      • Distinct Pro-Apoptotic Properties of Zhejiang Saffron against Human Lung Cancer Via a Caspase-8-9-3 Cascade

        Lung cancer is the leading cause of cancer-related death worldwide. Here we investigated the antitumor effect and mechanism of Zhejiang (Huzhou and Jiande) saffron against lung cancer cell lines, A549 and H446. Using high performance liquid chromatography (HPLC), the contents of crocin I and II were determined. In vitro, MTT assay and annexin-V FITC/PI staining showed cell proliferation activity and apoptosis to be changed in a dose- and time-dependent manner. The inhibition effect of Jiande saffron was the strongest. In vivo, when mice were orally administered saffron extracts at dose of 100mg/kg/d for 28 days, xenograft tumor size was reduced, and ELISA and Western blotting analysis of caspase-3, -8 and -9 exhibited stronger expression and activity than in the control. In summary, saffron from Zhejiang has significant antitumor effects in vitro and in vivo through caspase-8-caspase-9-caspase-3 mediated cell apoptosis. It thus appears to have more potential as a therapeutic agent.

        <P>A metal–organic framework, Cu<SUB>3</SUB>(BTC)<SUB>2</SUB>, was synthesized and applied as an electro-responsive electrorheological material dispersed in insulating oil.       • Research of Regional Forest Fire Prediction Method based on Multivariate Linear Regression

        In order to achieve the predicted speed, high accuracy, the use of simple purpose, forest fire prediction of the key issues is to choose the main predictors. Forest fire prediction involves many factors, some of which are stable factors such as climate, topography, forest characteristics; and some unstable factors, such as fuel moisture content, meteorological factors, and other sources of ignition. Currently leading factor in the prediction of forest fire is often used in the fuel moisture, precipitation or dry days, relative humidity, temperature and wind five factors. In this paper, some of the data Yichun fire nearly a decade predict the forest fire meteorological data analysis, using multivariate linear regression to derive forest fire prediction method in the wireless sensor networks.

      • Overcoming 5-Fu Resistance of Colon Cells through Inhibition of Glut1 by the Specific Inhibitor WZB117

        Background: 5-Fluorouracil (5-FU) is the most commonly used drug in colon cancer therapy. However, despite impressive clinical responses initially, development of drug resistance to 5-Fu in human tumor cells is the primary cause of failure of chemotherapy. In this study, we established a 5-Fu-resistant human colon cancer cell line for comparative chemosensitivity studies. Materials and Methods: Real time PCR and Western blotting were used to determine gene expression levels. Cell viability was measured by MTT assay. Glucose uptake was assess using an Amplex Red Glucose/Glucose Oxidase assay kit. Results: We found that 5-Fu resistance was associated with the overexpression of Glut1 in colon cancer cells. 5-Fu treatment at low toxic concentration induced Glut1 expression. At the same time, upregulation of Glut1 was detected in 5-Fu resistant cells when compared with their parental cells. Importantly, inhibition of Glut1 by a specific inhibitor, WZB117, significantly increased the sensitivity of 5-Fu resistant cells to the drug. Conclusions: This study provides novel information for the future development of targeted therapies for the treatment of chemo-resistant colon cancer patients. In particular it demonstrated that Glut1 inhibitors such as WZB117 may be considered an additional treatment options for patients with 5-Fu resistant colon cancers.

        Prediction of network security based on DS evidence theory

        Network security situation prediction is difficult due to its strong uncertainty, but DS evidence theory performs well in solving the problem of uncertainty. Based on DS evidence theory, this study analyzed the prediction of the network security situation, designed a prediction model based on the improved DS evidence theory, and carried out a simulation experiment. The experimental results showed that the improved method could predict accurately in the case of a large conflict, and had strong anti-jamming abilities as compared with the original method. The experimental results prove the effectiveness of the improved method in the prediction of the network security situation and provide some theoretical basis for the further application of DS evidence theory.

        Factors related to the intention of healthy eating behaviors based on the theory of planned behavior: focused on adults residing in Beijing, China

        Purpose: The theory of planned behavior (TPB) was used to investigate how the psychological constructs of attitude, subjective norms, and perceived behavioral control (PBC) affect the individual intention of behaviors in adults. Social support is also important in enabling the stability of healthy eating. This study examined the relationship between three major constructs of TPB as well as social support and the intention of healthy dietary behaviors in adults residing in Beijing, China using the extended TPB. Methods: The study questionnaire was based on previously validated items and an online survey was conducted from October to November 2020. Using a total of 244 Chinese adults in Beijing, multiple linear regression analysis was used to test the relationships between three major constructs of TPB as well as the social support and intention of healthy eating. Results: Among the three major constructs of TPB, subjective norms (p = 0.044) and PBC (p = 0.000) were significantly related to the behavioral intention of healthy eating (p = 0.000), and the model explained 76.6% of the variance of the behavioral intention from the three constructs of TPB included in the multiple linear regression model. The additional inclusion of social support to the model did not increase the explanatory power of the model to describe the behavioral intention of healthy eating. The subjective norms (p = 0.040) and PBC (p = 0.000) were still significant where social support did not explain the variance of the behavioral intention adequately. Conclusion: The subjective norms and PBC may be potential determinants of the behavioral intention of healthy eating in adults residing in Beijing, China. These study results can be used to promote healthy eating in Chinese adults living in urban areas. Large-scale intervention studies will be needed to determine if social norms and PBC predict the actual behaviors of healthy eating in Chinese adults.

