        전기영동과 산알카리처리 결합방법에 의한 돼지정액의 X 와 Y 정자 분리에 관한 연구

        The combination of the acid-alkali method and eletrophoresis for X-and Y-sperm separation of boar semen was performed We used lactic acid and sodium bicarbonate in acid-alkali method, and electrophoresed at 2.5V and 200 μA. The results obtained were summarized as follows : 1. The frequency of appearance of B-body from alkali treatment of whole semen was significantly higher than that from acid treatment. 2. The frequency of appearance of B-body from alkali treatment at the anode electrode was significantly higher compared with that from acid treatment at the same electrode, while that from acid treatment at the cathode was slightly higher than that from alkali treatment at the same electrode. 3. According to time of treatment, Appearance of B-body from alkali treatment at the anode after electrophonesis was decreased, and the result from acid treatment at the same condition was the opposite, while appearance of B-body from alkali treatment at the cathode after electrophoresis was not changed and that from acid treatment at the same condition was increased. 4. Appearance of B-body from alkali treatment at the anode for 5-10minutes showed the most useful result which was higher than the result (51.9%) from electrophoresis alone by 3 %.

        한우의 조정기능발달에 관한 연구 1 . 한우와 교잡종의 전반기 고환발육과 조정기능

        In order to study on the development of the spermatogenic function and the sexual maturity between the Korean purebred and three-way crossbred ((Angus × Korean purebred) × Holstein), this experiment was conducted to investigate the testicular development and the spermatogenetic function by means of repeating biopsy at one month intervals from one to 9 months of age with 6 Korean purebreds and 3 three-way crossbreds. 1. In the testicular development, the scrotum circumference of the Korean purebred and three-way crossbred increased steadily with increasing age. The scrotum circumferences of the Korean purebred and three-way crossbred were 19.4 ㎝ and 28.9 ㎝ at 9 months of age, respectively, and doubled to one month of age. The testicular width and length increased rapidly from 6-7 months of age, but the differences between two groups were not statistically significant. 2. The seminiferous tubules in the testis of two groups, solid at 3 months of age, increased appearently in diameter at 5 to 6 months of age, but were not reached mature size. 3. Only gonocytes and indifferent supporting cells were present in the tubules at 3 months of age and spermatogonia appeared from 5 months of age, spermatocytes increased rapidly from 6 months of age and spermatid were seen for the first time in the 7 months old Korean purebred and three-way crossbred. On the basis of the above results it is concluded that sexual maturity of three-way cross bulls was somewhat earlier than the Korean bulls, but the difference of two groups was not significant.

        정소 Biopsy 와 여름철 기온이 재래산양의 정액성상과 정소조직에 미치는 영향

        This experiment was carried out to determine the effects of hot environmental temperature in summer and repeated testicular biopsy on semen quality and testicular structure in Korean native goats. The testes of five goats were biopsied repeatedly once a month for 5 months from May 10, 1970. Semen was collected with an electro-ejaculator and artificial vagina, and evaluated once a week from the early June. The volumetric analyses for the testicular structures biopsied were also accomplised. The results obtained were as follows: 1) Repeated testicular biopsy of 5 times in total had no significantly detrimental effects on semen quality through the experimental period. 2) Significant decrease in semen volume, sperm concentration, sperm motility and % alive sperm were observed successively from the early August to the early September. 3) With the increase in pH of semen, high increase of morphologically abnormal spermatozoa(mostly tailless) appeared in all the goats used from the late August to the early September. 4) Between the two methods of semen collection, that is, electro-ejaculator and artificial vagina, there was significant difference in pH of semen, sperm motility, % alive sperm, concentration, but none in the other semen criterial evaluated. 5) The volumetric analysis for the testicular structures showed no significant monthly changes, though slightly fewer spermatids and more lumen were observed in September than in the other months. 6) From the above results, it could be recommended that the breeding male goats be sheltered under slightly coolish environment through the late July to the early September to improved in their fertility in the fall.

        성성숙기를 통한 재래산양의 정소 Biopsy 에 관한 연구

        Ten male of Korean native goats which were composed of 5 twins were weaned at 10 weeks of age. Both testicles of one of the twins were biopsied 4 times alternately at 2 week intervals from 12 weeks of age onwards. The time when spermatozoa first appeared in the ejaculates of all the bucks used were 24 weeks of age, and the body weight at that time averaged 10.7㎏ in the control and 12.7㎏ in the biopsy group. Since 24 weeks of age, the improvement of semen quality and sperm production has been observed till 32 weeks of age. There were no significant differences in the development of body and testes and some semen characteristics between the two groups. The first appearance of round spermatids was preceded to the spermatozoa presence in testes by 2 weeks and to the spermatozoa in ejaculates by 4 weeks.

