Since polyphenol-rich products such as red wine, grape juice, and grape extracts have been shown to induce potent endothelium-dependent relaxations, we have evaluated whether commercial fruit juices such as those from berries are also able to induce endothelium-dependent relaxations of isolated coronary arteries and, if so, to determine whether this effect is related to their phenolic content. Among the 51 fruit juices tested, 2/12 grape juices, 3/7 blackcurrant juices, 4/5 cranberry juices, 1/6 apple juices, 0/5 orange juices, 2/6 red fruit and berry juices, 3/6 blends of red fruit juices, and 0/4 non-red fruit juices were able to induce relaxations achieving more than 50% at a volume of 1%. The active fruit juices had phenolic contents ranging from 0.31 to 1.86 g GAE/L, which were similar to those of most of the less active juices with the exception of one active grape juice (2.14 g GAE/L) and one active blend of red fruit juices (3.48 g GAE/L). Altogether, these findings indicate that very few commercial fruit juices have the ability to induce potent endothelium-dependent relaxations, and that this effect is not related to their quantitative phenolic content, but rather to their qualitative phenolic composition.
Total Transit Time and Probiotic Persistence in Healthy Adults: A Pilot Study

Background/AimsMotility, stool characteristics, and microbiota composition are expected to modulate probiotics’ passage through the gut but their effects on persistence after intake cessation remain uncharacterized. This pilot, open-label study aims at characterizing probiotic fecal detection parameters (onset, persistence, and duration) and their relationship with whole gut transit time (WGTT). Correlations with fecal microbiota composition are also explored. MethodsThirty healthy adults (30.4 ± 13.3 years) received a probiotic (30 × 109 CFU/capsule/day, 2 weeks; containing Lactobacillus helveticus R0052, Lacticaseibacillus paracasei HA-108, Bifidobacterium breve HA-129, Bifidobacterium longum R0175, and Streptococcus thermophilus HA-110). Probiotic intake was flanked by 4-week washout periods, with 18 stool collections throughout the study. WGTT was measured using 80% recovery of radio-opaque markers. ResultsTested strains were detected in feces ~1-2 days after first intake and persistence after intake cessation was not significantly different for R0052, HA-108, and HA-129 (~3-6 days). We identified 3 WGTT subgroups within this population (named Fast, Intermediate, and Slow), which could be classified by machine learning with high accuracy based on differentially abundant taxa. On average, R0175 persisted significantly longer in the intermediate WGTT subgroup (~8.5 days), which was mainly due to 6 of the 13 Intermediate participants for whom R0175 persisted ≥ 15 days. Machine learning classified these 13 participants according to their WGTT cluster (≥ 15 days or < 5 days) with high accuracy, highlighting differentially abundant taxa potentially associated with R0175 persistence. ConclusionThese results support the notion that host-specific parameters such as WGTT and microbiota composition should be considered when designing studies involving probiotics, especially for the optimization of washout duration in crossover studies but also for the definition of enrollment criteria or supplementation regimen in specific populations.

<P>We create a baseline of the black hole (BH) mass (M-BH)-stellar-velocity dispersion (sigma) relation for active galaxies, using a sample of 66 local (0.02 < z < 0.09) Seyfert-1 galaxies, selected from the Sloan Digital Sky Survey (SDSS). Analysis of SDSS images yields AGN luminosities free of host-galaxy contamination, and morphological classification. 51/66 galaxies have spiral morphology. Out of these, 28 bulges have Sersic index n < 2 and are considered candidate pseudo-bulges, with eight being definite pseudo-bulges based on multiple classification criteria met. Only 4/66 galaxies show signs of interaction/merging. High signal-to-noise ratio Keck spectra provide the width of the broad H beta emission line free of Fe II emission and stellar absorption. AGN luminosity and H beta line widths are used to estimate M-BH. The Keck-based spatially resolved kinematics is used to determine stellar-velocity dispersion within the spheroid effective radius (sigma(spat,reff)). We find that sigma can vary on average by up to 40% across definitions commonly used in the literature, emphasizing the importance of using self-consistent definitions in comparisons and evolutionary studies. The M-BH-sigma relation for our Seyfert-1 galaxy sample has the same intercept and scatter as that of reverberation-mapped AGNs as well as that of quiescent galaxies, consistent with the hypothesis that our single epoch M-BH estimator and sample selection function do not introduce significant biases. Barred galaxies, merging galaxies, and those hosting pseudo-bulges do not represent outliers in the M-BH-sigma relation. This is in contrast with previous work, although no firm conclusion can be drawn on this matter due to the small sample size and limited resolution of the SDSS images.</P>

