      • Plasma Vascular Endothelial Growth Factors A and C in Patients undergoing Prostatic Biopsy and TURP for Suspected Prostatic Neoplasia

        Background: Formation of new blood vessels is necessary for the development and spread of neoplasms more than 1 mm3 in volume, angiogenesis being responsible for formation of new from pre-existing blood vessels. Vascular endothelial growth factor (VEGF) is pivotal and the best studied angiogenic factor in all human cancers. Therefore we designed this study to investigate the role of VEGF-A and VEGF-C in prostate cancer in comparison with BPH controls in a north Indian population. Methods: In this case-control study a total of 100 subjects were included on the basis of confirmed histopathological reports, out of which 50 were prostate cancer patients and the other 50 were BPH patients with PSA levels >2 ng/ml and abnormal digital rectal examination (DRE) findings during September 2009 to August 2011 from the Department of Urology, KGMU, Lucknow, India. Plasma levels of VEGF were determined using quantitative immunoassay (ELISA-enzyme linked immunosorbent assay). Statistical analysis was carried out using SPSS 15.0 version. Results: The mean age of prostate cancer ($67.6{\pm}5.72$) patients was significantly higher (p=0.005) than BPH ($63.6{\pm}7.92$) patients. Expression of VEGF-A was not significantly higher in disease stage C1 than D1 or D2 and A or B (p=0.13) while the level of VEGF-A was significantly higher (p=0.04) in prostate cancer as compared to BPH subjects (PCa=13.0 pg/ml, BPH=6.8 pg/ml). Levels of VEGF-C were similar in both groups (PCa=832.6 pg/ml, BPH=823.7 pg/ml). In ROC curve, the area under curve (AUC) was 0.70 (95%CI: 0.60-0.80) and the cut-off value for which a higher proportion of patients was correctly classified (20%) was 26.0 pg/mL. Conclusion: Although VEGF-A is increased in cancer prostate patients a statistically significant correlation could not be established in this study. VEGF-C was not found to be a useful biomarker.

        Monocrotophos Induces the Expression of Xenobiotic Metabolizing Cytochrome P450s (CYP2C8 and CYP3A4) and Neurotoxicity in Human Brain Cells

        <P>Expression of various cytochrome P450s (CYPs) in mammalian brain cells is well documented. However, such studies are hampered in neural/glial cells of human origin due to nonavailability of human brain cells. To address this issue, we investigated the expression and inducibility of CYP2C8 and CYP3A4 and their responsiveness against cyclophosphamide (CPA) and organophosphorus pesticide monocrotophos (MCP), a known developmental neurotoxicant in human neural (SH-SY5Y) and glial (U373-MG) cell lines. CPA induced significant expression of CYP2C8 and CYP3A4 in both types of cells in a time-dependent manner. Neural cell line exhibited relatively higher constitutive and inducible expression of CYPs than the glial cell line. MCP exposure alone could not induce the significant expression of CYPs, whereas the cells preexposed to CPA showed a significant response to MCP. Similar to the case of CPA induced expressions, neural cells were found to be more vulnerable than glial cells. Our data indicate differential expressions of CYPs in cultured human neural and glial cell lines. The findings were synchronized with protein ligand docking studies, which showed a significant modulatory capacity of MCP by strong interaction with CYP regulators-CAR and PXR. Similarly, the known CYP inducer CPA has also shown significant high docking scores with the two studied CYP regulators. We also observed a significant induction in reactive oxygen species (ROS), lipid peroxides (LPO), micronucleus (MN), chromosomal aberration (CA), and reduction in reduced glutathione (GSH) and catalase following the exposure of MCP. Moreover, the expressions of apoptotic markers such as caspase-3, caspase-9, Bax, and p53 were significantly upregulated, whereas the levels of antiapoptotic marker, Bcl2, was downregulated after the exposure of MCP in both cell lines. These findings confirm the involvement of ROS-mediated oxidative stress, which subsequently triggers apoptosis pathways in both human neural (SH-SY5Y) and glial (U373-MG) cell lines following the exposure of MCP.</P>

