        Insights Into the BR2/PGP1-mediated Patterns for Shoot and Root Growth in Maize Early Seedling Development by Comparative Transcriptome Sequencing

        Plant shoot and root growth is regulated by multiplefactors, including the hormone auxin. A maize brachytic2(br2) mutant displays a shortened shoot and root with highauxin concentration in their apexes, where BR2 encodes anauxin transporter P-glycoprotein1 (PGP1). We employedcomparative transcriptome sequencing to explore the BR2/PGP1-mediated patterns for shoot and root growth amongbr2, wild type (WT), and hybrid F1 (br2 × WT). The resultsshow 352 genes in the br2 shoot and 234 genes in the br2root are differentially expressed compared to WT and F1. Zm00001d045203 and Zm00001d031562 were identified astwo auxin-responsive genes in both the shoot and root. TheZm00001d045203 gene is down-regulated in br2 and encodesan Aux/IAA inhibitor involved in the SCFTIR1/AFB-mediatedauxin signaling pathway. The up-regulated Zm00001d031562gene encodes a small GTPase belonging to the Rab/Yptfamily, which takes part in auxin-mediated responses inArabidopsis. Cytology analysis reveals that reduced cellelongation affects the shoot and root growth of br2. TheZm00001d043477 gene is down-regulated and encodes amember of the cellulose synthase protein family, possiblyreducing cell elongation in the br2 shoot and root. Theseresults reveal the specific regulation of auxin actions and cellresponses by BR2/PGP1.

        Flutter performance of box girders with different wind fairings at large angles of attack

        The streamlined box is a common type of girders for long-span suspension bridges. Spanning deep canyons, long-span bridges are frequently attacked by strong winds with large angles of attack. In this situation, the flow field around the streamlined box changes significantly, leading to reduction of the flutter performance. The wind fairings have different effects on the flutter performance. Therefore, this study examines the flutter performance of box girders with different wind fairings at large angles of attack. Computational fluid dynamics (CFD) simulations were carried out to extract the flutter derivatives, and the critical flutter state of a long-span bridge was determined. Further comparisons of the wind fairings were investigated by a rapid method which is related to the input energy by the aerodynamic force. The results show that a reasonable type of wind fairings could improve the flutter performance of long-span bridges at large angles of attack. For the torsional flutter instability, the wind fairings weaken the adverse effect of the vortex attaching to the girder, and a sharper one could achieve a better result. According to the input energies on the girder with different wind fairings, the symmetrical wind fairings are more beneficial to the flutter performance.

        Modelling and Optimization of a Magnetic Spring Based Electromagnetic Vibration Energy Harvester

        This paper presents the development of an AA battery size electromagnetic vibration energy harvester with an aim to maximize the output power density. A tube shape and stacked opposing permanent magnets with magnetic spring were used to suit the shape constraint as well as to achieve high fl ux linkages. An initial prototype of electromagnetic vibration harvester with AA battery size was built and tested on a controllable shaker to obtain its output voltage and power level at diff erent frequencies for fi xed accelerations. A single magnet was fi xed at the bottom of the harvester to provide levitation force in this development in order to lower the resonant frequency. A special time-domain based analytical model was also developed using both Finite Element Analysis and Simulink simulation. The time-domain analytical model is easier to implement than other frequency domain based analytical models which generally applied in literatures for modelling of the electromagnetic vibration energy harvesters. The analytical model was verifi ed by the measured results obtained from the initial prototype. The validated analytical model was successfully applied to optimize the harvester. Two more generator prototypes were further built and tested after the optimization study. The optimized harvester using three stacked opposing permanent magnets could achieve a normalized power density of 12,655 μWcm −3 g −2 at 9.9 Hz frequency with 0.22 g acceleration, which is signifi cantly higher than other reported electromagnetic vibration energy harvesters

        Blockage effects on aerodynamics and flutter performance of a streamlined box girder

