        성공적 노화를 위한 노인 몸 디자인의 개념과 필요성 연구

        The aging of the population is appearing worldwide, and Korea is increasing the aging population faster than any other country. This means that your life as an elderly person will be longer. In order to live happily as an aged person, it is necessary to achieve successful aging. In successful aging, a healthy body is an important factor that determines the life of the elderly. A healthy body is an ecological body, which denotes that one’s mind and body are in a state of harmony. Straightening the elderly body and creating an ecological body by re-bodying the disordered body order is realized through movement. The process of organizing and internalizing these movements is necessary, and in this thesis this process was set as the body design process. This study is a study on the process of body design of the elderly for successful aging and proposes the direction of the following step-by-step design process: First, the process of designing an old person’s body starts with discovering problems while searching his or her body through movement, which necessarily leads to reflection of one’s life. Second, it is important to plan movements from an ecological point of view and perform them continuously and repeatedly to restore body order. Third, from an ecological point of view, it is a stage of practice that regenerates the harmony of consciousness and body. This is a very important process since it involves the continuous practice of movement to regenerate energy by properly re-arranging the body and smoothing circulation. Designing the elderly body in this way is directly related to health, and recovery of health is the first step in successful aging. New perspectives and changes in one’s view point on the elderly body will be an important driving force in re-examining the lives of all of us and fulfilling a healthy old life as an important member of the community.

        노인을 위한 스토리텔링 기반 예술통합교육의 인문학적 탐색

        This article examines the values of the humanistic approach to the Arts Integration program for the elderly based on Storytelling. In this age that acknowledges the life span of 100 years, ‘becoming elderly successfully’ and ‘being elderly creatively’ are key words for addressing many problems of the quality of life. ‘Becoming elderly successfully’ can be realized through physical, cognitive and emotional health as well as active social involvement. ‘Being elderly creatively’ can be achieved through practicing self-initiative and self-organization. The characteristics of the Arts Integration based Storytelling are ‘keeping existing’ and ‘peculiarity’ that make an individual more confident. At the same time, ‘meaningfulness’ and ‘communicativeness’ that can make one reorganize his or her life are important qualities in the Arts Integration based on Storytelling. The Arts Integration program for the elderly based on Storytelling pursues the artistic lives in which various artistic experiences meet lives with personal stories. In this process, the artistic experiences lead lives to changes and make individuals more able to redesign their lives. Such design of the lives for the elderly has significances as follows. First, it makes one’s existence being present through the narrative of life. Second, it makes one achieve life with self-initiative. Third, it makes one find his or her own ways to express peculiarity. Fourth, it realizes creative aging in which life and arts are combined. The Arts Integration program for the elderly based on Storytelling can offer a new solution for the physical, psychological, sociological problems of the elderly. With its new point of view and various artistic experiences for the problems of the elderly, the Arts Integration program for the elderly based on Storytelling can make the elderly reborn as existences with newly developed aesthetic sensibility.

        괭생이모자반 추출물의 기능성 원료 표준화를 위한 Fucose 분석법 개발 및 검증

        본 연구는 괭생이모자반의 기능성 원료로 개발하고자 원료 표준화를 위해 지표물질을 선정하여 분석법 개발 및 검증을 실시하였다. 괭생이모자반 추출물의 지표성분을 fucose로 설정하고 이에 대한 HPLC 분석법을 확립하여 유효성 검증을 실시하였다. 분석법 유효성 검증은 특이성, 직선성, 정확도, 정밀도(반복성 및 재현성)를 확인하였다. 그 결과 표준용액과 괭생이모자반 추출물 간의 HPLC 크로마토그램의 UV spectrum의 일치 여부를 확인하였고 다른 물질과 간섭 없이 피크가 분리된 것을 확인하였다. 또한 표준용액 검량선의 결정계수(R2)는 3회 분석 모두 0.998 이상으로 확인되어 적합하였다. 정확도는 시료 약 25 mg을 취한 후, 시료 중에 함유된 지표(기능)성분의 함량이 50%, 100%, 200%의 농도가 되도록 표준용액(237.9 ㎍/mL)을 0.25 mL, 0.5 mL, 1 mL씩 첨가하여 회수율을 확인한 결과 94.0~95.6%로 나타나 정확도가 있는 것으로 확인되었다. 정밀도 확인에서도 시료량의 50%, 100%, 200%에 해당하는 양을 정량하여 5회 이상 반복 측정한 결과 상대표준편차(%RSD)는 0.94%로 나타났고, 시료량을 정량하여 시험자, 기기, 일자를 달리하 여 5회 이상 반복 측정하여 상대표준편차를 구한 결과 상대표준편차(%RSD)는 0.47%로 나타나 재현성이 확인되었다. 이와 같은 분석 결과를 통해 확립된 괭생이모자반의 지표물질인 fucose의 HPLC 분석법은 적합한 시험법인 것으로 검증되었으며 본 시험법은 향후 괭생이모자반의 기능성 원료개발과 표준화를 위한 자료 및 품질관리를 위한 자료로 활용될 수 있을 것으로 보인다. This study developed and validated a high-performance liquid chromatography (HPLC) analysis method for the quantification of fucose in Sargassum horneri extracts. HPLC was performed using a YMC-Pack ODS-AQ column (250 ㎜×4.6 ㎜, 5 ㎛) with a flow rate of 1.0 mL/min at 40℃, and detection was conducted at a wavelength of 250 ㎚. The validation of the method was assessed by evaluating specificity, accuracy, precision, limit of detection, limit of quantitation, and linearity. With respect to specificity, the retention time of the fucose standard solution matched that of the fucose in the Sargassum horneri extract. The linearity exhibited a correlation coefficient (R²) above 0.998. The accuracy assessment revealed a recovery rate ranging from 94.0±2.5% to 95.6±0.4% for different concentrations. The overall relative standard deviation (%RSD) for precision was 0.94%, indicating good repeatability, and the mean value over three days of analysis was 9.69±0.05 ㎎/g with a %RSD of 0.47%, demonstrating reproducibility. The limits of detection and quantitation were determined as 2.04 ㎍/mL and 6.19 ㎍/mL, respectively. Based on these results, the developed HPLC analysis method for fucose in Sargassum horneri extract could be considered reliable and reproducible.

