Transport study of horseradish peroxidase and transferrin-horseradish peroxidase conjugate was performed using an in vitro Caco-2 cell cultured monolayer grown on a polycarbonate membrane of Transwell^ⓡ. Horseradish peroxidase was not transported across Caco-2 cell monolayer. Transferrin-horseradish peroxidase conjugate was transported through Caco-2 cell monolayer. The apparent membrane permeability coefficient (P_(app)) of transferrin horseradish peroxidase conjugate was 6.54×10^(-7) ㎝/sec. The P_(app) value of transferrin-horseradish peroxidase conjugate across Caco-2 cell monolayer was increased to 11.9×10^(-7) ㎝/sec in the presence of 50 ㎍/㎖ brefeldin-A. These results suggest the transferrin receptor mediated transcytosis of transferrin-horseradish peroxidase conjugate across Caco-2 cell monolayer.

Pharmacokinetics of epigallocatechin gallate(EGCG) was studied following i.v. bolus and oral administration in rats. The values of systemic clearance(CL) were 67.9±5.2 and 26.5±1.4 ㎖/min/㎏ following i.v. bolus administration of 1 ㎎ and 5 ㎎ EGCG, respectively. The values of volume of distribution at steady state (Vss) were 380±56 and 835±84 ㎖.㎏ after i.v. bolus administration of 1 ㎎ and 5 ㎎ EGCG, respectively. The decrease in the value of CL and the increase in the value of V_(SS) as a function of EGCG dose (1 ㎎ to 5 ㎎) suggest saturable mechanism(s) responsible for the distribution and elimination of EGCG. The fraction absorbed of EGCG after oral and intraduodenal administration of GTC were 13% and 22% of the dose, respectively. This result suggests a considerable degradation or elimination of EGCG in the gastrointestinal absorption after oral administration in rats.

Clear ceil sarcoma(CCS) is an uncommon soft tissue sarcoma that occurs in tendons and aponeuroses, usually of the lower extremities and is believed to be of neural crest origin that have a capability to produce melanin. These tumors commonly metastasize and have a very poor prognosis. The fine needle aspiration cytologic finding of CCS is not well documented. We recently experienced a case of CCS. The patient was a 54-year-old male with painful swelling of the right inguinal area. Fine needle aspiration cytology revealed polygonal or fusiform tumor cells with clear or granular cytoplasm and vesicular nuclei containing one or two nucleoli. Immunohistochemical staining for S-100 protein and HMB-45 revealed strong positivity and variable developing stages of premelanosomes were observed by electron microscopy in the excised specimen.
