        Extracts of Opuntia humifusa Fruits Inhibit the Growth of AGS Human Gastric Adenocarcinoma Cells.

        Opuntia humifusa (OHF) has been used as a nutraceutical source for the prevention of chronic diseases. In the present study, the inhibitory effects of ethyl acetate extracts of OHF on the proliferation of AGS human gastric cancer cells and the mode of action were investigated. To elucidate the antiproliferative mechanisms of OHF in cancer cells, the expression of genes related to apoptosis and cell cycle arrest were determined with real-time PCR and western blot. The cytotoxic effect of OHF on AGS cells was observed in a dose-dependent manner. Exposure to OHF (100 μg/mL) significantly induced (P<0.05) the G1 phase cell cycle arrest. Additionally, the apoptotic cell population was greater (P<0.05) in OHF (200 μg/mL) treated AGS cells when compared to the control. The expression of genes associated with cell cycle progression (Cdk4, Cdk2, and cyclin E) was significantly downregulated (P<0.05) by the OHF treatment. Moreover, the expression of Bax and caspase-3 in OHF treated cells was higher (P<0.05) than in the control. These findings suggest that OHF induces the G1 phase cell cycle arrest and activation of mitochondria-mediated apoptosis pathway in AGS human gastric cancer cells.

        Anticancer Properties of Extracts from Opuntia humifusa Against Human Cervical Carcinoma Cells

        In this study, we found that the total polyphenol and ascorbic acid levels in the fruit of Opuntia humifusa are higher than those in other parts of the plant. We further hypothesized that antioxidants in O. humifusa might affect the growth or survival of cancer cells. Hexane extracts of seeds and ethyl acetate extracts of fruits and stems significantly suppressed the proliferation of HeLa cervical carcinoma cells, but did not affect the proliferation of normal human BJ fibroblasts. Additionally, the extracts of O. humifusa induced G1 phase arrest in HeLa cells. The O. humifusa extracts reduced the levels of G1 phase-associated cyclin D1, cyclin-dependent kinase 4 (Cdk4), and phosphorylated retinoblastoma proteins. Moreover, p21WAF1/Cip1 and p53 expression significantly increased after treatment. We examined the effects of ethyl acetate extracts of O. humifusa fruit (OHF) on HeLa cells xenograft tumor growth. OHF treatment significantly reduced tumor volume and this decrease was correlated with decreased Cdk4 and cyclin D1 expression. Furthermore, flavonoids, trans Taxifolin, and dihydrokaempferol, were isolated from OHF. Thus, this extract may be a promising candidate for treating human cervical carcinoma.

        Effects of Opuntia humifusa Seed Powder on Serum Lipid Profile in Ovariectomized Rats

        Opuntia humifusa contains high levels of antioxidants including vitamin C, flavonoids and polyphenols, which may provide beneficial effects such as hypolipidemic activity and the reduction of atherosclerosis in postmenopausal women. This study was conducted to determine if the intake of O. humifusa seeds powder (OHS) regulates lipid concentrations, glutamate-oxaloacetate transaminase (GOT), and glutamate-pyruvate transaminase (GPT) in the serum of ovariectomized rats. Sprague-Dawley female rats were randomly assigned to either a sham-operated group (Sham) or one of the following four ovariectomy (OVX) subgroups: OVX with vehicle (OVX), OVX with 100, 200, and 500 mg/kg/day OHS (OHS100, OHS200, OHS500). Daily oral administration of OHS was initiated one week after ovariectomy and continued for seven weeks. Upon completion of treatments, organs were weighed and GOT, GPT, total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) and high-density lipoprotein cholesterol (HDL-C) levels were determined enzymatically. No significant differences in feed intake and organ index were observed among the groups. Significant decreases in GPT, TC and LDL-C (p<0.05) were observed in all of the OHS groups (OHS100, 200 and 500), while no significant changes in HDL-C were observed. In addition, the OHS200 and OHS500 treatment groups exhibited a lower level of serum GOT compared to the OVX group. These results indicate that supplementation with O. humifusa seeds could induce favorable changes in serum lipoprotein and lipid profiles, which frequently worsen with inadequate estrogen availability.

