        Sprague-Dawley계 흰쥐에서 현지초 부탄올 분획물의 항비만 및 항고지혈증 효과

        The purpose of this study was to investigate anti-obesity and anti-hyperlipidemic effects of extracts from Geranium thunbergii (GT) in high-fat-diet-induced obese rats. Animals were randomly divided into four groups [normal diet, high fat diet, MeOH extract of GT (GTM), and BuOH fraction of GT (GTB)] and GT samples were treated with dose of 100 mg/kg for 8 weeks. It was observed that GTB-treated group significantly reduced body weight gain, food intake, epididymal fat weight, and triglyceride level in serum and liver compared to control group. The rats fed GTB also decreased contents of thiobarbituric acid-reactive substances (TBARS), hydroxyl radical, and xanthin oxidase (XO) increased by high fat diet. Furthermore, the anti-oxidant enzymes such as superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase were increased by GTB treatment. The experimental results indicate that GTB has anti-obesity and anti-hyperlipidemic effects, as well as radical scavenging activity.

        UPLC-DAD를 이용한 불환금정기산의 다성분 동시분석법 개발

        Bulhwangeumjeonggisan (BHGJGS) is a traditional herbal formulation generally used in the treatment of cold and gastritis. BHGJGS consists of eight herbal plants; Atractylodis Rhizoma, Magnoliae Cortex, Citri Pericarpium, Glycyrrhizae Radix, Agastachis Herba, Pinelliae Rhizoma, Zingiberis Rhizoma and Zizyphi Fructus. Complete standardization of this formulation has not been done yet. So, a simple and accurate method was developed and validated using Ultra Performance Liquid Chromatography (UPLC) with Diode Array Detector (DAD) for the standardization of BHGJGS. UPLC conditions were optimized using a c18 RP-Amide column with mobile phase; 0.1% phosphate buffer and acetonitrile, detection wavelength; 210 and 325 nm. The linearities of calibration curves were acceptable (R2>0.9994), and the limit of detection and quantification were within the ranges of 0.011-0.091 and 0.034-0.277 mg/ml respectively. The relative standard deviation (RSD) of intra- and inter-day precisions were under 3.61%. The RSD of repeatability was under 0.68 %. The results of recovery test were 94.4-107.9%, and the RSD were under 4.6%. The developed method was used to find the contents of standard constituents in BHGJGS mix extract powder, and two commercial formulation (A and B). The data show that the developed method was specific, sensitive, accurate, and precise for analysis of BHGJGS components.

        새로운 생약복합제제(HPL-4)의 품질관리를 위한 분석법 개발

        HPL-4 is a new herbal formulation developed for the treatment of osteoarthritis. In this study, we took HPL-4 to develop a method for simultaneous determination of nine marker compounds (protocatechuic acid, chlorogenic acid, liriodendrin, nodakenin, b-D-(3-O-sinapoyl)frucofuranosyl-a-D-(6-O-sinapoyl)glucopyranoside, quercitrin, 6-gingerol, decursin and decursinol angelate) present in the formulation. The analytes were separated by UPLC system consisting of diode array detector at 205 nm and RP-amide column, and solvent system of ACN/0.1%H3PO4. Analytical method was validated to evaluate its linearity, limit of detection (LOD), limit of quantification (LOQ), precision, and accuracy. All standards showed good linearity (R2>0.99) in the rage of 0.25-400.0 mg/mL. The LOD and LOQ were within the range of 0.021-0.148 and 0.070-0.448 mg/mL, respectively. Intra- and inter-day precision was less than RSD 4.0% and the accuracy was range from 92.00-104.81% with RSD<4.2%. The results suggest that the developed UPLC method is precise, accurate and effective, and could be applied for quality control of HPL-4 formulation.