        Effect of selected pesticides on larval mortality of the neuropteran predator, Chrysopa lacciperda Kimmins, of the lac insect, Kerria lacca (Kerr)

        Laboratory bioassays were carried out to evaluate the bioefficacy of some pesticides against larval Chrysopa lacciperda Kimmins, a lac insect predator, to develop a suitable strategy for field management of this serious neuropteran pest of Indian lac insect, Kerria lacca (Kerr). Seven insecticides (lambdacyhalothrin, carbosulfan,spinosad, indoxacarb, fipronil, alphamethrin and ethofenprox) were identified based on field trials against the lac insect. They were evaluated for their bioefficacy against C. lacciperda by spraying the insect directly and by exposing the insect to a residual film. Direct spray of lambdacyhalothrin (0.005 and 0.008% a.i.), carbosulfan (0.02 and 0.03% a.i.), fipronil (0.005 and 0.01% a.i.), alphamethrin (0.005 and 0.01% a.i.), spinosad (0.02% a.i.),indoxacarb (0.02% a.i.), and ethofenprox (0.02% a.i.) exhibited 100% mortality of C. lacciperda within 24 h of treatment. Fipronil (0.005 and 0.01% a.i.) and indoxacarb (0.02% a.i.) were equally effective as 100% mortality was observed within 24 h of treatment with both modes of treatment. For most insecticides, direct spray was more effective compared to residual films. It is therefore, suggested that lambdacyhalothrin, carbosulfan,indoxacarb, spinosad, fipronil, alphamethrin and ethofenprox shall be incorporated in IPM programs for the effective management of this neuropteran predator of lac insect without adversely affecting the lac insect.

        Estimation of Rumen Microbial Protein Supply Using Urinary Purine Derivatives Excretion in Crossbred Calves Fed at Different Levels of Feed Intake

        A study was carried out to study the response of total purine derivatives (PD) excretion in urine to determine microbial N (MN) supply at four fixed levels of feed intake (namely 95, 80, 60 and 40% of voluntary intake). The crossbred (CB) calves were allocated according to a $4{\times}4$ Latin Square Design and fed wheat straw and concentrate (1:1). The rate of PD excretion (mmol/d) as a linear function of feed intake was 15.85/kg DMI and 20.12/kg DOMI. Based on the endogenous and PD excretion rates obtained in this study, a relationship between daily urinary PD excretion (Y, mmol) and daily microbial protein supply (X, mmol) was developed for crossbred calves as Y = 0.83X+0.296 kg $W^{0.75}$. The derived microbial N values using this equation differed (p<0.001) among the 4 groups and was the highest in L-95 followed by L-80, L-60 and L-40. The relationship between urinary nitrogen loss (Y, g/d) and DOMI (X, kg/d) was established as: Y = 6.038X+21.753 ($r^2$ = 0.663, p<0.01). When urinary excretion of PD (Y, mmol/d) was plotted against intake of DM and DOM (X, kg/d), the equations obtained were: Y = 7.1711X+8.674 ($r^2$ = 0.889, p<0.01) and Y = 12.434X+7.683 ($r^2$ = 0.896, p<0.01), respectively. The proportional contribution of allantoin and uric acid to total PD remained stable irrespective of level of feed intake. Similarly, urinary excretion of creatinine did not differ (p>0.05) between animals fed at different levels. The MN supply was the highest to animals at intake levels L-95, and decreased linearly with corresponding decrease in feed intake. However, the MN supply when expressed per kg DOMI remained statistically (p>0.05) similar irrespective of level of intake. The results revealed that the excretion of urinary purine derivatives were positively correlated with the level of feed intake as well as rumen microbial supply and thus it could be a good indicator for measuring the microbial protein supply and nutritional status of animals.

