Twenty novel tetramic acid derivatives (E)-3-(1-(alkyloxyamino)ethylidene)-1-alkylpyrrolidine-2,4-diones were synthesized by the reaction of 3-(1-hydroxyethylidene)pyrrolidine-2,4-diones with O-alkyl hydroxylamines. The title compounds were confirmed by IR, 1H NMR, MS and elemental analysis. The structure of compound 6r was further verified by X-ray diffraction crystallography. The bioassays showed that most of the title compounds exhibited noticeable herbicidal and fungicidal activities.

Catalase (CAT) is an essential antioxidant in organisms, its can eliminate hydrogen peroxides produced in the cellular environment. Previous studies have focused mainly on the functions of CAT by pathogen challenge, but the functions of CAT by metals stress remain unclear. In order to investigate the function of CAT in Acrossocheilus fasciatus (AfCat), we cloned CAT complete cDNA sequences and analyzed its functions by cadmium (Cd) stress. qPCR analysis illustrated that the expression of CAT changed rapidly and dynamically in response to Cd from 0.05 mg L-1 to 1.25 mg L-1. Enzyme activity assay result showed that CAT activity was up-regulated significantly in response to Cd from 0.05 mg L-1 to 1.25 mg L-1. Histochemistry analysis showed that the structure of liver, testis and kidney were severely damaged by exposed to Cd of 1.25 mg L-1. Collectively, the results of our study indicate that AfCat may play an indispensable role in maintaining redox balance under Cd exposure, but AfCat expression will be inhibited when Cd accumulation reaches to the critical concentration. This study will illuminate the potential functions of AfCat in the defense mechanisms of reproductive and metabolic systems under Cd exposure in A. fasciatus.

A novel anthraquinone diamino-bridged bis($\beta$ -cyclodextrin) 2 was synthesized. The inclusion complexation behaviors of the native $\beta$ -cyclodextrin 1 and the novel bis($\beta$ -cyclodextrin) 2 with guests, such as acridine red (AR), neutral red (NR), ammonium 8-anilino-1-naphthalenesulfonate (ANS), sodium 2-(p-toluidinyl) naphthalenesulfonate (TNS) and rhodamine B (RhB) were investigation by fluorescence, circular dichroism and 2D NMR spectroscopy. The spectral titrations were performed in phosphate buffer (pH 7.20) at 25 ${^{\circ}C}$ to give the complex stability constants (Ks) and Gibbs free energy changes (−${\Delta}G^0$) for the stoichiometric 1:1 inclusion complexation of host 1 and 2 with guests. The results indicated that the novel bis($\beta$ -cyclodextrin) 2 greatly enhanced the original binding affinity of the native $\beta$ -cyclodextrin 1. Typically, bis($\beta$ -cyclodextrin) 2 showed the highest binding constant towards ANS up to 34.8 times higher than that of 1. The 2D NMR spectra of bis($\beta$ -cyclodextrin) 2 with RhB and TNS were performed to confirm the binding mode. The increased binding affinity and molecular selectivity of guests by bis($\beta$ -cyclodextrin) 2 were discussed from the viewpoint of the size/shape-fit concept and multipoint recognition mechanism.
Hydraulic fracture initiation pressure of anisotropic shale gas reservoirs

Shale gas formations exhibit strong mechanical and strength anisotropies. Thus, it is necessary to study the effect of anisotropy on the hydraulic fracture initiation pressure. The calculation model for the in-situ stress of the bedding formation is improved according to the effective stress theory. An analytical model of the stresses around wellbore in shale gas reservoirs, in consideration of stratum dip direction, dip angle, and in-situ stress azimuth, has been built. Besides, this work established a calculation model for the stress around the perforation holes. In combination with the tensile failure criterion, a prediction model for the hydraulic fracture initiation pressure in the shale gas reservoirs is put forward. The error between the prediction result and the measured value for the shale gas reservoir in the southern Sichuan Province is only 3.5%. Specifically, effects of factors including elasticity modulus, Poisson's ratio, in-situ stress ratio, tensile strength, perforation angle (the angle between perforation direction and the maximum principal stress) of anisotropic formations on hydraulic fracture initiation pressure have been investigated. The perforation angle has the largest effect on the fracture initiation pressure, followed by the in-situ stress ratio, ratio of tensile strength to pore pressure, and the anisotropy ratio of elasticity moduli as the last. The effect of the anisotropy ratio of the Poisson's ratio on the fracture initiation pressure can be ignored. This study provides a reference for the hydraulic fracturing design in shale gas wells.
An MRTF-A–ZEB1–IRF9 axis contributes to fibroblast–myofibroblast transition and renal fibrosis