        Piceatannol Inhibits Phorbol Ester-Induced NF-kappa B Activation and COX-2 Expression in Cultured Human Mammary Epithelial Cells.

        <P>There are multiple lines of evidence supporting that inflammation is causally linked to carcinogenesis. Abnormal upregulation of cyclooxygenase-2 (COX-2), a rate-limiting enzyme in the prostaglandin biosynthesis, has been implicated in carcinogenesis. Trans-3,4,3',5'-tetrahydroxystilbene (piceatannol), a naturally occurring hydroxylated stilbene with potent anti-inflammatory and antioxidative activities, has been shown to inhibit the proliferation of several cancer cells by inducing apoptosis or blocking cell cycle progression. In this study, we examined the effect of piceatannol on activation of the nuclear transcription factor NF-kappa B, one of the major transcription factors that regulate proinflammatory COX-2 gene transcription, in human mammary epithelial (MCF-10A) cells treated with the tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA). When pretreated to MCF-10A cells, piceatannol markedly inhibited TPA-induced NF-kappa B DNA binding to a greater extent than resveratrol and oxyresveratrol, stilbene analogs structurally related to piceatannol. Piceatannol also inhibited TPA-induced phosphorylation and degradation of Ikappa Balpha as well as nuclear translocation of the phosphorylated form of p65, the functionally active subunit of NF-kappa B. Likewise, TPA-induced expression of COX-2 was abrogated by piceatannol pretreatment. The thiol reducing agent dithiothreitol abolished the inhibitory effects of piceatannol on NF-kappa B DNA binding activity, suggesting that piceatannol may directly modify NF-kappa B or its regulator through reaction with the cysteine thiol(s).</P>

        Deep removal of sulfur from real diesel by catalytic oxidation with halogen-free ionic liquid

        A halogen-free task-specific ionic liquid (TSIL) of 1-(2-carboxylic acid) ethyl-3-methylimidazolium bisulfate ([(CH2)2COOHmim] [HSO4]) is applied for deep oxidative desulfurization of real diesel as a catalyst and reaction media. The sulfur level of real diesel can be reduced from 200 to 20.5 μg/g at room temperature with small amount of ionic liquid using H2O2 as an oxidant. The used ionic liquid can be easily recycled and no obvious decrease in catalytic activity can be found after recycling five times.

        The CREG1-FBXO27-LAMP2 axis alleviates diabetic cardiomyopathy by promoting autophagy in cardiomyocytes

        Autophagy plays an important role in the development of diabetic cardiomyopathy. Cellular repressor of E1A-stimulated genes 1 (CREG1) is an important myocardial protective factor. The aim of this study was to investigate the effects and mechanisms of CREG1 in diabetic cardiomyopathy. Male C57BL/6 J mice, Creg1 transgenic mice and cardiac-specific knockout mice were used to establish a type 2 diabetes model. Small animal ultrasound, Masson’s staining and western blotting were used to evaluate cardiac function, myocardial fibrosis and autophagy. Neonatal mouse cardiomyocytes (NMCMs) were stimulated with palmitate, and the effects of CREG1 on NMCMs autophagy were examined. CREG1 deficiency exacerbated cardiac dysfunction, cardiac hypertrophy and fibrosis in mice with diabetic cardiomyopathy, which was accompanied by exacerbated autophagy dysfunction. CREG1 overexpression improved cardiac function and ameliorated cardiac hypertrophy and fibrosis in diabetic cardiomyopathy by improving autophagy. CREG1 protein expression was decreased in palmitate-induced NMCMs. CREG1 knockdown exacerbated cardiomyocyte hypertrophy and inhibited autophagy. CREG1 overexpression inhibited cardiomyocyte hypertrophy and improved autophagy. LAMP2 overexpression reversed the effect of CREG1 knockdown on palmitate-induced inhibition of cardiomyocyte autophagy. CREG1 inhibited LAMP2 protein degradation by inhibiting the protein expression of F-box protein 27 (FBXO27). Our findings indicate new roles of CREG1 in the development of diabetic cardiomyopathy.