        Measurement of Microbial Protein Supply in Murrah Buffaloes (Bubalus bubalis) Using Urinary Purine Derivatives Excretion and PDC Index

        A study was conducted to predict the rumen microbial protein production based on urinary excretion of purine derivatives in buffaloes fed a diet of wheat straw and concentrate (40:60) at four fixed levels of feed intake. (95, 80, 60 and 40% of preliminary voluntary feed intake) following experimental protocol of IAEA (Phase I). The buffaloes were allocated according to a $4{\times}4$ latin square design. The urinary allantoin, uric acid, total PD excretion (mmol/d) in treatments L-95, L-80, L-60 and L-40 was 20.13, 16.00, 12.96 and 9.17; 1.88, 2.12, 2.11 and 2.15; 22.01, 18.12, 15.07 and 11.32, respectively and were significantly (p<0.05) different among treatments except for uric acid. The rate of PD excretion (mmol/d) was positively correlated with the digestible organic matter intake. Variations were observed in PD and creatinine concentration in spot samples collected at 6-hour interval. However, daily PD:Creatinine ratio (PDC index) appears to be a reasonably good predictor of microbial-N supply. The contribution of basal purine excretion to total excretion of purine derivatives (PD) was determined in pre-fasting period followed by a fasting period of 6 d (Phase II). Daily PD and creatinine excretion (mmol/kg $W^{0.75}$) during fasting averaged 0.117 and 0.456 respectively for buffaloes. The excretion rates of PD decreased significantly (p<0.01) during fasting compare to pre-fasting period, the urinary creatinine excretion remained almost similar. Except for creatinine, plasma concentration of target parameters significantly (p<0.01) declined during fasting. Likewise, glomerular filtration rate (GFR) and renal clearance of allantoin and uric acid also decreased. Based on the PD excretion rates during fasting and at different levels of feed intake obtained in this study, a relationship between daily urinary PD excretion (Y-mmol) and microbial purine absorption (X-mmol) was developed for buffaloes as Y = 0.74X+0.117 kg $W^{0.75}$. The microbial N supply (g/kg DOMI) remained statistically similar irrespective of dietary treatment. The results showed that excretion of urinary purine derivatives is positively correlated with the levels of feed intake in Murrah buffaloes and thus, estimation of urinary purine derivatives and PDC index could be used to determine microbial nitrogen supply when there is large variation in level of feed intake.

        Antioxidants Supplementation on Acid Base Balance during Heat Stress in Goats

        The effects of vitamin C and vitamin E with selenium on acid-base balance and some stress hormones were evaluated during heat stress in goats. Goats, 1.5 years of age, were divided into control, heat stress and antioxidant treatment groups 1, 2 and 3. Except for the control, all groups were exposed to a temperature of $40{\pm}2^{\circ}C$ with a relative humidity of 30% for 5 h/d for 21 days in a psychrometric chamber. Rectal temperature and respiratory rates were recorded daily post exposure. Blood samples were collected on every 3rd day for estimation of plasma vitamins C and E, total antioxidant activity and hormones, and separate blood samples were taken to estimate acid-base status. The rectal temperature and respiratory rates were increased (p<0.05) in the heat stress group only. Except for pH and $pO_2$, which were increased significantly (p<0.05) other parameters of acid-base balance such as $pCO_2$, $HCO_3^-$, $TCO_2$, BEb, BEcef, PCV and Hb were significantly decreased (p<0.05) in the heat stress group. An improvement in acid-base status was noted in the antioxidant supplemented groups. Prolactin and cortisol levels were significantly (p<0.05) higher and free T3 and T4 levels were significantly (p<0.05) lower in the heat stress group. Levels of prolactin and cortisol were decreased and free T3 and T4 were increased in antioxidant treatment groups. Different levels of antioxidant supplementation resulted in similar protection against heat stress.

