (SBF) for various time periods. X-ray diffraction and structural refinements, scanning electron microscopy and X-ray energy-dispersive spectra measurements were applied to investigate apatite formation before and after immersion in SBF. HA can easily form flower-like nanostructures with nano-needles even when soaked in SBF for several hours. The <I>in vitro</I> bioactivity of CaB2O4 was attributed to easy formation of B-OH groups in the CaB2O4 structure when soaked in SBF solutions. In the process of mineralization, the luminescence evolution of Eu3+ ions, a well-known structural probe, was detected by photoluminescence spectra and photoluminescence decay curves. This suggested that the process of mineralization can be monitored by the luminescence intensity of Eu3+ ions in the mineralization products. The current study will open up a new and simple <I>in vivo</I> avenue for <I>in situ</I> monitoring of hydroxyapatite conversion using a fiber luminescence spectrometer.</P>

      • A Smart Strategy for Speculative Execution Based on Hardware Resource in a Heterogeneous Distributed Environment

        Qi Liu,Weidong Cai,Zhangjie Fu,Jian Shen,Nigel Linge 보안공학연구지원센터 2016 International Journal of Grid and Distributed Comp Vol.9 No.2

MapReduce, as a popular programming model for processing large data sets, has been widely applied. MapReduce 2.0 (MRV2) is a newly adopted one, which has a better performance. Those machines which have a lower performance in a cluster usually play a role who pull down the pace of job execution time. Speculative execution known as an approach for dealing with the above problems works by backing up those tasks running on a low performance machine to a higher one. Although multiple speculative execution strategies have been proposed, there are still a lot of pitfalls existing in the strategies. In this paper, Some pitfalls in proposed strategy have been modified and computer hardware has been taken into consideration (HWC-Speculation). In Hadoop-2.6, we have implemented it, called Hadoop-HWC. Experiment results show that our method can find a slow task correctly, also, the performance of MRV2 is improved.

Effectiveness and safety of acupuncture for angina pectoris: An overview of systematic reviews

        Qi Wen-Chuan,Fu Hong-Juan,Sun Rui-Rui,Li Xiang,Cai Ding-Jun,Wang Chao,Liang Fan-Rong 한국한의학연구원 2022 Integrative Medicine Research Vol.11 No.3

Background: The number of systematic reviews or meta-analyses (SRs/MAs) on the effectiveness of acupuncture for angina pectoris (AP) is increasing. Due to the inconsistent conclusions and unknown quality of these SRs/MAs, this overview aimed to systematically evaluate and synthesize the existing SRs/MAs, attempting to provide more reliable evidence for the effectiveness and safety of acupuncture in the treatment of AP. Methods: SRs/MAs were searched via eight databases from inception to March 14, 2022. The risk of bias was evaluated using the Risk of Bias in Systematic reviews (ROBIS) tool. The quality of the methodology, reporting, and evidence were assessed by the Assess Systematic Reviews 2 (AMSTAR-2), the Preferred Reporting Item for Systematic Review and Meta-analysis for Acupuncture (PRISMA-A), and the Grading of Recommendations Assessment, Development, and Evaluation (GRADE) system, respectively. Results: Sixteen SRs/MAs were included and fifteen SRs/MAs were considered being of critically low quality according to AMSTAR-2. Only three SRs/MAs were rated at low risk of bias. No study reported all the items listed in the PRISMA-A checklist. No high-quality evidence with GRADE assessment was found. With the low-quality evidence, acupuncture combined with other interventions was superior to monotherapy (medications or Chinese medicine) in the angina symptom and electrocardiogram recovery. No adverse effects owing to acupuncture were reported. Conclusions: Owing to the lack of high-quality evidence provided by the current SRs/MAs, the effective-ness of acupuncture for AP still warrants further proof. Further researches with more critical design and methodology are needed for providing more convincing evidence.
