        A New Large-Scale Plane-Strain Permeameter for Gravelly Clay Soil under Stresses

        A large-scale plane-strain permeameter was designed to investigate the hydraulic characteristics of gravelly clay base soil and a filter under various stresses and different water heads. The apparatus provides a specimen size about 80 cm long, 40 cm wide and 40cm high. It is equipped with a horizontal and vertical loading system with capacities of 1000 kN in the horizontal direction and 2000kN in the vertical direction and a water pressure system which can provide water head up to 300 m. The components of the planestrain permeameter are described in detail in this paper. The performance of the permeameter is demonstrated through a group of tests on specimens combining gravelly clay soil and a filter.

        Effects of a traditional Chinese medicine formula and its extraction on muscle fiber characteristics in finishing pigs, porcine cell proliferation and isoforms of myosin heavy chain gene expression in myocytes

        Objective: This study evaluated the effects of a traditional Chinese medicine formula (TCMF) on muscle fiber characteristics in finishing pigs and the effects of the formula's extract (distilled water, ethyl acetate and petroleum ether extraction) on porcine cell proliferation and isoforms of myosin heavy chain (MyHC) gene expression in myocytes. Methods: In a completely randomized design, ninety pigs were assigned to three diets with five replications per treatment and six pigs per pen. The diets included the basal diet (control group), TCMF1 (basal diet+2.5 g/kg TCMF) and TCMF2 (basal diet+5 g/kg TCMF). The psoas major muscle was obtained from pigs at the end of the experiment. Muscle fiber characteristics in the psoas major muscle were analyzed using myosin ATPase staining. Cell proliferation was measured using 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) dye and cytometry. Isoforms of MyHC gene expression were detected by real-time quantitative polymerase chain reaction. Results: The final body weight and carcass weight of finishing pigs were increased by TCMF1 (p<0.05), while the psoas major muscle cross-sectional area was increased by TCMF (p<0.05). The cross-sectional area and diameter of psoas major muscle fiber Ι, IIA, and IIB were increased by TCMF2 (p<0.05). The cross-sectional area and fiber diameter of psoas major muscle fiber IIA and IIB were increased by diet supplementation with TCMF1 (p<0.05). Psoas major muscle fiber IIA and IIB fiber density from the pigs fed the TCMF1 diet and the type IIB fiber density from the pigs fed the TCMF2 diet were lower compared to pigs fed the control diet (p<0.05). Pigs fed TCMF2 had a higher composition of type Ι fiber and a lower percentage of type IIB fiber in the psoas major muscle (p<0.05). The expression levels of MyHC Ι, MyHC IIa, and MyHC IIx mRNA increased and the amount of MyHC IIb mRNA decreased in the psoas major muscle from TCMF2, whereas MyHC Ι and MyHC IIx mRNA increased in the psoas major muscle from TCMF1 (p<0.05). Peroxisome proliferator-activated receptor ${\gamma}$ $coactivator-1{\alpha}$ and CaN mRNA expression in the psoas major muscle were up-regulated by TCMF (p<0.05). Porcine skeletal muscle satellite cell proliferation was promoted by $4{\mu}g/mL$ and $20{\mu}g/mL$ TCMF water extraction (p<0.05). Both $1{\mu}g/mL$ and $5{\mu}g/mL$ of TCMF water extraction increased MyHC IIa, MyHC IIb, and MyHC IIx mRNA expression in porcine myocytes (p<0.05), while MyHC Ι mRNA expression in porcine myocytes was decreased by $5{\mu}g/mL$ TCMF water extraction (p<0.05). Porcine myocyte MyHC Ι and MyHC IIx mRNA expression were increased, and MyHC IIa and MyHC IIb mRNA expression were down-regulated by $5{\mu}g/mL$ TCMF ethyl acetate extraction (p<0.05). MyHC Ι and MyHC IIa mRNA expression in porcine myocytes were increased, and the MyHC IIb mRNA expression was decreased by $1{\mu}g/mL$ TCMF ethyl acetate extraction (p<0.05). Four isoforms of MyHC mRNA expression in porcine myocytes were reduced by $5{\mu}g/mL$ TCMF petroleum ether extraction (p<0.05). MyHC IIa mRNA expression in porcine myocytes increased and MyHC IIb mRNA expression decreased by $1{\mu}g/mL$ in a TCMF petroleum ether extraction (p<0.05). Conclusion: These results indicated that TCMF amplified the psoas major muscle cross-sectional area through changing muscle fiber characteristics in finishing pigs. This effect was confirmed as TCMF extraction promoted porcine cell proliferation and affected isoforms of MyHC gene expression in myocytes.

        Effect of sputtering pressure on the properties of large area IWO thin films deposited by direct current magnetron sputtering

        Tungsten doped indium oxide (In2O3: W, IWO) thin films have been attracting increasing attention due to their excellent optoelectronic properties. Here, a series of IWO thin films were prepared using direct current (DC) magnetron sputtering method, by varying the sputtering pressure. Analysis revealed that the IWO films prepared under sputtering pressure of 0.4 Pa exhibited excellent optoelectronic performance, with low square resistance, resistivity, high carrier concentration and mobility. The resulting semi-transparent perovskite solar cells (ST-PSCs), with IWO fabricated under 0.4 Pa, yield a PCE of 15.71 % for the large area modules of 100 cm2 (active area 64.8 cm2).