        육성 - 비육돈에 대한 Selenium 의 급여효과

        This experiment was carried out with growing-finishing swine to investigate the effects of Selenium on the growth rate, feed efficiency and carcass qualities. Fifty heads of weaned piglets were allotted to following three groups. 1. Twenty heads for selmix which was produced by Jung Ang Chemical (Pharmaceutical Co.). 2. Ten heads for natural selenite which was produced by Hyochang Mineral Research Center. 3. Twenty heads for control group. Selmix and Selenite were added to supply 0.3 ppm. of Selenium to basal ration which was formulated by Won-Kang Co. Ltd. This study had been lasted for 108 days from May 27, 87 to September 12. 87. and the results were obtained as follows a) The daily body gain: The best result was obtained in Sehnix group, ave, daily gain, 708.6 gr. (P$lt;0.05) compared to Selenite group, 674.7 gr. and control group, 663.6 gr. b) Feed efficiency: There were no significant differences in the feed consumed and feed conversion. c) Carcass qualities: There were no significant differences in the carcass qualities; backfat thickness, carcass length, carcass weight and melting point of leaf fat, except significant differences in the eye muscle area (P$lt;0.05) and backfat thickness (P$lt;0.05).

        Propylthiouracil 과 Diethylstilbestrol 이 성장웅추의 성선 및 제2차 성미에 미치는 영향

        This experiment was conducted to investigate the influence of Diethylstilbestrol and Propylthiouracil upon the gonad and secondary sex characters of growing cockerels. This trial was alloted four treatments; Control, Diethylstilbestrol, Diethylstilbestrol plus Propylthiouracil, Propylthiouracil. Each lot had 13 growing cockerels which were five weeks old. At the day of testing, three cockerels were randomly sampled from every lot on each week by third week. And at the fourth week, seven cockerels were sampled from each lot. The results obtained were as follow; 1. Testicles weights had no significant differences among treatments by third week. But in fourth week, all the treatments had decreased the weight of testicles compared to control. 2. At fourth week after treatment, diameters of seminiferous tubules of treated individuals were significantly decreased from control. (p$lt;0.01) 3. Comb and wattle areas were significantly decreased in all treatments compared to control.

        유우의 좌우난소간 배란빈도차에 대한 조사연구

        The primary objectives of this study were to observe (1) the variation of functional activity between the right and left ovaries in the dairy cows, and (2) the effect of age to the variation of functional activity and (3) environmental effects around the ovary uterine horn to the rate of fertilization. Data were obtained from 1218 mature follicles and 155 pregnant cows. The result obtained were as follows; 1. The right ovary was more active than the left. And the frequencies of follicle maturation in the right and left ovaries were 763(62.46%) and 455(37.36%) follicles respectively. The variation of this two frequency is significant at 1% level. 2. Under the 6 years the frequencies of follicle maturation in the right and left ovaries were not affected by the age. As compared over the 7 years with under the 6 years, the age did not significantly affect the variation of ovulation frequency between the right and left ovary. 3. The environments around the left ovary and uterine horn did not significantly affect the rate of fertilization and implantation of fertilized egg.

