β-Glucosidase, which hydrolyzes cellobiose into two glucoses, plays an important role in the process of saccharification of the lignocellulosic biomass. In this study, we optimized the activity of β-glucosidase of brown-rot fungus Fomitopsis pinicola KCTC 6208 using the response surface methodology (RSM) with various concentrations of glucose, yeast extract and ascorbic acid, which are the most significant nutrients for activity of β-glucosidase. The highest activity of β-glucosidase was achieved 3.02% of glucose, 4.35% of yeast extract, and 7.41% ascorbic acid where ascorbic acid was most effective. The maximum activity of β-glucosidase predicted by the RSM was 15.34 U/mg, which was similar to the experimental value 14.90 U/mg at the 16th day of incubation. This optimized activity of β-glucosidase was 23.6 times higher than the preliminary activity value, 0.63 U/mg, and was also much higher than previous values reported in other fungi strains. Therefore, a simplified medium supplemented with a cheap vitamin source, such as ascorbic acid, could be a cost effective mean of increasing β-glucosidase activity.
Preferential Ganglion Cell Loss in the Nasal Hemiretina in Patients With Pituitary Tumor

<P>Background: Analysis of retinal nerve fiber layer thickness and macular ganglion cell layer loss provides a noninvasive method to assess optic chiasmal compression. These techniques may provide valuable data for patient evaluation and management when combined with findings on clinical examination and neuroimaging results. Methods: Data from 20 eyes of 10 patients with pituitary tumor treated at Inha University Hospital from 2011 to 2013 were collected. This included results of ophthalmologic examination, fundus photography, spectral domain optical coherence tomography (SD-OCT), automated visual field testing, and brain magnetic resonance imaging (MRI). Abnormal color patterns on thickness and deviation maps obtained by macular ganglion cell analysis (GCA) were evaluated and compared with visual field defects. Results: Patients with pituitary tumor showed preferential ganglion cell loss in the nasal hemiretina and characteristic vertical midline-respecting perimacular ganglion cell-inner plexiform layer defects, which anatomically matched the visual field defects. Conclusions: Macular GCA using SD-OCT can be used to complement visual field assessment and brain MRI findings during evaluation of patients with pituitary tumor. New Constitutively Active Phytochromes Exhibit Light-Independent Signaling Activity

<P>Plant phytochromes are photoreceptors that mediate a variety of photomorphogenic responses. There are two spectral photoisomers, the red light-absorbing Pr and far-red light-absorbing Pfr forms, and the photoreversible transformation between the two forms is important for the functioning of phytochromes. In this study, we isolated a Tyr-268-to-Val mutant of Avena sativa phytochrome A (AsYVA) that displayed little photoconversion. Interestingly, transgenic plants of AsYVA showed light-independent phytochrome signaling with a constitutive photomorphogenic (cop) phenotype that is characterized by shortened hypocotyls and open cotyledons in the dark. In addition, the corresponding Tyr-303-to-Val mutant of Arabidopsis (Arabidopsis thaliana) phytochrome B (AtYVB) exhibited nuclear localization and interaction with phytochrome-interacting factor 3 (PIF3) independently of light, conferring a constitutive photomorphogenic development to its transgenic plants, which is comparable to the first constitutively active version of phytochrome B (YHB; Tyr-276-to-His mutant). We also found that chromophore ligation was required for the light-independent interaction of AtYVB with PIF3. Moreover, we demonstrated that AtYVB did not exhibit phytochrome B activity when it was localized in the cytosol by fusion with the nuclear export signal and that AsYVA exhibited the full activity of phytochrome A when localized in the nucleus by fusion with the nuclear localization signal. Furthermore, the corresponding Tyr-269-to-Val mutant of Arabidopsis phytochrome A (AtYVA) exhibited similar cop phenotypes in transgenic plants to AsYVA. Collectively, these results suggest that the conserved Tyr residues in the chromophore-binding pocket play an important role during the Pr-to-Pfr photoconversion of phytochromes, providing new constitutively active alleles of phytochromes by the Tyr-to-Val mutation.</P>
Psychological Aspects of Glaucoma

<P>Glaucoma is a group of optic neuropathies that is more prevalent among the elderly population and commonly associates with comorbidities, including mental disorders in that population. This article reviews the relationship between glaucoma and mental disorders. In it, we discuss the coexistence of glaucoma and mental illnesses, including Alzheimer's disease, depression, and personality disorder. We also focus on the proper treatment approaches for glaucoma patients with mental comorbidity and poor treatment adherence. We summarize some cautiously recommended psychotherapeutic medications, while also discussing the psychologically adverse effects of antiglaucoma medications.</P>

자연정화공법을 이용한 소형 축산폐수처리장치의 최적여재 선정

소규모 축산농가로부터 발생되는 축산폐수를 효과적으로 처리하기 위한 인공습지를 개발하기 위하여 인공습지에서 가장 중요한 인자 중 하나인 최적 여재의 선정에 관하여 연구하였다. 최적여재를 선정하기 위하여 여재를 왕사, 쇄석, 제올라이트 및 방해석으로 달리하여 충진한 후 각 여재별 축산폐수 처리효율 조사한 결과 COD 처리효율은 왕사가 다른 여재에 비하여 높았고, SS 처리효율은 모든 여재에서 94~95%로 큰 차이 없었다. 또한 T-N 처리효율은 제올라이트가 다른여재에 비해 높았고, T-P 처리효율은 방해석이 다른여재에 비해 높았다. 이상의 결과에서 COD,SS, T-N 및 T-P 모두를 안정적으로 처리할 수 있는 최적여재는 왕사였다. 소형 축산폐수처리장치에서 질소와 인 처리효율을 극대화 하기 위해 호기성조 (왕사:쇄석:방해석=3:2:1)와 혐기성조 (왕사:쇄석:제올라이트=3:2:1)에 혼합여재를 주입한 결과 최적 여재인 왕사에 비해 질소 및 인 처리효율이 각각 15% 및 7% 향상되었다. In order to develop livestock wastewater treatment technology by natural purification method, the optimum filter media in small-scale livestock wastewater treatment apparatus for treating livestock wastewater were studied. Removal rates of pollutants under different filter media were in the other of coarse sand ≒ broken stone > zeolite > calcite for COD, zeolite >> broken stone ≒ coarse sand ≒ calcite for T-N, and calcite > coarse sand ≒ broken stone ≒ zeolite for T-P. Based on the above results, the optimum filter media was coarse sand in small-scale livestock wastewater treatment apparatus. To meet acceptable effluent quality standard for livestock wastewater and to improve T-N and T-P removal efficiencies, removal efficiencies of pollutants in small-scale livestock wastewater treatment apparatus with mixed filter media were studied. The removal rates of COD, SS, T-N and T-P in effluent were 84, 94, 65 and 98% in small-scale livestock wastewater treatment apparatus with mixed filter media, respectively. For increasing the T-N and T-P removals in small-scale livestock wastewater treatment apparatus, the mixed filter media are recommended.
