B7-H4 is a member of B7 family of co-inhibitory molecules and B7-H4 protein is found to be overexpressed in many human cancers and which is usually associated with poor survival. In this study, we developed a therapeutic vaccine made from a fusion protein composed of a tetanus toxoid (TT) T-helper cell epitope and human B7-H4IgV domain (TT-rhB7-H4IgV). We investigated the anti-tumor effect of the TT-rhB7-H4IgV vaccine in BALB/c mice and SP2/0 myeloma growth was significantly suppressed in mice. The TT-rhB7-H4IgV vaccine induced high-titer specific antibodies in mice. Further, the antibodies induced by TT-rhB7-H4IgV vaccine were capable of depleting SP2/0 cells through complement-dependent cytotoxicity (CDC) in vitro. On the other hand, the poor cellular immune response was irrelevant to the therapeutic efficacy. These results indicate that the recombinant TT-rhB7-H4IgV vaccine might be a useful candidate of immunotherapy for the treatment of some tumors associated with abnormal expression of B7-H4. To Prevent Crack for Thin-wall Workpieces during Milling Process by Torque Feedback

Thin-wall tubular-geometry workpieces have been widely applied in aircraft and medical industries. Though numerous research reports are available that the tool path can be planned on the basis of preset surface profile before actual milling operation is performed, it is still difficult to predict the real-time surface profile errors for peripheral milling of thin-wall tubular workpieces. This research is focused on how to real-time formulate the appropriate applied cutting torque via feedback of spindle motor current. A few suitable cutting conditions which are able to prevent potential break/crack of thin-wall workpieces and enhance productivity but almost retain the same cutting quality is proposed. To achieve this goal, estimated surface profile error on machined parts due to deflections caused by both tool and workpiece is studied. The torque estimation approach by spindle motor current feedback and the corresponding fuzzy logic controller are employed. Compared with constant cutting torque during milling operation in tradition manner, it is observed that the time consumption of milling cycle by aid of the aforesaid fuzzy logic controller is greatly shortened while the resulted cutting accuracy upon finish of workpiece can be almost retained.

Purpose: The aim of this study was to evaluate the clinical efficacy of a Tanner-Whitehouse 3 (TW3)-based fully automated bone age assessment system on hand-wrist radiographs of Korean children and adolescents. Materials and Methods: Hand-wrist radiographs of 80 subjects (40 boys and 40 girls, 7-15 years of age) were collected. The clinical efficacy was evaluated by comparing the bone ages that were determined using the system with those from the reference standard produced by 2 oral and maxillofacial radiologists. Comparisons were conducted using the paired t-test and simple regression analysis. Results: The bone ages estimated with this bone age assessment system were not significantly different from those obtained with the reference standard (P>0.05) and satisfied the equivalence criterion of 0.6 years within the 95% confidence interval (- 0.07 to 0.22), demonstrating excellent performance of the system. Similarly, in the comparisons of gender subgroups, no significant difference in bone age between the values produced by the system and the reference standard was observed (P>0.05 for both boys and girls). The determination coefficients obtained via regression analysis were 0.962, 0.945, and 0.952 for boys, girls, and overall, respectively (P=0.000); hence, the radiologist-determined bone ages and the system-determined bone ages were strongly correlated. Conclusion: This TW3-based system can be effectively used for bone age assessment based on hand-wrist radiographs of Korean children and adolescents.

To estimate the genetic susceptibility for childhood lymphoma, we conducted an association study for 23 cases and 148 controls. Total 1536 tag single nucleotide polymorphisms (SNPs) were selected in 138 candidate gene regions related to immune responses, apoptosis, the cell cycle, and DNA repair. Twelve SNPs were significantly associated with the risk of lymphoma ($P_{trend}$ <0.05) in six genes ($IL1RN$, $IL2$, $IL12RB1$, $JAK3$, $TNFRSF13B$, and $XRCC3$). The most significant association was seen for $IL2$ variant rs2069762 ($OR_{TG+GG}$ vs. TT=3.43 (1.29-9.11), $P_{trend}$=0.002, min$P$=0.005). These findings suggest that common genetic variants in $IL2$ might play a role in the pathogenesis of childhood lymphoma.

Brain-based Correlations Between Psychological Factors and Functional Dyspepsia

Background/Aims Increasing evidence shows involvement of psychological disorders in functional dyspepsia (FD), but how psychological factors exert their influences upon FD remains largely unclear. The purpose of the present study was to explore the brain-based correlations of psychological factors and FD. Methods Based on Fluorine-18-deoxyglucose positron emission tomography-computed tomography, the altered cerebral glycometabolism was investigated in 40 FD patients compared with 20 healthy controls during resting state using statistical parametric mapping software. Results FD patients exhibited increased glucose metabolism in multiple regions relative to controls (P < 0.001, family-wise error corrected). After controlling for the dyspeptic symptoms, increased aberrations persisted within the insula, anterior cingulate cortex (ACC), middle cingulate cortex (MCC) and middle frontal cortex (midFC), which was related to anxiety and depression score. Interestingly, FD patients without anxiety/depression symptoms also showed increased glycometabolism within the insula, ACC, MCC and midFC. Moreover, FD patients with anxiety/depression symptoms exhibited more significant hypermetabolism within the above 4 sites compared with patients without anxiety/depression symptoms.Conclusions Our results suggested that the altered cerebral glycometabolism may be in a vicious cycle of psychological vulnerabilities and increased gastrointestinal symptoms.

