High Performance Liquid Chromatography에 의한 Porphyomonas gingivalis W50의 대사산물 분석

Porphyomonas gingioalis(P. gingivalis) has been implicated as a strong pathogen in periodontal disease and short chain fatty acids(SCFA) which present in periodontal pockets have been shown to play a role in periodontal diseases. The pupose of This study is intended to analyse metabolic acids of P. gingivalis and then to detect the presence of SCFA. Metabolic acids extracted from a 48 hours PYG culture and measured by high performance liquid chromatography. These results suggest that the metabolic acids of P. gingivalis W50 are acetic acid, pyruvic acid, lactic acid, fumaric acid, propionic acid, isobutyric acid, butyric acid, hydroxyphenyl acetic acid, isovaleric acid, valerie acid and phenylacetic acid.
배양 시간에 따른 Streptococcus mutans 10449의 성장도

It is reported that the oral bacterium, containing Streptococcus species, continues growing as adjusting to metabolic activity according to the surrounding environments that might be affected by temperature, pH, nutrition, etc. The purpose of this study is to know the growth rate of Streptococcus mutans 10449 according to incubation time. Growth rate of Streptococcus mutans 10449 was measured by spectrophotometer (optical density) and counting CFU(Colony Forming Unit). Streptococcus mutans 10449 increased in number nearly straight from 0 to 3 hours of incubation time. A lag phase was observed from 0 to one hours of incubation time by serial dilution. From 3 to 10 hours, the growth rate of them was fluctuant with a little statistical significance but didn't have a marked variation. They decreased straight from 10 to 15 hours and PABS(Percentage of absorbance) showed no change after 15 hours. These results suggest that growth curve scale of Streptococcus mutans 10449 might have a semilogarismic scale and show a additional stationary phase. Further study is needed to know the growth curve scale of Streptococcus mutans 10449 as various environment change (particularly pH of media, nutrition, temperature etc.).
