N/A To evaluate the utility of autoradiographic technique in the detection of TRH receptor changes in brain after the various kinds of stimulation or drug administration, we tried the characterization of TRH receptor in mouse brain and autoradiography in rat brain as a preliminary study. The Kd value of [3-H] MeTRH to TRH receptors of adult male ICR mouse brain (cebellum and spina1 cord were excluded) was 3.55+0.6 nM and Bmax was 3.44+0.52 fmol/mg wet tissue by saturation analysis. The Kd value of TRH to TRH receptors was 133.8+28.2 nM by competition analysis. And we could visualize the distrbution of TRH receptors in rat brain by autoradiographic technique.
경련성 환아에서 Valproate에 의한 혈중 지질의 변화와 영향을 주는 인자에 관한 연구

목 적 : 소아기 경련성 질환에서 장기간 valproate의 치료에 의한 혈중 지질 변화와 혈중 지질 변화에 영향을 줄 수 있는 인자로 성별, 치료 전 체질량지수, valproate의 혈중농도 등에 따른 지질의 변화를 알아보기 위해 본 연구를 시행하였다 방 법 : 24개월 이상 valproate를 복용 중인 30명의 환아를 대상으로 치료시작, 치료 후 6개월, 12개월, 24개월에 공복 시 TC, TG, LDL, HDL를 측정하여 valproate 치료에 의한 혈중 지질의 변화와 성별, 치료 전 체질량지수, valproate 혈중농도 등이 혈중지질 변화에 미치는 영향을 분석하였다. 결 과 : 1) Valproate 치료에 의해 TC는 12개월까지 현저한 감소를 보였으며(P<0.05) LDL는 치료 후 12개월까지 감소하였으나 통계적인 의미는 없었다. TG, HDL은 치료 전과 의미 있는 차이를 보이지 않았다. TC/HDL 비는 치료 전 3.2±0.8에서 6개월 3.0±0.5로 치료에 따라 감소되었으나 통계적으로 의미 있는 차이는 없었다. 2) 성별 및 치료 전의 체질량지수에 따른 TC, LDL, HDL, TC/HDL의 차이는 없었으며, TG가 체질량지수 20 이하 군에서 20 이상 군보다 증가를 보였는데 이는 통계학적으로도 차이가 있었다(P<0.05). 3) Valproate 혈중농도가 증가할수록 LDL는 감소했으며(r=-0.2915, P<0.05), TC, TG, HDL valproate 혈중농도와 상관관계를 보이지 않았다. 결 론 : Valproate의 치료는 혈중 지질의 증가를 초래하지 않았으나 비만한 환아 특히 사춘기 전후의 비만한 여아에서는 항경련제의 선택에 이를 고려해야 할 것으로 생각된다. Purpose : Antiepileptic drugs may alter serum lipid status in epileptic patients. We conducted this study to assess the effect of valproate on serum levels of total cholesterol(TC), triglycerides(TG), low-density lipoprotein(LDL), high-density lipoprotein(HDL), and TC/HDL ratio, and to investigate the factors affecting serum lipid status in children with epilepsy who had been receiving valproate therapy. Methods : Thirty epileptic children(16 males, 14 females, mean age 7.4±3.3 years) were evaluated for serum lipid status at the onset and the 6, 12 and 24 months of valproate therapy, and were analysed changes and potential factors of affecting changes such as sex, body mass index, valproate concentration, in serum lipid levels during valproate therapy. Results : TC were significantly lowered during first 12 months of valproate theraphy (P<0.05). LDL were lowered during first 12 months. HDL and TC/HDL ratio were not changed and TG were increased during valproate theraphy but not reach to statistical significance. TC, TG, LDL, and HDL return to pretreatment levels after 24 months of valproate theraphy. TC, LDL, HDL, and TC/HDL ratio changes were not significantly different by sex and initial body mass index, but TG were significantly increased in group of BMI below 20(P<0.05). LDL levels were significantly decreased correlation to serum valproate concentration(r=-0.2915. P<0.05). Conclusion : Our results suggest that valproate therapy would not increase a risk for atherosclerotic disorders in adulthood, but weight gain with a metabolic consequence of obesity would increase risk for atherosclerotic disorders in adulthood.

N/A According to the development of monoclonal antibodies against cyclosporine, it became avaiiable to replace the conventional polyclonal antibody method with new monoclonal antibody method to measure the blood leve1 of cyclosporine by radioimrnunoassay. We compared the results obtained hy the two methods: polyclonal antibody and monoclonal antibody method. The results were ohtaincd as follows: 1) We obtained mean value 137,3 ng/ml and CV 16.1% from plasma sample, and mean values 495.7ng/ml and 1053.8 ng/ml and CVs 19.3% and 17.4% respectively from whole blood sample by polyclonal antibody method. 2) For the two control groups, 100 ng/ml 400 ngjml each, we obtained that the CVs were 20,2% and 14.0% respectively from plasma sample, and 11.9% and 13.1% respectively from whole blood samaple by monoclonal antibody method. In conclusion, we found that ryclosporine RIA was a relatively reliable method to measure blood or plasma concentration. Especially RIA using monocloanl antibody showed less degree of error in measurement compared to polyclonal antibody method.