Chronic liver diseases with portal hypertension are characterized by a progressive vasodilatation, endothelial dysfunction, and NADPH oxidase-derived vascular oxidative stress, which have been suggested to involve the angiotensin system. This study evaluated the possibility that oral intake of polyphenol-rich blackcurrant juice (PRBJ), a rich natural source of antioxidants, prevents endothelial dysfunction in a rat model of cirrhosis induced by chronic bile duct ligation (CBDL), and, if so, determined the underlying mechanism. Male Wistar rats received either control drinking water or water containing 60 mg/kg gallic acid equivalents of PRBJ for 3 weeks before undergoing surgery with CBDL or sham surgery. After 4 weeks, vascular reactivity was assessed in mesenteric artery rings using organ chambers. Both the acetylcholine-induced nitric oxide (NO)- and endothelium-dependent hyperpolarization (EDH)-mediated relaxations in mesenteric artery rings were significantly reduced in CBDL rats compared to sham rats. An increased level of oxidative stress and expression of NADPH oxidase subunits, COX-2, NOS, and of the vascular angiotensin system are observed in arterial sections in the CBDL group. Chronic intake of PRBJ prevented the CBDL-induced impaired EDH-mediated relaxation, oxidative stress, and expression of the different target proteins in the arterial wall. In addition, PRBJ prevented the CBDL-induced increase in the plasma level of proinflammatory cytokines (interleukin [IL]-1α, monocyte chemotactic protein 1, and tumor necrosis factor α) and the decrease of the anti-inflammatory cytokine, IL-4. Altogether, these observations indicate that regular ingestion of PRBJ prevents the CBDL-induced endothelial dysfunction in the mesenteric artery most likely by normalizing the level of vascular oxidative stress and the angiotensin system.
H0LiCOW VII: cosmic evolution of the correlation between black hole mass and host galaxy luminosity

BROAD H<i>β</i>EMISSION-LINE VARIABILITY IN A SAMPLE OF 102 LOCAL ACTIVE GALAXIES

<P>A sample of 102 local (0.02 <= z <= 0.1) Seyfert galaxies with black hole masses M-BH > 10(7)M(circle dot) was selected from the Sloan Digital Sky Survey (SDSS) and observed using the Keck 10 m telescope to study the scaling relations between M-BH and host galaxy properties. We study profile changes of the broad H beta emission line within the three to nine year time frame between the two sets of spectra. The variability of the broad H beta emission line is of particular interest, not only because it is used to estimate M-BH, but also because its strength and width are used to classify Seyfert galaxies into different types. At least some form of broad-line variability (in either width or flux) is observed in the majority (similar to 66%) of the objects, resulting in a Seyfert-type change for similar to 38% of the objects, likely driven by variable accretion and/or obscuration. The broad H beta line virtually disappears in 3/102 (similar to 3%) extreme cases. We discuss potential causes for these changing look active galactic nuclei. While similar dramatic transitions have previously been reported in the literature, either on a case-by-case basis or in larger samples focusing on quasars at higher redshifts, our study provides statistical information on the frequency of H beta line variability in a sample of low-redshift Seyfert galaxies.</P>
