A high-performance liquid chromatographic method with fluorometric detection was evaluated for analysis of ofloxacin in plasma. Biological fluids (plasma, $200\;{\mu}L$) were prepared for assay by protein precipitation with chlorofurm. The detection of ofloxacin and triamterene as an internal standard were performed at 358 nm for excitation and 495 nm for emission. The HPLC separation was carried out on Ultrasphere ODS column (4.6 mm${\times}25\;cm,\;5\;{\mu} m$) with acetonitrile $(45\%)$-phosphoric acid $(1.5\%)\;containing\;0.3\%$ sodium laurylsulfate as the mobile phase. The flow-rate was 1.0 mL/min. The calibration graphs were linear from 3.0 to 80 ng/mL with r=0.998. The minimal detectable concentration in plasma was 1.5 ng/mL. The proposed technique is reproducible, selective, reliable and sensitive.
다당유도체로 공유결합된 카이랄 칼럼에서 이동상과 분석물질의 용매가 거울상 이성질체의 광학분할에 미치는 영향

공유결합된 다당유도체에서 유도된 카이랄 칼럼인 Chiralpak IC을 사용하여 N-BOC $\alpha$-amino acid와 그들의 ethyl ester 유도체의 액체크로마토그래피 거울상 이성질체의 분리를 수행하였다. 다양 한 이동상에서 공유결합된 카이랄 칼럼인 Chiralpak IC에서 거울상 이성질체가 분리된 실험결과를 나 타내었으며, 광학분할의 선택성과 분리인자가 이동상에 따라 크게 영향을 받았다. 또한 동일한 이동 상에서 분석물질의 녹이는 용매에 따라 거울상 이성질체의 분리에 미치는 영향과 분석용 컬럼에서 거울상 이성질체의 분리의 여러 예를 나타내었다. Liquid chromatographic enantiomer separation of N-tert-butoxycarbonyl (BOC) $\alpha$-amino acids and their ethyl esters was performed on covalently immobilized chiral column (Chiralpak IC) derived from polysaccharide derivative. The solvent versatility of the covalently immobilized Chiralpak IC in enantiomer separation of N-BOC $\alpha$-amino acid ethyl ester derivatives was shown and the chromatographic parameters of their enantioselectivities and resolution factors were greatly influenced by the nature of the mobile phase. Also the effect on the dissolving solvent for these analytes on the enantiomer separation using the same mobile phase and the examples of preparative enantiomer separation on analytical column were shown.
Liposome - Insulin 의 토끼직장투여에 있어서 혈당치의 변화

Insulin administration rectally with the liposome has the hypoglycemic effect in the rabbits. The reducton in blood sugar was maximum at about 60 minutes after administration and continued for 4 hrs. at low level in these experiments. No hypoglycemic effect was observed in control administrated rectally without liposome. Rectal absorption of insulin has been effected by addition of the bile salt, as the protective agent which prevented denaturation and the phastransition of insulin in liposome-encapsulation. As a matter of the fact, a significant hypoglycemic action was obtained when the insulin-liposome was given by rectal administration. The use of this agent to enhance insulin absorption offers the possibility of a new approach to rectal insulin therapy.