        Effects of Cheonnyuncho (Opuntia humifusa) Seeds Treatment on the Mass, Quality, and the Turnover of Bone in Ovariectomized Rats

        Cheonnyuncho (Opuntia humifusa), rich in polyphenols and flavonoids, is known to have medicinal benefits in the treatment of arteriosclerosis and inflammation. This study was undertaken to evaluate the effects of cheonnyuncho seed powder (OHS) on postmenopausal osteoporosis in ovariectomized rats. OHS treatment prevented an ovariectomy (OVX)-induced decrease in biomechanical quality of femurs. OHS also dose-dependently inhibited bone mineral density (BMD) decreases in femurs, which were accompanied by a significant decrease in skeletal remodeling, as evidenced by decreased levels of the bone turnover markers osteocalcin, alkaline phosphatase (ALP)activity, and serum and urinary Ca excretion. Micro-CT analysis of femoral metaphysics showed that OHS prevented the OVX-induced decrease in bone volume/tissue volume,trabecular thickness, and trabecular number. Concluded,the treatment with OHS improves bone biomechanical quality through modification of BMD and trabecular microarchitecture without hyperplastic effects on the uterus. For these reasons, OHS may be suitable as an alternative treatment of postmenopausal osteoporosis.

        식품산업 현황과 R&D 미래 대응전략: 공공부문의 역할과 추진방향을 중심으로

        식품산업은 국민 삶과 국가 경제에서 매우 중요한 역할을 하며 꾸준한 성장세를 유지하고 있다. 향후에도 식품은 필수재라는 상품 특성상 경기침체 영향이 작고, 식생활 및 소비성향 변화와 함께 시장규모가 지속적으로 확대될 전망이다. 이에 전반적인 국내 식품산업 현황과 주요 이슈를 살펴보고, 연구개발 측면에서 도출된 문제점을 분석하여 향후 대응방안을 모색하고자 한다. 특별히 식품산업의 활력 제고 및 중소기업 경쟁력 강화를 위해 공공부문에서 중점적으로 수행해야 할 역할과 연구개발 추진방향을 제시하고자 한다. 식품산업이 새로운 국가성장 동력으로서 지속적으로 발전하기 위해서 정부는 거시적 방향성 조율을 통해 일관성 있는 R&D 로드맵을 제공하고, 공공 연구기관 및 민간 기업은 연구개발 현장에서 체감 가능한 실질적이며 구체적인 협업 방안을 마련할 필요가 있다. 또한 공공부문의 식품 R&D는 기초원천연구와 공공 플랫폼기술 중심으로 핵심 요소기술의 전략적 중요도 및 글로벌 기술 수준차이를 고려하여 투자 방향을 설정할 필요가 있다. 마지막으로 식품 연구를 통한 국가·사회적 문제 해결 또는 식품분야 난제 해결을 위한 융합연구 등 식품기술 중심의 아젠더를 발굴하고 대형 연구개발사업으로 연계하는 노력이 보다 절실하게 필요하다. Current status of the domestic food industry and major issues are reviewed, and some problems derived from the R&D aspect are analyzed. Particularly, the role of the public sector and the direction of R&D are presented in order to enhance the vitality of the food industry and strengthen the competitiveness of small and mediumsized businesses. At first, the government needs to provide a consistent R&D roadmap through macroscopic coordination, and public institutes and private companies should come up with practical and concrete collaborative measures. It is also necessary to set the investment direction for food R&D in the public sector, taking into account the strategic importance of core technology and the global level difference, targeting on basic research and public platform technology. More efforts to discover agendas focused on food technology and link them to large-scale R&D projects are urgently needed to solve national and social problems through food research.

