Upregulation of Mir-34a in AGS Gastric Cancer Cells by a PLGA-PEG-PLGA Chrysin Nano Formulation

Background: Nano-therapy has the potential to revolutionize cancer therapy. Chrysin, a natural flavonoid, was recently recognized as having important biological roles in chemical defenses and nitrogen fixation, with anti-inflammatory and anti-oxidant effects but the poor water solubility of flavonoids limitstheir bioavailability and biomedical applications. Objective: Chrysin loaded PLGA-PEG-PLGA was assessed for improvement of solubility, drug tolerance and adverse effects and accumulation in a gastric cancer cell line (AGS). Materials and Methods: Chrysin loaded PLGA-PEG copolymers were prepared using the double emulsion method (W/O/W). The morphology and size distributions of the prepared PLGA-PEG nanospheres were investigated by 1H NMR, FT-IR and SEM. The in vitro cytotoxicity of pure and nano-chrysin was tested by MTT assay and miR-34a was measured by real-time PCR. Results: 1H NMR, FT-IR and SEM confirmed the PLGA-PEG structure and chrysin loaded on nanoparticles. The MTT results for different concentrations of chrysin at different times for the treatment of AGS cell line showed IC50 values of 68.2, 56.2 and $42.3{\mu}M$ and 58.2, 44.2, $36.8{\mu}M$ after 24, 48, and 72 hours of treatment, respectively for chrysin itslef and chrysin-loaded nanoparticles. The results of real time PCR showed that expression of miR-34a was upregulated to a greater extent via nano chrysin rather than free chrysin. Conclusions: Our study demonstrates chrysin loaded PLGA-PEG promises a natural and efficient system for anticancer drug delivery to fight gastric cancer.
Curcumin and Silibinin Inhibit Telomerase Expression in T47D Human Breast Cancer Cells

Background: Telomerase has been considered as an attractive molecular target for breast cancer therapy. The main objective of this work is to assess the inhibitory effects of silibinin and curcumin, two herbal substances, on telomerase gene expression in breast cancer cells. Materials and Methods: For determination of cell viability tetrazolium-based assays were conducted after 24, 48, and 72 h exposure times and expression of human telomerase reverse transcriptase gene was measured with real-time PCR. Results: Each compound exerted cytotoxic effects on T47D cells and inhibited telomerase gene expression, both in a time-and dose-dependent manner. The mixture of curcumin and silibinin showed relatively more inhibitory effect on growth of T47D cells and hTERT gene expression as compared with either agent alone. Conclusions: These findings suggest that cell viability along with hTERT gene expression in breast cancer cells could be reduced by curcumin and silibinin.

Background: Herbal compounds such as curcumin which decrease telomerase and gene expression have been considered as beneficial tools for lung cancer treatment. In this article, we compared the effects of pure curcumin and curcumin-loaded NIPAAm-MAA nanoparticles on telomerase and PinX1 gene expression in a lung cancer cell line. Materials and Methods: A tetrazolium-based assay was used for determination of cytotoxic effects of curcumin on the Calu-6 lung cancer cell line and telomerase and pinX1 gene expression was measured with real-time PCR. Results: MTT assay showed that Curcumin-loaded NIPAAm-MAA inhibited the growth of the Calu-6 lung cancer cell line in a time and dose-dependent manner. Our q-PCR results showed that the expression of telomerase gene was effectively reduced as the concentration of curcumin-loaded NIPAAm-MAA increased while expression of the PinX1 gene became elevated. Conclusions: The results showed that curcumin-loaded-NIPAAm-MAA exerted cytotoxic effects on the Calu-6 cell line through down-regulation of telomerase and stimulation of pinX1 gene expression. NIPPAm-MAA could be good carrier for such kinds of hydrophobic agent.
Corms as a Source of Explants for the Successful Clonal Propagation of $Crocus$ $cancellatus$

The genus $Crocus$ comprises plants with a potential to be developed as a new ornamental crop but to date, there are not many reports on $in$ $vitro$ propagation of many members of this genus. The present study involves $in$ $vitro$ propagation of $Crocus$ $cancellatus$ with ornamental and horticultural value. Two different types of corm explants (apical and basal halves of corms) were cultivated onto Murashige and Skoog's (MS) medium supplemented with different levels of ${\alpha}$-naphthalene acetic acid (NAA) and 6-benzylaminopurine (BAP). One to five cormlets emerged from every responding explant through direct organogenesis. Apical halves of corms were more highly responsive than basal halves and produced a maximum multiplication rate with $3.45{\pm}0.06$ cormlets per explant in $95.33{\pm}2.33%$ of the explants in MS medium supplemented with 3% sucrose and $2mg\;L^{-1}$ NAA and $1mg\;L^{-1}$ BAP. The effect of cold storage temperature on in vitro cormlets sprouting was studied. Cormlets stored at $4^{\circ}C$ for 8 weeks had more statistically significant positive effects on cormlets sprouting from the controls. $In$ $vitro$ rooting of cormlets was induced on MS medium without plant hormones.

