        말뚝버섯 자실체의 항산화, 항콜린에스테라제 및 염증 저해 활성

        Phallus impudicus var. impudicus L. is an edible mushroom that has long been used as folk medicine in China. The aim of this study was to evaluate the antioxidant, anti-cholinesterase, and inflammation inhibitory activities of a methanol extract of fruiting bodies of P. impudicus var. impudicus L. The extract exhibited good 1,1-diphenyl-2-picrylhydrazyl scavenging activity, excellent ferrous ion chelating activity, and moderate hydroxyl radical scavenging activity compared with BHT at 2.0 mg/ml. However, the reducing power of the extract was significantly lower than that the BHT positive control. Although the inhibitory activities of methanol extract on acetylcholinesterase and butyryl cholinesterase were significantly lower than the galanthamine positive control at the concentration tested, the inhibition of acetylcholinesterase and butyryl cholinesterase was 52.83% and 55.17%, respectively, at 1.0 mg/ml. The methanol extract also demonstrated excellent inhibition of inflammation-related activities, such as production of nitric oxide in lipopolysaccharide-induced RAW 264.7 macrophage cells and acute edema induced by administration of carrageenan on the hind paw of rats. The collective results suggest that the fruiting body of P. impudicus var. impudicus L. might be a good source of antioxidant, anti-cholinesterase, and anti-inflammation compounds.

        Gamma-irradiation Reduces Allergenicity of Cochineal Extracts

        Cochineal extract and carmine are natural red dyes, widely used in the food, cosmetics,and pharmaceutical industries. However, since they are extracted from the female cochineal insects, they are also prone to inducing food allergies such as anaphylaxis and asthma. In this study, cochineal extract solutions were subjected to multiple gamma-irradiation dosages (0, 10, 30, and 50 kGy), and measured the quantitative changes in carminic acid and allergenicity as well as the Hunter color values changes. The carminic acid level of gamma-irradiated cochineal extracts were significantly reduced at higher irradiation dosages. The allergenicity of cochineal extracts was evaluated through a competitive ELISA cross-reacting with a polyclonal antibody to carmine. Gamma-irradiation significantly reduced the allergenicity of cochineal extract as the irradiation dose increased. Additionally, the Hunter color values of gamma-irradiated cochineal extract were reduced, which resulted in darker red color. In conclusion, gamma-irradiation can reduce the allergenicity of cochineal extracts with improving its color.

        In vitro antioxidant, anti-hyperglycemic, anti-cholinesterase, and inhibition of nitric oxide production activities of methanol and hot water extracts of Russula rosacea mushroom

        Russula rosacea, a mycorrhizal fungus, has been used for edible and medicinal purposes. This study was conductedto evaluate the in vitro antioxidant, anti-hyperglycemic, anti-cholinesterase, and nitric oxide inhibitory effects of the fruiting bodiesfrom R. rosacea extracted with methanol, and hot water. The 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activitiesof the methanol and hot water extracts (2.0 mg/ml) of R. rosacea were comparable with BHT, the positive control. The chelatingeffects of the mushroom and hot water extracts were significantly higher than that of BHT. The reducing power of methanol andhot water extract (6 mg/ml) were significantly lower than that of BHT. Seven phenolic compounds were detected from acetonitrileand hydrochloric acid solvent extract of the mushroom. alpha-amylase and alpha-glucosidase inhibitory activities of methanol andhot water extracts were lower than that of acarbose, the positive control. The acetylcholinesterase and butyrylcholinesteraseinhibitory effects were moderate compared with galanthamine, the standard drug. Nitric oxide (NO) production inlipopolysaccharide (LPS) induced RAW 264.7 cells were inhibited significantly by the mushroom extracts in a concentrationdependent manner. Therefore, we demonstrated that fruiting bodies of R. rosacea possess in vitro antioxidant, anti-hyperglycemic,anti-cholinesterase, and NO production inhibitory activities. The experimental results suggest that the fruiting bodies of R. rosaceaare good natural antioxidant, anti-hyperglycemic, anti-cholinesterase, and anti-inflammatory sources.