      • Bleomycin 纖維肺症의 彈力組織의 變化

        저자는 폐의 결합조직이 증가하는 질환의 탄력조직의 미세구조의 변화를 규명하기 위하여 bleomycin 폐섬유증의 간질조직을 검색하였다. 채중 약 280gm 정도의 Sprague-Dawely계 흰쥐 수컷에 bleomycin 1.5 단위를 기관지로 주입하여 1주, 2주, 4주 및 8주에 폐조직을 광학 및 전자현미경으로 관찰하였으며 아울러 탄력조직의 미세구조를 검색하기 위해 Kajikawa 염색을 하여 관찰하였다. 광학현미경적으로는 bleomycin투여후 1주부터 호중구 및 대식세포가 증가되었으며 2주부터 부분적으로 폐포벽의 비후가 일어나 4주에는 현저하였으며 그 이후 호중구는 감소되었으나 섬유폐는 지속되었다. 전자현미경적으로는 탄력섬유의 양은 bleomycin 투여후부터 감소되었으며 8주에서는 다시 증가되었다. 탄력조직의 미세구조는 bleomycin 투여후 초기에는 주로 미세섬유가 적은 elastin이었으나 8주에서는 미세섬유가 풍부한 elastin이 많았다. 이상의 소견으로 보아 bleomycin투여 후에는 탄력조직이 감소되며 8주에서 다시 신생하는 것으로 생각된다. 그리고 탄력조직의 감소는 bleomycin 투여 후 증가된 호중구와 대식세포에서 유래된 elastin의 작용에 기인될 것이라 생각된다. This study was carried out to investigate the ultrastructural changes of elastic tissue in fibrotic lung disease increasing connective tissues in the interstitium of the lungs. Male Sprague-Dawley rats were given 1.5 unit of bleomycin which induces pulmonary fibrosis. The rats were observed by light microscopy, electron microscopy, and cytochemical electron microscopy(Kajikawa stain) for ultrastructure of elastic tissue. On light microscopy, neutrophils and alveolar macrophages increased in number at 1 week after bleomycin administration. Two weeks after administration alveolar wall was thickened and more pronounced thereafter. But infiltration of neutrophils was recovered. On electron microscopy, the amount of elastic tissue was decreased by bleomycin administration and almost of elastic tissue disappeared at 2 to 4 weeks after bleomycin administration. The elastic tissue was restored at 8 weeks after administration. The ultrastructure of the elastic tissue was predominantly elastin with few microfilaments at early stages of belomycin lung but in late stage restored elastic tissue was newly formed elastin with abundant microfilaments. Based on these findings, it was considered that the elastic tissue was disappeared in early stage of bleomycin lung and was restored at late stage. The possible mechanism of volume change of elastic tissue after bleomycin administration is activity of elastase derived from the neutrophils and alveolar macrophages which increaced due to neutrophilic alveolitis.

        Rat 대동맥 평활근 세포에서 배양액내 Nitrite 농도에 미치는 Guanylate Cyclase Inhibitor의 영향

        Purpose: Nitric oxide (NO) exerts the relaxant effect in vascular smooth muscle cells (VSMC) by activating soluble guanylate cyclase (sGC), which produces cyclic guanidine monophosphate (cGMP) in the cell. This study was undertaken to investigate the mechanism of the inhibitory actions of sGC inhibitors, LY 83583 and methylene blue in the VSMC. Methods: VSMC was primarily cultured from rat aorta and confirmed by immunocytochemistry of anti-smooth muscle myosin antibody. Bacterial lipopolysacchride (LPS), an inducer of inducible nitric oxide synthase (iNOS) and sodium nitroprusside (SNP), an NO donor, were uesd to increase NO within VSMC. The changes in concentrations of nitrite in culture media by an addition of LPS or SNP with a pretreatment of LY 83583 or methylene blue were measured by the spectrophotometry with griess regent and absorbance at 550 nm. Western blot and RT-PCR for iNOS and iNOS mRNA, respectively were performed. Results: LPS and SNP increased nitrite concentration. LY 83583 potentiated the increase in nitrite concentration by LPS and SNP. LY 83583 also increased expressions of iNOS protein and mRNA induced by LPS. Methylene blue has no effect on nitrite concentration increased by LPS or SNP, and it did not affect the expressions of iNOS protein or mRNA induced by LPS. Conclusion: These results suggest that the mechanism of inhibitory actions of LY83583 and methylene blue on sGC are different each other; LY83583 interferes the interaction of sGC and NO resulting positive feedback increase in iNOS gene expression, but methylene blue eliminates NO from cytosol inducing no compensatory effect.