Myofibroblasts, characterized by the expression of the matricellular protein periostin (Postn), mediate the profibrogenic response during tissue repair and remodeling. Previous studies have demonstrated that systemic deficiency in myocardin-related transcription factor A (MRTF-A) attenuates renal fibrosis in mice. In the present study, we investigated the myofibroblast-specific role of MRTF-A in renal fibrosis and the underlying mechanism. We report that myofibroblast-specific deletion of MRTF-A, achieved through crossbreeding Mrtfa-flox mice with Postn-CreERT2 mice, led to amelioration of renal fibrosis. RNA-seq identified zinc finger E-Box binding homeobox 1 (Zeb1) as a downstream target of MRTF-A in renal fibroblasts. MRTF-A interacts with TEA domain transcription factor 1 (TEAD1) to bind to the Zeb1 promoter and activate Zeb1 transcription. Zeb1 knockdown retarded the fibroblast–myofibroblast transition (FMyT) in vitro and dampened renal fibrosis in mice. Transcriptomic assays showed that Zeb1 might contribute to FMyT by repressing the transcription of interferon regulatory factor 9 (IRF9). IRF9 knockdown overcame the effect of Zeb1 depletion and promoted FMyT, whereas IRF9 overexpression antagonized TGF-β-induced FMyT. In conclusion, our data unveil a novel MRTF-A–Zeb1–IRF9 axis that can potentially contribute to fibroblast–myofibroblast transition and renal fibrosis. Screening for small-molecule compounds that target this axis may yield therapeutic options for the mollification of renal fibrosis.

Background: Evidence supporting an association between the 8q24 rs4242382-A polymorphism and prostate cancer (PCa) risk has been reported in North American and Europe populations, though data from Asian populations remain limited. We therefore investigated this association by clinical detection in China, and meta-analysis in Asian, Caucasian and African-American populations. Materials and Methods: Blood samples and clinical information were collected from ethnically Chinese men from Northern China with histologically-confirmed PCa (n=335) and from age-matched normal controls (n=347). The 8q24 (rs4242382) gene polymorphism was genotyped by polymerase chain reaction-high-resolution melting analysis. We initially analyzed the associations between the risk allele and PCa and clinical covariates. A meta-analysis was then performed using genotyping data from a total of 1,793 PCa cases and 1,864 controls from our study and previously published studies in American and European populations, to determine the association between PCa and risk genotype. Results: The incidence of the risk allele was higher in PCa cases than controls (0.222 vs 0.140, $P=7.3{\times}10^{-5}$), suggesting that the 8q24 rs4242382-A polymorphism was associated with PCa risk in Chinese men. The genotypes in subjects were in accordance with a dominant genetic model (ORadj=2.03, 95%CI: 1.42-2.91, $Padj=1.1{\times}10^{-4}$). Presence of the risk allele rs4242382-A at 8q24 was also associated with clinical covariates including age at diagnosis ${\geq}65$ years, prostate specific antigen >10 ng/ml, Gleason score <8, tumor stage and aggressive PCa, compared with the non-risk genotype ($P=4.6{\times}10^{-5}-3.0{\times}10^{-2}$). Meta-analysis confirmed the association between 8q24 rs4242382-A polymorphism and PCa risk (OR=1.62, 95%CI: 1.39-1.88, $P=1.0{\times}10^{-5}$) across Asian, Caucasian and African American populations. Conclusions: The replicated data suggest that the 8q24 rs4242382-A variation might be associated with increased PCa susceptibility in Asian, Caucasian and African American populations. These results imply that this polymorphism may be a useful risk biomarker for PCa in multi-ethnic populations.
Effect of Nd Substitution for Mischmetal on Magnetic Properties in (MM,Nd)-Fe-B Ribbons

Mischmetal-based permanent magnets of (MM,Nd)12Fe82B6 were prepared by melt-spinning method via varying the relative content of mischmetal and Nd. The coercivity is low, and the squareness of hysteresis loop is poor in MM12Fe82B6 ribbons. The x-ray diffraction pattern, maximum of δm value as well as the temperature dependence of magnetization indicates that the partial substitution of Nd for MM could improve the crystallinity in the mischmetal-based magnets, and both the coercivity and squareness of hysteresis loop increase with Nd substitution for mischmetal. The increase of squareness should be partially attributed to the improvement in the crystallinity of R₂Fe14B crystal phase. For more than 4 at.% Nd substitution the coercivity is not less than that in corresponding (Ce,Nd)-based magnets, and the coercivity of 7.81 kOe and the maximum energy product of 15.41 MGOe were obtained in MM8Nd₄Fe82B6 ribbons. These investigations show that it is reasonable to use the low cost mischmetal to prepare the resource-saving rare-earth magnets, and that optimizing the addition amount of Nd is necessary to improve the crystallinity and enhance the magnetic properties.