        장수진흙버섯 자실체 추출물의 다양한 생리활성 효과

        본 연구에서는 장수진흙버섯 자실체로부터 메탄올과 열수를 이용해 추출한 물질의 항당뇨, 항콜린에스테라아제, 항염증 효과를 탐색하였다. 항당뇨 실험에서 α-amylase 효소에 대한 메탄올과 열수 추출물의 저해 효과는 1.0~2.0 mg/ml의 농도에서 각각 50.67~61.33%와 52.67~65.00%를 보여 양성대조군인 acarbose의 83.67~96.33%에 비해 크게 낮았으나 α-glucosidase에 대한 메탄올과 열수 추출물의 1.0~2.0 mg/ml의 농도에서의 저해 효과는 각각 74.33~89.67%와 75.67~91.00%를 보여 53.67%와 84.67%를 보인 양성대조군 acarbose에 비해 유의하게 높아 탄수화물 분해효소인 α-glucosidase에 대한 저해 효과가 특이적으로 높았다. 메탄올과 열수 추출물의 아세틸콜린 에스테라아제에 대한 저해 효과는 1.0 mg/ml의 농도에서 각각 96.05%와 94.58%를 보여 양성대조군인 galanthamine (97.80%)과 유사하였으나 butyrylcholinesterase에 대한 저해효과는 1.0 mg/ml의 농도에서 각각 91.05%와 82.27%를 보여 galanthamine의 저해 효과 81.12%에 비해 유의하게 높았다. 염증 저해 효과 실험에서 RAW 264.7 대식세포가 배양되고 있는 배지에 메탄올과 열수 추출물을 각각 전처리 후 염증 매개 물질인 LPS를 1 ㎍/ml 처리하여 24시간 배양 후 NO 생성의 저해 효과를 조사한 결과 각각의 추출물 농도가 증가함에 따라 생성된 NO의 양도 점차 유의하게 감소하는 경향을 나타내었다. Carrageenan의 주사에 의해 흰쥐의 뒷발에 유도된 부종 저해실험에서는 투여한 메탄올 추출물의 농도가 증가함에 따라 뒷발에 유발된 부종의 용적도 농도 의존적으로 점차 감소하는 추세를 나타냈다. 따라서 장수진흙버섯의 자실체에 함유된 물질은 항당뇨, 항콜린에스테라아제 및 항염증 효과를 지니고 있어서 천연 건강식품으로의 개발 가능성이 높은 것으로 사료 된다. Phellinus baumii, a white-rot fungus, has been used for centuries as folk medicine in China, Japan, and Korea. This study aimed to evaluate the in vitro anti-diabetic, and anti-cholinesterase, and in vivo anti-inflammatory effects of the fruiting bodies of P. baumii. The methanol (ME) and hot water (HE) extracts (2.0 mg/mL) of P. baumii fruiting bodies suppressed α-amylase activity, exactly 61.33%, and 65.00%, respectively; of note, acarbose, the positive control, inhibited 93.33% of the α-amylase activity. Moreover, the ME and HE (2.0 mg/mL) inhibited 89.67% and 91.00%, respectively, of the activity of α-glucosidase activity, whereas the same concentration of acarbose suppressed 84.67% of the α-glucosidase activity. The ME and HE (1.0 mg/mL) also inhibited 96.05% and 94.58%, respectively, of the acetylcholinesterase (AChE) activity; galanthamine, the positive control, led to an inhibition of 81.12%. The butyrylcholinesterase (BChE) activity was also inhibited by ME and HE (1.0 mg/mL; 91.05% and 82.27%, respectively); of note, the same concentration of galanthamine suppressed 81.12% of the BChE activity. The production of NO in LPS-induced RAW 264.7 macrophages was significantly suppressed by both ME and HE treatments. Importantly, the carrageenan-activated rat hind-paw edema was significantly reduced 2-6 h after ME administration (50 mg/mL). Taken together, the results suggest that the fruiting bodies of P. baumii have α-amylase, α-glucosidase, α-cholinesterase, and anti-inflammatory activities, and, therefore, may be good natural sources for the promotion of human health.

        Bioconversion of ethanol from various sugars and cellulosic materials by brown rot fungus Phaeolus schweinitzii

        A novel brown rot fungus Phaeolus schweinitzii IUM 5048 was firstly used for ethanol production. It was found that this fungus produced ethanol with various sugars, such as glucose, mannose, galactose and cellobiose at 0.28, 0.22, 0.06, and 0.22 g of ethanol per g of sugar consumed, respectively. This fungus showed relatively good ethanol production from xylose at 0.23 g of ethanol per g of sugar consumed. However, the ethanol conversion rate of arabinose was relatively low (at 0.08 g of ethanol per g sugar). P. schweinitzii was capable of producing ethanol directly from rice straw and corn stalks at 0.11 g and 0.13 g of ethanol per g of substrates, respectively, when the fungus was cultured in a basal medium supplemented with 20 g/L rice straw or corn stalks. These results suggest that P. schweinitzii can hydrolyze cellulose or hemicellulose to fermentable sugars and convert them to ethanol simultaneously under oxygen limited condition.

        Antioxidant and anti-inflammatory activities from fruiting body extracts of Lyophyllum decastes

        Lyophyllum decastes has been used for culinary purpose. The present study was conducted to evaluate the antioxidant and anti-inflammatory effects from methanol, acetone, and hot water extracts of L. decastes fruiting bodies. The acetone and methanol extracts showed the higher 1,1-diphenyl-2-picryl-hydrazy radical scavenging activities than that of the hot water extract at 0.5–2.0 mg/mL and was comparable to the BHT, the positive control. The ferrous ion chelating effects of the mushroom extracts at 0.5–2.0 mg/mL were significantly higher than that of BHT. The reducing power of acetone extract (2.12) was significantly lower than that of BHT (2.73) at 2.0 mg/ mL. The mushroom extracts also showed inhibitory effects on production of nitric oxide (NO), and expression of inducible nitric oxide synthase (iNOS) in lipopolysaccharide-induced murine macrophage cells in a concentration dependent manner. In vivo anti-inflammatory experiment on carrageenan-induced hind-paw edema of rat model, the acetone extract of the mushroom significantly suppressed the carrageenan-induced rat hind paw edema of rats in a dose dependently. The results suggest that the fruiting bodies of Lyophyllum decastes are a good natural resource of antioxidant and anti-inflammation.
