      • Combination of <i>Garcinia cambogia</i> Extract and Pear Pomace Extract Additively Suppresses Adipogenesis and Enhances Lipolysis in 3T3-L1 Cells

        <P><B>Background:</B></P><P>Inhibition of adipogenesis has been a therapeutic target for reducing obesity and obesity-related disorders such as diabetes, hypertension, atherosclerosis, and cancer. For decades, anti-adipogenic potential of many herbal extracts has been investigated. One example is <I>Garcinia cambogia</I> extract (GE) containing (-)-hydroxycitric acid as an active ingredient. GE is currently marketed as a weight loss supplement, used alone or with other ingredients. Pear pomace extract (PE), another natural product, has been also shown to have anti-adipogenic activity in a recent report.</P><P><B>Objective:</B></P><P>It was tested if the mixture of PE and GE (MIX) would produce more effective anti-adipogenic activity than PE or GE alone.</P><P><B>Materials and Methods:</B></P><P>Differentiation of 3T3-L1 preadipocyte was induced by adding insulin, dexamethasone, and isobutylmethylxanthine and lipid accumulation was measured by Oil Red O staining. Cellular markers for adipogenesis and lipolysis such as CCAAT/enhancer binding protein (C/EBP-α), peroxisome proliferator-activated receptor gamma (PPAR-γ), fatty acid synthase (FAS), and hormone-sensitive lipase (HSL) was measured using immunocytochemistry.</P><P><B>Results:</B></P><P>MIX, compared to PE or GE alone, showed greater inhibition of lipid accumulation. Furthermore, MIX reduced the expression of adipogenesis-related factors C/EBP-α, PPAR-γ, and FAS more than PE or GE alone did. In contrast, the expression of HSL the enzyme required for lipolysis was further enhanced in MIX-treated adipocytes compared to the PE or GE alone treated groups.</P><P><B>Conclusions:</B></P><P>Anti-adipogenic effect of PE and GE appears synergistic, and the MIX may be a useful therapeutic combination for the treatment of obesity and obesity-related diseases.</P><P><B>SUMMARY</B></P><P><P>PE and GE efficiently inhibited adipocyte differentiation by suppressing the expression of adipogenic transcription factor CEBP-α and PPAR-γ.</P><P>PE and GE significantly decreased the expression of adipogenic enzyme FAS.</P><P>PE and GE increased the expression of lipid degrading enzyme HSL.</P><P>Mixture of PE and GE exhibited additive or moderately synergistic effect on adipocyte differentiation and lipid accumulation.</P></P><P><B>Abbreviations used:</B> CEBP-a: CCAT/enhancer binding protein alpha, CI: Combination Index, FAS: Fatty acid synthase, GE: Garcinia cambogia extract, HSL: Hormone sensitive lipase, PE: Pear pomace extract, PPAR-γ: Peroxisome proliferator-activated receptor gamma.</P> >[FIG OMISSION]</BR>

        Effect of Feeding Mixture of Soybean Peptides, L-Carnitine and Garcinia Cambogia Extract on Body Weight and Lipid Metabolism in Rats

        This study was performed to investigate effects of the experimental mixture containing soybean peptides, L-carnitine and Garcinia Cambogia extract on body weight and lipid metabolism in rats. Male Sprague-Dawley rats (n=40) of eight weeks old were raised for four weeks with high fat diet (40% fat as calorie) to induce obesity. After induction of obesity, rats were feed control (C) diet, containing either 0.16% (+1D), 1.6% (+10D), 8% (+50D) of experimental mixture for eight weeks. Plasma aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activity and total protein and albumin concentration were not different among groups. The Body weight gain was significantly lower in experimental mixture diet group compared to control group. Weights of perirenal fat pad and epididymal fat pad in the +50D group were significantly lower than those in the +1D and +10D groups. Plasma total lipid and liver total cholesterol levels in the experimental groups were significantly lower than those in the control group. Fecal total lipid and total cholesterol excretions were highest in +50D group. These results suggest that the experimental mixture containing peptides, L-carnitine and Garsinia Canbogia extract is effective for reducing the body weight and adipose tissue weight which may be due to the modulation of lipid metabolism and the increased fecal excretion of lipid.

        비만 피험자에서 블록앤번의 유효성 및 안전성을 평가하기 위한 위약대조 비교 인체시험

        The purpose of the present study was to evaluate the efficacy and safety of Block and burn®, a 4:1 mixture of Phaseolus vulgaris and Garcinia cambogia extracts, in reducing body weight, as the main outcome measure. Secondary measures of the study were body composition change in overweight human subjects. A randomized, double-blinded, placebocontrolled study was conducted on 52 pre-selected, slightly overweight volunteers, whose weight had been essentially stable for at least 2 weeks, The volunteers were divided into two groups, homogeneous for age, gender, and body weight. The test product (1.2 g) containing Phaseolus vulgaris (200 mg) and Garcinia cambogia (50 mg) and the placebo (1.2 g) were taken 6 tablets per day for 56 consecutive days, respectively. Each subject's body weight, body mass index (BMI), body fat, body fat ratio, and waist hip ratio (WHR) were measured. After 56 days (8 weeks), subjects receiving Block and burn ® had significantly greater reduction of body weight. Moreover, a significant changes of the BMI, body fat, and body fat ratio were observed in the test group compared to placebo. However, WHR was not statistically different between groups. These results indicated that Block and burn® produced significant decrements in body weight and suggested decrements in fat mass in the face of maintained lean body mass.