The cotton mealybug Phenacoccus solenopsis Tinsley (Hemiptera: Pseudococcidae) is one of the polyphagous and destructive pests in the world. It has caused a lot of damage in cotton fields and settles on other host plants, especially Chinese hibiscus shrubs Hibiscus rosa-sinensis L. and causes irrecoverable damages to these ornamental plants and urban landscape. Midge fly Dicrodiplosis manihoti Harris (Dip.: Cecidomiidae) is one of the most common fly species which was collected in mealybugs colonies. In this study, the biology and age-stage, two-sex life table parameters of Dicrodiplosis manihoti, was evaluated at four constant temperatures (22, 27, 32 and 37 ± 1 ◦ C, 65 ± 5 % RH and a photoperiod of 16: 8 (L: D) h) in the laboratory conditions. The developmental pre-adult period was found as 36.12 ± 0.38, 24.09 ± 0.41, 14.96 ± 0.34 and 12.68 ± 0.26 days and the average egg produced by females was 23.65 ± 2.86, 11.14 ± 2.09, 12.32 ± 3.94 and 8.57 ± 0.67 at those temperatures, respectively which decreased as the temperature increased from 22 to 37 ◦ C, respectively. The population intrinsic rate of increase (r m ) was 0.0247 ± 0.0051, 0.0291 ± 0.0099, 0.0374 ± 0.0206 and 0.0103 ± 0.0129 (d -1 ), and the population finite rate of increase (λ) for the above mentioned temperatures was 1.0249 ± 0.0052, 1.0295 ± 0.0101, 1.0384 ± 0.0212 and 1.0104 ± 0.0130 (d -1 ), respectively. Also, the net reproductive rate (R 0 ) at the above mentioned temperatures was 2.62 ± 0.5, 2.19 ± 0.53, 2.05 ± 0.73 and 1.18 ± 0.22 offspring respectively. The shortest (T = 14.88 ± 0.45 days) and the longest (T = 38.28 ± 0.65 days) mean generation time was recorded at 37 and 22 ◦ C, respectively. According to the results of this study D. manihoti has a good potential to control of Ph. solenopsis and the temperature of around 32 ◦ C is the best for mass rearing this predator.
Sustained release coating of ibuprofen pellets at Wurster fluidization: statistical approach

The Wurster fluidization coating is recognized in many pharmaceutical industries for its precise film coating onto particulate materials such as pellets, granules, and powders. The current work focused on some process variables to study sustained release behavior of ibuprofen pellets at a lab-scale Wurster fluidized bed coater. Applying design of experiment, the effect of mean pellets size (dp), air flow rate (Qa), coating solution flow rate (Qs), and drying time duration (td) were investigated statistically on the mean dissolution time (MDT) of ibuprofen pellets. Results in analysis of variance illustrated that a decrease in dp make rising of MDT due to decreasing the ibuprofen diffusivity in the media owing to trapping role of ethyl cellulose. It was also found increasing of Qs resulted in extending of MDT due as the coating solution deposited on the pellets surface was increased. While the td did not play an important role on the MDT, the MDT was increased by Qa because of pellets circulation within the bed.

Wetted rice particles were dried in a fluidized bed and the corresponding passive acoustic emissions signals (AES) were recorded at a given frequency to study the drying phenomena and bed hydrodynamic changes as well. The results show that the end time of the constant rate zone and the end of the falling rate can be determined from the variation of standard deviation and kurtosis of AES, respectively. Frequency domain analysis was also used to quantify the moisture content of solids. For this end, the original signal was decomposed into ten sub-signals, and it was found that the energy of the 4th sub-signal can be correlated with the moisture content. The results show that the acoustic emission measurement is applicable as a practical method for on-line condition monitoring of drying process in fluidized bed dryers.