        Performance of Crossbred Cattle Fed Chopped Green Sugarcane Tops and Supplemented with Wheat Bran or Lentil Chuni Concentrates

        Fifteen crossbred cattle bulls of about 22-24 months age (mean body weight ranged from 291.0 to 298.1 kg) were randomly divided into 3 groups of 5 animals following randomized block design. Animals in group 1 were fed concentrate mixture containing wheat bran 97%, mineral mixture 2% and salt 1% plus ad libitum chopped green sugarcane tops (SCT) as a roughage source. Animals in group 2 were fed concentrate mixture containing lentil chuni 97%, mineral mixture 2% and salt 1% while group 3 concentrate contained wheat bran 48.5%, lentil chuni 48.5%, mineral mixture 2% and salt 1% plus ad libitum chopped green SCT to meet their maintenance requirements. During metabolism trial period, the mean total intake and digestibility of DM in groups 1, 2 and 3 was $8.09{\pm}0.41$, $8.41{\pm}0.49$ and $7.86{\pm}0.16kg/d$ and 60.24, 63.24 and 65.05%, respectively. The intake of CP (p<0.05) and EE (p<0.01) was significantly higher in group 2 and group 1, respectively as compared to remaining 2 groups, which were comparable. Digestibility of CP and EE was significantly (p<0.01) higher in group 3 and that of CP was comparable with group 2. The intake and digestibility of total carbohydrates, NDF, ADF, Hemi-cellulose and cellulose were similar among 3 groups. The mean body weight changes in group 1, 2 and 3 were 173.4, 253.4 and 203.4 g/d, respectively, which was significantly (p<0.05) higher in group 2. The total nitrogen (N) and phosphorus (P) intakes were significantly higher in group 2 and group 1, respectively. The total volatile fatty acids, ammonia-N, trichloro-acetic acid precipitable nitrogen (TCA ppt.-N; p<0.05), total-N concentrations and pH were significantly (p<0.01) higher in group 3, however, pH and TCA ppt.-N was comparable with group 2 and 1, respectively. From the results, it may be deduced that the feeding of chopped green SCT supplemented with a concentrate mixture having both wheat bran (48.5%) and lentil chuni (48.5%) has shown a better performance with respect to intake and digestibility of nutrients and growth as well.

        Effect of Sources of Supplementary Protein on Intake, Digestion and Efficiency of Energy Utilization in Buffaloes Fed Wheat Straw Based Diets

        Sixteen adult male buffaloes (average body weight $443{\pm}14kg$) were equally distributed into four groups in an experiment to study the effect of supplementary protein sources on energy utilization efficiency in buffaloes fed a wheat straw-based diet. The animals in the control group were offered a basal diet composed of 700 g deoiled ground nut cake and ad libitum wheat straw. Animals of other groups were offered 1.8 kg of soyabean meal (SBM), linseed meal (LSM) or mustard cake (MC) along with the basal diet. Protein supplementation increased the digestibility of DM (p<0.01), OM (p<0.01) CP (p<0.01) and CF (p<0.05). Maximum CP digestibility was observed on SBM, followed by LSM and MC when compared to the control. Total DMI and DOMI was significantly (p<0.01) higher in protein supplemented groups with no differences between treatment groups. Digestible crude protein (DCP) intake and N balance were significantly (p<0.01) different between the groups; maximum response was obtained with SBM supplementation, followed by LSM and MC. Faecal energy was significantly (p<0.01) lower in SBM and LSM groups in comparison to other groups. Methane production (% DEI) was significantly (p<0.05) lower on the SBM treatment. Metabolizable energy (ME) intake increased significantly due to protein supplementation. Metabolizable energy intake (MEI) of animals in the MC group was less than LSM and SBM. Energy balance was increased significantly (p<0.01) due to protein supplementation and within supplement variation was also significant with maximum balance in SBM followed by LSM and MC groups. Protein supplementation significantly (p<0.05) increased the digestibility and metabolizability of energy from whole ration. Metabolizable energy (ME) content (Mcal/kg DM) of SBM, LSM and MC was 4.49, 3.56 and 2.56, respectively. It was concluded that protein supplementation of wheat straw increased intake, digestibility and metabolizability of energy and maximum response could be obtained when soybean meal was used as a supplement.

