국화에 발생하는 반쪽시들음병균 Verticillium dahliae 검출용 등온 증폭법 개발

Verticillium wilt disease is caused by a fungal plant pathogen Verticillium dahliae, which attacks commercial crops such as chrysanthemum. The conventional methods so far used to identify this fungal pathogen require high expertise and are time-consuming. Therefore, in this study, we developed an assay for the rapid and specific detection of V. dahliae infection using loop-mediated isothermal amplification (LAMP) method. For this assay, four primers for LAMP were designed for targeting cellulose-growth-specific protein partial mRNA gene in Verticillium dahliae. Under standard condition, the optimum reaction temperature for amplification is around 60 ℃ within 60 minutes. This LAMP assay was designed to amplify only present in V. dahliae. When this LAMP assay applied to the DNAs for four other soil-borne fungi and host plants, no amplification was detected. Therefore, this LAMP assay we developed for V. dahliae is expected to do detection at the early stage of its infection. The fast and reliable detection method will allow us to develop effective management system to monitor and control infection of this pathogen in chrysanthemum plant.
양파 노균병균 Peronospora destructor의 분자계통학적 유연관계 분석과 PCR 검출기술 개발

우리나라 양파 재배지의 주요 곰팡이병인 양파 노균병의 원인균 Peronospora destructor를 채집하여, internal transcribed spacer (ITS) 영역을 포함한 4개 유전자의 염기서열을 분석하여 상동성 비교와 분자계통학적 유연관계를 분석하였다. 그 결과, 경남 창녕군, 함안군, 합천군과 전남 무안군, 신안군, 해남군에 발생하는 P. destructor의 4종의 유전자 염기서열 모두 100% 상동성을 보였고, 유연관계 분석에서도 모두 동일한 계통으로 확인되었다. 본 연구에서는 ITS 영역 유전자 염기서열을 이용하여 P. destructor 검출용 PCR법을 개발하였고, 노균병균만을 특이적으로 검출하였다. 또한 식물체를 포함한 total genomic DNA로 검출한계를 검정한 결과, 0.07 ng/μL까지 가능하였다. 양파 노균병균 검출용 PCR법은 육안상 병징이 나타나지 않을 때도 충분히 감염유무가 확인되었다. Onion downy mildew, caused by Peronospora destructor, is a major disease in onion cultivation areas in Korea. The causal fungi were collected and analyzed based on sequence similarity and molecular phylogenetic relationships of multi-gene sequences, including the internal transcribed spacer (ITS) region. All isolates from Changnyeong-gun, Hamyang-gun, and Hapcheon-gun in Gyeongnam province, and Muan-gun, Haenam-gun, and Sinan-gun in Jeonnam province were identical in the four types of gene sequences, indicating they were genetically the same strains. In this study, a PCR method was developed based on the ITS gene sequences to amplify the specific DNA fragment for P. destructor only. The detection limit of was total genomic DNA of the P. destructor and the plant 0.7 ng/μL. Therefore, the developed PCR method could be used to detect P. destructor effectively from symptomless onion leaves.
도마운동 Cuervo Stretched 동작의 국면별 역학적 분석

