A cytotoxic sesquiterpene against L1210 and Hl-60 cells was isolated from Angelicae Koreanae Radix (bulk-kang-hwal). The component was identified as bisabolangelone by means of chemical and physical methods. The $ED_{50}$ values of it were $1.20{\;}\mu\textrm{g}/ml$ against L1210 cells and $2.30{\;}\mu\textrm{g}/ml$ against HL-60 cells. Bisabolangelone was found in bulk-kang-hwal but not in kang-hwal.

The total ethanol extract of Angelica koreana radix (UmbeUiferae) showed potent inhibitory effect on tumor necrosis factors (TNF-alpha) production from LPS-stimulated RAW264.7 cells in screening studies on 118 Korean medicinal plants. The activity-guided fractionation of the total ethanol extract resulted in the isolation of 4 compounds, including active substances. The chemical structures of the compounds isolated were established by chemical and spectrometric analyses as imperatorin, isoimperatorin, osthol and oxypeucedanin. Among them, osthol, oxypeucedariin and isoimperatorin showed significant inhibitory activities on TNF-alpha production as 54.3+/-2.1%, 33.6+/-1.0% and 17.7+/-3.0%, respectively, at 12.5mcg/ml.

Prostaglasdins biosynthesis and nitric oxide production have been implicated in the process of inflammation In this study, we investigated on the effects of ethyl acetate extract of Angelica Koreana Radix (EAKR) on the activities of prostaglandin E2 (PGE2), proteoglycan (PG) degradation and nitric oxide synthase (NO) in inflammation cytokines-activated rabbit articular chondrocytes. EAKR exhibited inhibitory activities on NO production and PGE2 production as 73.08% and 89.49%, respectively at 20 ㎍/㎖ and inhibited the degradation of PG in a concentration-dependent manner. Zelatin zymog- raphy analysis demonstrated that EAKR significantly inhibited MMR-2, 9 expression in chondrocytes. In vivo, EAKR was shown to have inhibitory effects on acetic acid-induced pain. This study suggests that modulation of PGE2, NO, PG degradation and MMP-2, 9 by EARK may be important in the prenebtion of inflammation and osteroarthritis.