        <P>Panax ginseng is an important cash crop in the Asian countries due to its pharmaceutical effects, however the plant is exposed to various abiotic stresses, lead to reduction of its quality. One of them is the Aluminum (Al) accumulation. Plant growth promoting bacteria which able to tolerate heavy metals has been considered as a new trend for supporting the growth of many crops in heavy metal occupied areas. In this study, twelve bacteria strains were isolated from rhizosphere of diseased Korean ginseng roots located in Gochang province, Republic of Korea and tested for their ability to grow in Al-embedded broth media. Out of them, four strains (Pseudomonas simiae N3, Pseudomonas fragi N8, Chryseobacterium polytrichastri N10, and Burkholderia ginsengiterrae N11-2) were able to grow. The strains could also show other plant growth promoting activities e.g. auxins and siderophores production and phosphate solubilization. P. simiae N3, C. polytrichastri N10, and B. ginsengiterrae N11-2 strains were able to support the growth of Arabidopsis thaliana stressed by Al while P. fragi N8 could not. Plants inoculated with P. simiae N3, C. polytrichastri N10, and B. ginsengiterrae N11-2 showed higher expression level of Al-stress related genes, AtAIP, AtALS3 and AtALMT1, compared to non-bacterized plants. Expression profiles of the genes reveal the induction of external mechanism of Al resistance by P. simiae N3 and B. ginsengiterrae N11-2 and internal mechanism by C. polytrichastri N10. Korean ginseng seedlings treated with these strains showed higher biomass, particularly the foliar part, higher chlorophyll content than non-bacterized Al-stressed seedlings. According to the present results, these strains can be used in the future for the cultivation of ginseng in Al persisted locations.</P>

        A comparison of electrochromic properties of sol-gel derived amorphous and nanocrystalline tungsten oxide films

        A pristine acetylated peroxotungstate sol with and without 4 wt% of oxalic acid dihydrate (OAD) yielded nanocrystalline and amorphous tungsten oxide (WO3) films respectively by dip coating technique. Contrary to the expected trend, whereby, the nanostructured 4% OAD film with a triclinic modification should have shown superior electrochromic efficiency, its amorphous 0% OAD counterpart exhibits higher optical modulation and coloration efficiency at photopic wavelengths. This anomalous behavior of the amorphous 0% OAD film was correlated to a higher W5+ content in its colored state. The colored nanocrystalline 4% OAD film contained a lower proportion of the W5+ color centers. Under the same level of lithiation, while the 4% OAD film was also constituted by W4+ states, its 0% OAD counterpart did not contain any 4+ states of tungsten. X-ray photoelectron spectroscopic (XPS) investigations also confirmed that the single peak at 1.4 eV in the absorption coefficient–wavelength spectrum of the colored 0% OAD film arises from the small polaronic transitions between the W6+ and W5+ states of tungsten whereas the W6+–W5+ and the W5+–W4+ charge transitions produce two distinct peaks at 1.2 and 1.6 eV in the a–k spectrum of the colored 4% OAD film. The microstructure of the 4% OAD film, characterized by an interconnected network of nanocrystallites and pores promotes rapid ion insertion and extraction. Therefore, the larger magnitudes of NIR reflectance modulation, diffusion coefficient for lithium, electrochemical activity and faster color–bleach kinetics observed for the 4% OAD film, are a direct consequence of its structure. Band gap widening upon lithium insertion observed for both films, is a repercussion of Burstein–Moss effect and structural changes that occur upon coloration.