Three traditional Chinese medicines, Agrimonia pilosa Ledeb, Grifola umbellata (pers.) Pilat, and Gambogia, are combined to form a compound extract, AGC. In this study, the in vitro and in vivo inhibitory effects of AGC on human gastric carcinoma MGC-803 cells were demonstrated, and the molecular mechanisms underlying these effects are investigated. Our results indicate that AGC inhibited MGC-803 cell growth in a dose-dependent manner as measured by a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay, with an IC?? of about 6.045±0.69 ㎍/㎖. In vivo, AGC inhibited growth of human gastric carcinoma in xenograft tumors in nude mice, and the inhibitory rate reached 55.2% at 300 ㎎/㎏. The pro-apoptotic activity of AGC was attributed to its ability to decrease the expression of Bcl-2 and Pro-caspase3 and increase the expression of Bax. These results demonstrate that AGC can effectively induce programmed cell death and may be a promising anti-tumor drug in human gastric carcinoma.

전면증착법에 의한 W공정에서 부착특성고양층으로 사용되는 TiN막에 대한 CVD W막의 부착특성을 인장법(pulling method)과 스크래치법(scratch method)을 사용하여 조사하고, 주사전자현미경과 반사도측정에 의한 표면거칠기측정, 응력측정 및 SIMS depth profiling 등에 의하여 그 원인을 분석하였다. 스퍼터링법으로 형성한 TiN막상에 바로 W막을 증착한 경우와 TiN막을 열처리한 후에 W막을 증착한 경우 간에 두 막간의 부착특성은 큰 차이를 보였다. 전자의 경우가 후자의 경우보다 부착특성이 더 우수한 것으로 나타났는데, 이것은 열처리하지 않은 TiN막이 열처리한 TiN막에 비해 표면이 더 거칠고, 응력수준이 더 낮으며, 열처리한 TiN막내에는 산소성분이 존재하는 반면, 열처리 하지 않은 TiN막내에는 산소성분이 거의 들어있지 않기 때문이다. 또한 TiN막 두께가 증가함에 따라 응력의 증가로 인하여 TiN막에 대한 W막의 부착강도가 저하되었다. Adhesion of W to TiN which is used as a glue layer in the blanket tungsten process has been investigated using the pulling and scratch methods and analysed using surface roughness measurements by SEM and the reflectivity measurement, the stress measurement and the SIMS depth profiling Significant differences between the adhesion of the CVD tungsten film to the s-TiN and that to the a-TiN were found. It appeared that the fomer were better than the latter, which is due to the facts that the s-TiN film has a rougher surface and a lower stress level and a higher oxygen content than the a-TiN film. Also the ashesion strength was degreaded with increasing the TIN film thickness because of the increase of the TiN film stress.

Adhesion of CVD W to the TiN glue layer in the blanket W process which is a promising candidate for filing contact holes in subhalfmicron ULSIs has been investigated. The adhesion was enhanced with increasing the W film thickness due to the decrease of the TiN film stress. The adhesion strength was increased by the sputter etching of the TiN surface prior to the W deposition owing to the removal of contaminants and the increase of the surface roughness. The adhesion of the W film to the TiN glue layer property was also improved by Ar ion implantation of the TiN surface owing to the activation of the TiN surface. Subhalfmicron ULSI의 컨택홀을 메꾸는 기술로 각광받고있는 전면도포(blanket W)법에서 부착특성고양층(glue layer)인 TiN막에대한 CVD-W의 부착특성을 조사하였다. 부착강도는 텅스텐막의 두께가 증가함에 따라 TiN막내의 응력의 감소로 인해 부착특성이 향상되었다. TiN막표면을 스퍼터 에칭하는 진처리를 실시할 경우 막표면의 불순물 제거, 표면 거칠기 증가등의 효과로 부착특성이 개선되었다. 또한 TiN막표면에 Ar이온 주입에 의한 손상을 줄 경우에도 TiN막 표면이 활성화되어 부착특성이 향상되었다.

MicroRNAs (miRNAs) are endogenous non-coding genes that participate in post-transcription regulation by either degrading mRNA or blocking its translation. It is considered to be very important in regulating insect development and metamorphosis. Insects are the largest group of animals and are extremely valuable in biological and agriculture research. Insects are also important pests to human health and agriculture, and efforts are necessary protect both humans and plants from disease and damage. Despite their importance, insects lag behind mammals, nematodes, and plants in miRNA research. At present, only 279 insect miRNAs have been identified from Drosophila melanogaster, Anopheles gambiae, Apis mellifera, Bombyx mori, and D. pseudoobscura in miRBase, and most of these miRNAs were computationally predicted without experimental validation. Functional analysis of insect miRNAs has only been conducted in D. melanogaster.