본 연구의 목적은 도마 Cuervo Stretched 동작의 도약 국면별 운동역학적 변인의 차이가 제 2비약의 공중 회전운동과 착지 제어에 어떠한 영향이 미치는가를 규명하는데 있다. 이를 위해 국가대표 남자 기계체조 선수 3명을 대상으로 고속 비디오 카메라를 이용한 3차원적 영상분석과 구름판과 도마에 가해지는 힘의 측정을 통하여 운동역학적 변인들을 비교 분석한 결과 다음과 같은 결론을 얻었다. 1. 발구름 시 우수한 동작일수록 고관절을 좁히면서 구름판에 접근하고 신체중심을 앞으로 빠르게 이동시키며 동시에 견관절을 빠르게 좁혀주어 구름판 이륙 시 신체를 앞으로 많이 숙이는 것으로 나타났다. 이는 제 1비약의 체공시간을 짧게 하고 다리를 뒤로 차주는 동작을 용이하게 하여 신체의 회전 속도를 빠르게 유발시키는 것으로 나타났다. 2. 도마 접촉 국면 시 견관절각의 큰 신전과 함께 대퇴분절의 빠른 각속도 유지는 도마 접촉 시간을 짧게 하고 수직성분을 강화시키며 도마 이륙각을 수직에 가깝게 유지시킴으로서 제 2비약의 체공 높이와 거리를 증가시키고 제 2비약의 정점에서 공중 회전거리를 많이 확보할 뿐만 아니라 도마에 가해지는 힘을 크게 하여 탄성을 일으키는데 큰 기여를 하는 것으로 나타났다. 3. 발구름 시 강한 수평반력에 따른 제 1비약의 수직성분 강화는 도마 접촉 시 소요시간을 길게 하고 견관절의 각변위를 작아지게 만드는 것으로 나타났으며, 이는 신체를 앞으로 쓰러지게 하고 수직속도를 약화시킴으로서 제 2비약의 도약력 상승과 공중 회전운동에 나쁜 영향을 제공하는 것으로 나타났다. 또한 공중 회전운동 초반의 소극적인 움직임은 뒤늦게까지 회전운동의 강화에 치중하게 되어 착지 시 좁은 고관절각과 낮은 중심고를 제공하여 불안정한 착지 동작을 유발시키는 것으로 나타났다. This paper aims to closely examine how the difference of kinetic variables by take-off state of horse vaulting Cuervo stretched motion would have an effect on the rotation of the post flight and landing. For this, to three male national artistic gymnastics athletics a three dimensional image analysis using the high speed video camera was made and the force applied to the board was measured, and as a result of comparing and analyzing their kinetic variables, following conclusion was obtained. 1. It was indicated that more skilful motion at contact board approaches the contact board with hip joints in narrow, and moves cog. forward rapidly, and narrows shoulder joint, resulting in making the body bending forward at take-off state. In was indicated that this shortens the duration of pre-flight, and facilitates kicking motion, resulting the rotation velocity of the body. 2. Indicated that at the phase of contacting horse vaulting, large extension of shoulder joint angle and maintenance of rapid angular velocity shortens the contact time of horse vaulting, consolidates vertical components, keeps the take-off angle of horse vaulting close to the vertical state, increases the post-flight height and distance, secures rotation distance in the airborne at the peak of the post-flight, and generates elasticity at horse vaulting. 3. It was indicated the consolidation of the vertical component in the pre-flight according to strong horizontal reaction power prolongs required time at the time of contact horse vaulting, and reduces the angular displacement of shoulder joint, which makes the body fall ahead, and weakens vertical velocity, and provides disadvantage to increase and rotation motion of the take-off power at the post-flight, and the initial passive motion of air rotation motion focuses on the consolidation of rotation motion till late, so provides narrow hip joint angle and low cog. height at landing, resulting in unstable landing motion.

This study has intended to devise a comprehensive rural planning, driven by residents based on ecological rural amenity paradigm for Busu area, Boeun-gun, South Korea. To reach the goal, this study has performed the following processes. First, we have analyzed the elements of threat, opportunity, weakness, and strength in both inside and outside village through `SWOT` analysis. Second, through strategic analysis and consultation, we have proposed developmental directions of Busu area. Third, based on an ecological rural amenity planning system composed of ecological economy system, ecological environment system, ecological history-culture system, ecological image system, and ecological society system, we have suggested research projects of each system. Fourth, we selected projects through a general meeting with all stakeholders. Fifth, the selected projects were applied to Busu area by village residents and experts. Finally, the projects, which were appraised and revised by experts, residents, and governmental officers, were composed of ecological scenic agriculture center, resident`s site for city dweller, energy independent village, eco-road, eco-pond, ecological park and parking lot, restoration of traditional culture, zone of ecological scenic agriculture, eco-tunnel, eco-fence, landmark, corporate identity, community center, forum and seminar, and education for residents` empowerment.