        소에 있어서 비외과적 방법에 의한 수정란의 채란 기술개발에 관한 연구

        These experiments were conducted to develop embryo collection by nonsurgical methods and. establish basis of embryo transfer in bovine. The recovery rate of embryos between natural and superovulated cows, the ovarian response following 3 to 4 times repeated uses of 2000 IU PMSG with some intervals, cleavage stage of embryo and expansion of uterine cervix with laminaria were studied with korean native cows. The results obtained were as follows: 1. Laminaria No. 1 was expanded into 3.2 times in diameter within 3 hrs and 4 times in diameter within 4 hrs when it was set in warm water at 38℃ and uterine cervix, respectively. After removing it from uterine cervix, foley catheters (18-22FR) were easily inserted into uterus without rectal manipulation. 2. Average numbers of corpus lutea on 9-12 days following estrus induced with 2000 IU PMSG in I, II, III and IV replications taking for 60±15, 90±15 and 150 days intervals among replications were 6.0, 10.3, 7.3 and 7.0, respectively. Accordingly, appropriate interval between replications is thought to be 3 to 4 months and can be shorter or extended according to variation of estrus cycle in the individual. 3. Ovarian response following PMSG injection was greater in fall than in summer and winter. 4. The cows showed standing estrus at average 71±2 hrs following PMSG injection and 85% of the cows showed the estrus between 3 to 4 days following PMSG injection. 5. 92.6% of the flushing fluid was recovered by flushing and 24 eggs (34.3%) among 70 corpus lutea were collected from the superovulated cows. 6. The recovery rates of embryos from I. II, III and IV replications were 50, 3.6, 8.6 and 100%, respectively. The shorter the interval between replications was, the lower the recovery rate of embryo was. 7. Six embryos (100%) were collected from the six cows in natural ovulation by nonsurgical method. 8. Cleavage stages of embryos recovered respectively on 5,7,8,10 and 15 days following estrus ware 16-cell, morula, tight morula, early hatching blastocyst and late hatching blastocyst, respectively. 9. Abnormal, non-fertilized and broken ova among 30 ova recovered were 3(10%), 2(6.7%) and 2(6.7%), respectively. 10. In case of cows in natural ovultion, embryo was not recovered in 1S fractions obtained only by flushing, but mostly recovered in 2nd to 5th factions obtained by flushing with massage of uterus. 11. The embryo recovered on the same day following estrus by PMSG showed 1½±½ difference in cleavage stage between Holstein and Korean native cows and also showed 1±½ difference between the natural and superovulated cows.

        Dimethylacetamide 첨가와 동결융해방법이 닭 동결정액의 정자생존성과 수정율에 미치는 영향

        This study was carried out to investigate the effects of DMA level and equilibration, and freezing-thawing methods on sperm viability, motility and fertility in fowl frozen semen. The results were summarized as follows: 1. Higher DMA levels (0, 4, 7, 10 and 14%) decreased sperm viability and motility of diluted semen and there was significant (P$lt;0.05) difference among DMA levlels. 2. The semen frozen with 7% DMA showed the hightest sperm viability (35.0%), and the sperm viability of semen frozen with rapid-freezing process was higher (14%) than that with slow-freezing process (P$lt;0.05). 3. The sperm viability of semen frozen in 7% DMA with and without equilibration was significantly (P$lt;0.05) different between rapid freezing-rapid thawing 43.3 and 31.7 and slow freezing-slow thawing 11.7 and 8.3, and the sperm viability of frozen semen with 1 hr equilibration was higher than that without equilibration. 4. The fertility of fresh semen inseminated before, after and together with 7% DMA deposition was lower (89, 80 and 70%, respectively) than that of without DMA deposition (94%). 5. The fertility of semen frozen with 7% DMA, 1 hr equilibration and rapid freezing-rapid thawing process was 21%.

        돼지의 조정기능발달에 관한 연구

        In order to study on the spermatogenic function and the sexual maturity in male three-way crossbred ((Berkshire × Hampshire) × Landrace), this experiment was conducted to investigate the testicular development and the spermatogenic function by means of repeating biopsy testes at one month internals from one to 4 months of age and 15 days intervals from 4 to 6 months of age and the quality of semen collected after 27 weeks of age. 1. In the testicular development, the scrotum circumference increased steadily with increasing age up to 3 months of age, but increased rapidly after 4 months of age. The correlation between body weights, testicular lengths, testicular widths, and scrotum circumferences were statistically highly significant (p$lt;0.01). 2. The seminiferous tubules increased steadily at 3 months of age, increased apparently in diameter at 4 to 6 months of age. The percentage of intratubular tissue in the testis increased apparently at 4 to 6 months of age. 3. Only indifferent supporting cells, spermatogonia and spermatocytes were present in the seminiferous tubules at 3 months of age. Spermatids were appeared from 4 months of age, the percentage of which increased rapidly up to 5.5 months of age. 4. The first ejaculation of spermatozoa were observed in 1 of the 4 boars at 27 weeks of age, in 2 of the 4 boars at 29 weeks and in all of the 4 boars at 30 weeks. 5. The total volume ejaculated, sperm concentration and percentage of alive spermatozoa per ejaculate averaged 196.7㎖, 284.77x 10^6/㎖, and 80.0, respectively, at 35 weeks of age. On the basis of the above results it is concluded that three-way crossbred boars reached puberty at 27 seeks of age and can be used for breeding at about 35 weeks of age.
