The purpose of this study is to examine how the media handles and reproduces the Olympics by grasping the trend of photographic reporting at the Tokyo Olympics in daily newspapers, identifying topics and reports, and analyzing photographic images published in newspapers. It is also important to observe how our media reproduce by looking at the differences in media that appear through Olympic photo images. For this study, a total of 539 photos were analyzed by selecting press photos posted in general daily newspapers such as the Chosun Ilbo, the Hankyoreh Newspaper, and the Hankook Ilbo from July 23 to August 9, 2021. As a result of the analysis, the area average of the photos showed significant differences between media. In addition, in the case of a front-page article, the media to be analyzed reported more than one related photo during the Olympics. Even when the subject of photography was women, the degree of exposure differed from medium to medium. The main contents of the photo report were “physical athletic ability” and “the joy, pleasure and cheers of victory.” However, the Chosun Ilbo showed a lower percentage of reports related to COVID-19 than the other two media companies. The theme of all newspapers was “victory advantage.” However, the increase in the proportion of humanism is interpreted as meaning that the tendency to report gold-oriented past Olympics is changing.
시호(Bupleurum falcatum L.)의 모상근 배양을 통한 Saponin 생산 I. 캘러스, 부정근, 모상근과 재배근의 Saponin 양상과 함량 비교

In order to survey possibility to produce saikosaponin from in vitro hairy root culture, culture of callus, adventitous root, and hairy root of Bupleurum falcatum L. were estabilished, and quantitative and qualitative aspects in saikosaponin extracted from these were compared with those of cultivated root. Callus grew well in MS medium containing 0.9 $\mu$M 2, 4-D. In contrast, both of adventitous root and hairy root grew well in hormone-free MS medium. However, hairy root showed more rapid growth with extensive lateral root branches, characteristics of lower content of water and softer than in adventitous root. Among the selected lines of adventitous root and hairy root were observed difference in the growth rate. Mannopine, one of opine synthesized in the transformed tissue with Agrobacterium rhizogenes. A4 were detected in the extract of hairy root lines. Pattern and content of crude saponin from adventitous and hairy root showed no difference, but somewhat difference from those of cultivated root. However, in callus, distinct production-aspect of saponin was not observed.

Saikosaponin productivity was examined in a Bupleurum falcatum L. BFHR2 hairy root culture in re-sponse to changes in the sucrose content (2~8%), nitrogen content (0~250 mM NH4NO3), phosphate content (0~12 mM NaH2PO4), and the potassium content (0~87.2 mM KCl) of the culture media. We found that the conditions for maximal saikosaponin production differed from those for optimal root growth. Highest saikosaponin yield was achieved for 8% sucrose, 62 mM NH4NO3, 1.2 mM NaH2PO4, and 0.5 mM KCl.
미치광이풀 모상근 배양에서 적정 sucrose 농도 및 hydroxyapatite 첨가에 의한 tropane alkaloid 생산성 향상

미치광이풀(Socopolia parviflora Nakai) 모상근 배양에서 모상근 생장과 tropane alkaloid 생산에 대한 sucrose농도와 일부 흡착제의 효과를 조사하였다. 모상근 클론 SP 11은 5% sucrose 농도를 함유한 1/2 B5 배지에서 최적인 생장과 tropane alkaloid 생산을 보였다. 모상근에서 tropane alkaloids 생성양상은 모식물에서와 약간의 차이를 보였고 그 중 식물체에서 검출되지 않는 littorine의 생성이 모상근에서 두드러졌다. 조사된 흡착제(0.01% 1%) 중에서, charcoal 0.01%와 XAD-II l%에서 약간의 생장촉진효과를 보인 것을 제외하고는 그 외 농도나 흡착제(amberlita chitosan)에서는 억제효과 또는 유의적인 변화를 보이지 않았다. 반면에 hydroxyapatite는 미치광이풀 모상근의 생장과 tropane alkaloid 생산에 있어 촉진효과가 있음이 확인되었다. The effects of sucrose concentration and some absorbents on growth and tropane alkaloid production in hairy root cultures of Scopolia parviflora were investigated. The maximum effect on growth and tropane alkaloid production in hairy root clone SP11 was obtained for 1/2 B5 medium containing 5% sucrose. The production pattern of tropane alkaloid in hairy roots was some different from that of rhizome of mother plant, particulary showing high littorine contents, which was not found in ordinary roots. Among absorbents examined, charcoal 0.01% and XAD-II 1% made a slight growth promotion effect, whereas the other concentration of charcoal, XAD-II and absorbents (amberlite and chitosan) showed inhibition or no significant effect. The addition of hydroxyapatite enhanced the production of tropane alkaloids significantly than control cultures.

시호 모상근 (BFHR2 클론) 배양에서 생장과 saikosaponin 생산에 미치는 기본배지, 배지의 다량무기염농도 변화 및 IAA와 IBA의 효과를 조사하였다. 조사한 배양액 중 RCM 배지에 다량무기염농도를 2배로 증가시킨 배지에서 생장이 가장 양호하였다. 또한 2RCM배지에서는 saikosaponin인 a, c와 d의 생성을 확인하였으며, 반면에 MS배지에서는 saikosaponin의 생성이 극히 저조하였다. IBA는 0.5mg/L에서 약 60%의 생장촉진효과를 보였으나, IAA와 IBA두 가지 모두 0.01~5mg/L 까지의 농도에서 뿌리형태의 외적인 변화 없이 saikosaponin 생성을 강하게 억제하는 작용을 나타내었다. We examined the effects of various basal media and exogenous auxins on the growth and saikosaponin production in hairy root cultures of Bupleurum falcatum. Of the various media tested, 2RCM medium was superior for growth and saikosaponin content, in which saikosaponin a, c, and d was produced, however MS medium was resulted in extremely low level in saikosaponin production. By the treatment of 0.5 mg/L IBA the growth rate was increased by about 60% in 2RCM medium, whereas both IAA and IBA showed a potent inhibition effect in saikosaponin biosynthesis by the treatment of 0.01 mg/L~5mg/L.