        천년초선인장 열매추출물의 폴리페놀, 플라보노이드 함량과 유방암 세포(MCF-7)에 대한 성장 억제효과

        본 연구에서는 천년초선인장 열매의 총 폴리페놀과 플라보노이드 함량을 분석하고, 열매의 추출물이 인간의 유방 상피조직에서 유래한 암세포인 MCF-7의 증식과 세포사멸에 미치는 영향을 조사하였다. 천년초선인장 열매의 총 폴리페놀 함량은 약 4.49 g/100 g, 플라보노이드 함량은 약 1.31 g/100 g로 나타났다. 열매의 물 추출물 100과 500 ㎍/mL 농도에서 정상세포인 BJ에 대한 세포독성을 나타내지 않으면서 MCF-7 세포의 살아있는 세포수를 농도 의존적으로 감소시키는 효과를 보였다. 또한 세포배양액에 열매의 물 추출물을 첨가한 경우, G1 arrest가 일어나 세포주기 진행이 지연되었으나, apoptosis에는 영향을 주지 않음이 확인되어 천년초선인장 열매 물 추출물이 apoptosis보다는 G1 arrest를 유도하여 유방암세포를 억제한다는 것을 규명하였다. 이러한 결과들은 천년초선인장 열매의 물 추출물이 세포독성에는 영향을 주지 않으면서 유방암세포의 증식과 세포주기 진행을 억제하는 효과적인 항암물질이 될 수 있는 기능성 소재로 활용 가능하다는 것을 암시한다. Opuntia humifusa, widely distributed in the southern regions of the Korean peninsula, is known to have bioactive functions and medicinal benefits for treating various diseases such as arteriosclerosis, diabetes mellitus, gastritis, and hyperglycemia. In this study total polyphenol and flavonoid contents of fruit and its anticarcinogenic effects on human breast cancer were investigated. As expected, O. humifusa showed high concentrations of total polyphenol as well as flavonoid as compared to other kinds of cactus. Effects of the water extracts of O. humifusa on the proliferation, G1 arrest and apoptosis of the MCF-7 human breast cancer cells were also examined using the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assays, and G1 cycle arrest and apoptotic effect of O. humifusa were analyzed by flow cytometry. When MCF-7 cells were treated with different concentrations of hexane, ethyl acetate and water extracts of O. humifusa, water extracts of the fruit significantly decreased viable cell numbers in a concentration dependent manner. A G1 arrest in MCF-7 cells was induced as well. The overall results indicate that water extracts of fruit of O. humifusa would inhibit MCF-7 human breast cancer cell proliferation and induce G1 arrest.

        천년초선인장의 부위별 영양성분 분석과 정상 및 암세포에 미치는 효과

        In order to examine the nutritional value of prickly pear(Opuntia humifusa), contents of ash, protein, fat, minerals and vitamins were determined on freeze-dried stem, fruit, seed and root from plants harvested in autumn. The average moisture contents for stem, fruit, seed and root were 67~87%. Crude ash content determined on dry weight basis was 2~3%. Crude protein existed mostly in seed(2.95%) and root(2.37%). Crude fat was detected mainly in seed(4.49%). Contents of major minerals(㎎/100 ㎎ dry weight) was generally higher in stem. Ca in stem(4,142.30) and fruit(2,790.86) were much higher than in seed(43.37). P in stem, seed and fruit were 448.19, 263.20 and 161.59, respectively. Stem also displayed more abundant Mg(1,110.86), Zn(35.62) and Mn(37.07). However, fruit contained higher amounts of Fe(13.38) and Se(0.15). Vitamin A was negligible in all plant parts. Vitamin E contents in fruit and stem were 1.78 ㎎ and 1.22 ㎎/㎎ dry weight, respectively. Vitamin C was detected mostly in fruit(445.40) and stem(260.94). Use of a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide-based microtiter assay of cell viability demonstrated an anti-proliferative effect of O. humifusa extract on the MCF-7 estrogen-dependent human breast cancer cell line.