        Effects of boundary layer and liquid viscosity and compressible air on sloshing characteristics

        In this paper, numerical investigations for tank sloshing, based on commercial CFD package FLUENT, are performed to study effects of boundary layer grid, liquid viscosity and compressible air on sloshing pressure, wave height and rising time of impact pressure. Also, sloshing experiments for liquids of different viscosity are carried out to validate the numerical results. Through comparison of numerical and experimental results, a computational model including boundary layer grid can predict the sloshing pressure more accurately. Energy dissipation due to viscous friction leads to reduction of sloshing pressure and wave elevation. Sloshing pressure is also reduced because of cushion effect of compressible air. Due to high viscosity damping effect and compressible air effect, the rising time of impact pressure becomes longer. It is also found that liquid viscosity and compressible air influence distribution of dynamic pressure along the vertical tank wall.

        Anatase Phase TiO2 Anode of a Dye-Sensitized Solar Cell Prepared by using RMFMS

        Anatase phase TiO2 films were deposited by using reactive medium frequency magnetron sputtering (RMFMS) technology under the conditions of a pure 99.9 % Ti disk as a target in an Ar+O2 atmosphere without additional external heating. The effect of the total pressure and bias on the phase composition, grain sizes and morphologies of the TiO2 films were characterized by using X-ray diffraction (XRD) and scanning electron microscopy (SEM). The results showed that the crystalline anatase phase could be obtained under a total pressure of 0.4 5 Pa. The grain size of the anatase TiO2 was between 24 37 nm at a bias of 0 −200 V. Preferred (101) growth was found to occur at total pressures of below 1 Pa. We also discovered the existence of many holes distributed within the TiO2 films.       • Melatonin inhibits the Migration of Colon Cancer RKO cells by Down-regulating Myosin Light Chain Kinase Expression through Cross-talk with p38 MAPK

        Background: Melatonin, which is mainly produced by the pineal gland, has a good inhibitory effect on cell growth of multiple cancer types. However, the underlying molecular mechanisms of anti-tumor activity for colon cancer have not been fully elucidated. In this study, we investigated the effects of melatonin on migration in human colon cancer RKO cells and the potential molecular mechanisms. Materials and Methods: The viability of RKO cells was investigated by MTT assay after treatment with melatonin, SB203580 (p38 inhibitor) and phorbol 12-myristate 13-acetate (PMA, MAPK activator) alone or in combination for 48h. The effects of melatonin, and ML-7, a selective inhibitor of myosin light chain kinase (MLCK), and SB203580, and PMA on the migration of RKO cells were analyzed by in vitro scratch-wound assay. The relative mRNA levels of MLCK was assessed by real-time quantitative RT-PCR. Western blotting analysis was performed to examine the expression of MLCK, phosphorylation of myosin light chain (pMLC) and p38 (pp38). Results: The proliferation and migration of human colon cancer RKO cells were inhibited significantly after treatment with melatonin. The expression levels of MLCK and phosphorylation of MLC of RKO cells were reduced, and real-time quantitative RT-PCR showed that melatonin had significant effects on suppressing the expression of MLCK. Furthermore, the phosphorylation level of p38, which showed the same trend, was also reduced when cells were treated by melatonin. In addition, ML-7 (25umol/l) could down-regulate the phosphorylation of p38. Conclusions: Melatonin could inhibit the proliferation and migration of RKO cells, and further experiments confirmed that p38 MAPK plays an important role in regulating melatonin-induced migration inhibition through down-regulating the expression and activity of MLCK.

        Modelling the Construction of a High Embankment Dam

        Appropriate layer quantity has been considered to model the sequential construction of a high embankment dam over 300 m and its effect on the predicted settlement of the 314 m high Shuangjiangkou dam has been discussed. The simulation results have demonstrated that at least 25 layers are required to accurately model the stage construction of a high embankment dam over 300 m. The stress and deformation within the dam and the foundation during the dam construction and reservoir filling have been simulated using finite element analysis. Saturated-unsaturated seepage theory is used to analyse the transient seepage field in the dam and in the foundation. Two different cases about the construction and the operation are modelled. One involves gradual reservoir filling after the completion of sequential construction, whereas the other involves sequential construction and reservoir impounding by several interleaved stages. The simulation results for both cases have been compared and discussed.

      • TERT rs2736098 Polymorphism and Cancer Risk: Results of a Meta-analysis

        Objective: Several studies have demonstrated associations between the TERT rs2736098 single nucleotide polymorphisms (SNPs) and susceptibility to cancer development. However, there are conflicting results. A systematic meta-analysis was therefore performed to establish the cancer risk associated with the polymorphism. Methods: In this meta-analysis, a total of 6 case-control studies, including 5,567 cases and 6,191 controls, were included. Crude odds ratios with 95% confidence intervals were used to assess the strength of associations in several genetic models. Results: Our results showed no association reaching the level of statistical significance for overall risk. Interestingly, in the stratified analyses (subdivided by ethnicity), significantly increased risks were found in the Asian subgroup which indicates the TERT rs2736098 polymorphism may have controversial involvement in cancer susceptibility. Conclusions: Overall, this meta-analysis indicates that the TERT rs2736098 polymorphism may have little involvement in cancer susceptibility.