        Wind tunnel test is one of the most important means to study the flutter performance of bridges, but there are blockage effects in flutter test due to the size limitation of the wind tunnel. On the other hand, the size of computational domain can be defined by users in the numerical simulation. This paper presents a study on blockage effects of a simplified box girder by computation fluid dynamics (CFD) simulation, the blockage effects on the aerodynamic characteristics and flutter performance of a long-span suspension bridge are studied. The results show that the aerodynamic coefficients and the absolute value of mean pressure coefficient increase with the increase of the blockage ratio. And the aerodynamic coefficients can be corrected by the mean wind speed in the plane of leading edge of model. At each angle of attack, the critical flutter wind speed decreases as the blockage ratio increases, but the difference is that bending-torsion coupled flutter and torsional flutter occur at lower and larger angles of attack respectively. Finally, the correction formula of critical wind speed at 0o angle of attack is given, which can provide reference for wind resistance design of streamlined box girders in practical engineering.

        Synthesis and Application of Phosphorus-containing Flame Retardant Plasticizer for Polyvinyl Chloride

        A novel phosphorus-containing flame retardant plasticizer (PFRP) derived from castor oil acid methyl ester (COME) was synthesized to substitute dioctyl phthalate (DOP) for plasticizing polyvinyl chloride (PVC) products. The chemical structures of PFRP were confirmed by fourier transform infrared spectroscopy (FTIR) and 1H nuclear magnetic resonance (1H NMR). Meanwhile, the plasticizing effect, flammability and thermal stability of plasticized PVC films were investigated by dynamic mechanical analyzer (DMA), limiting oxygen index (LOI) test, scanning electron microscope (SEM) and thermogravimetric analysis (TGA). As the PFRP content increasing from 0 wt% to 50 wt% amount of plasticizers, the plasticizing efficiency and the mechanical properties showed a slightly decreasing tendency compared with that of DOP, while the LOI value of plasticized PVC increased remarkably from 21.5 % to 25.2 %, showing a combined plasticizing efficiency and flame retardancy. SEM and TGA analysis indicated that PFRP had little effect on thermal stability but was effective to promote the formation of compact carbon residue.

        Subproteomic profiling from renal cortices in OLETF rats reveals mutations of multiple novel genes in diabetic nephropathy

        Background: Ovarian cancer is a common gynecological malignancy among female patients and poses a serious threat to women's health. Although it has been established that Fos-like antigen 2 (FOSL2) is linked to ovarian cancer (OC), its exact role in the development of OC remains unknown. Objective: This article aims to investigate the role of FOSL2 in ovarian cancer development. Methods: FOSL2 expression in ovarian carcinoma and adjacent tissues was assessed using real-time fluorescent quantitative PCR and western blot. We constructed OE/sh-FOSL2 plasmids and Caspase-1 specific inhibitors (Yvad-CMK) and transfected A 2780 cells with them to identify the relevant cell functions. Furthermore, we used western blot assay to determine the changes in expression of apoptosis-associated speck-like protein containing a CARD (ASC), cysteine aspartate-specific proteasezymogen procaspase 1 (pro-caspase-1), cysteinyl aspartate-specific proteinase-1 (caspase-1), interleukin-1β precursor (pro-IL-1β), interleukin-1β (IL-1β), interleukin-18 precursor (pro-IL-18), and interleukin-18 (IL-18). In addition, we measured the concentration of IL-1β and IL-18 using an enzyme-linked immunosorbent assay (ELISA). Moreover, Tthe level of lactate dehydrogenase (LDH) in the cell supernatant was measured by LDH release assay kit. Results: The expression of FOSL2 was significantly higher compared with the surrounding tissues. The proliferation, migration, and invasion of A2780 cells were enhanced after transfection with OE-FOSL2 plasmids; however, the cell apoptosis was significantly decreased. When FOSL2 was overexpressed, the inflammasome-associated proteins such as ASC, caspase-1, IL-1β, and IL-18 were downregulated. Furthermore, FOSL2 induced apoptosis and activated the production of inflammasomes in A2780 cells. Co-therapy with Yvad-CMK and substantially inhibited apoptosis and activation of inflammasomes. Conclusions: Inhibition of FOSL2 promotes the apoptosis of OC cells by mediating the formation of an inflammasome.
