Over the last decade, comprehensive genome-wide sequencing studies have enabled us to find out unexpected genetic alterations of metabolism in cancer. An example is the identification of arginine missense mutations of isocitrate dehydrogenases-1 and -2 (IDH1/2) in glioma, acute myeloid leukemia (AML), chondrosarcomas, and cholangiocarcinoma. These alterations are closely associated with the production of a new stereospecific metabolite, (R)-2-hydroxyglutarate (R-2HG). A large number of follow-up studies have been performed to address the molecular mechanisms of IDH1/2 mutations underlying how these events contribute to malignant transformation. In the meanwhile, the development of selective mutant IDH1/2 chemical inhibitors is being actively pursued in the scientific community and pharmaceutical industry. The present review article briefly discusses the important findings that highlight the molecular mechanisms of IDH1/2 mutations in cancer and provides a current status for development of selective mutant IDH1/2 chemical inhibitors. The unequivocal solid-state structure and stereochemistry of the hetaryl leuco-TAM dye, 2,2’-(2-phenyl propane- 1,3-diylidene) bis(1,3,3- trimethylindoline) derivatives were established using X-ray single crystal analysis. The X-ray crystal analysis showed that the (Z, E)-isomers only formed stereoselectively, with a so-called “threebladed propeller” conformation, from the reaction of a Fischer base and benzaldehyde derivatives. These isomers were stacked in a juxtaposition to form a dimer or a double dimer, adopting either a triclinic, with P-1, or monoclinic crystal system with a space group P21/n in the unit cell of the crystal.
Regulation of Nrf2-Mediated Phase II Detoxification and Anti-oxidant Genes

The molecular mechanisms by which a variety of naturally-occurring dietary compounds exert chemopreventive effects have been a subject of intense scientifl c investigations. Induction of phase II detoxifl cation and anti-oxidant enzymes through activation of Nrf2/ ARE-dependent gene is recognized as one of the major cellular defense mechanisms against oxidative or xenobiotic stresses and currently represents a critical chemopreventive mechanism of action. In the present review, the functional signifl cance of Keap1/ Nrf2 protein module in regulating ARE-dependent phase II detoxifl cation and anti-oxidant gene expression is discussed. The biochemical mechanisms underlying the phosphorylation and expression of Keap1/Nrf2 proteins that are controlled by the intracellular signaling kinases and ubiquitin-mediated E3 ligase system as well as control of nucleocytoplasmic translocation of Nrf2 by its innate nuclear export signal (NES) are described.

The position of the thermodynamic equilibrium for reverse thermochromic spiropyran 6-carboxylates (SP-COOHs) was easily determined in aqueous binary mixtures, such as water-methanol, water-acetonitrile and water-dimethyl sulfoxide. The existence of more than one type of interconvertible species of the ring-opened form of SP-COOH in aqueous binary solvents enables us to evaluate the molar extinction coefficients of the ring-opened species of SP-COOH and to obtain the thermodynamic parameters.

Background: Intravenous hypnotics are used in pregnancy, labor and delivery. The aim of the present study was to investigate and compare the relaxant effects of propofol, thiopental, ketamine, and etomidate on isolated rat uterine smooth muscles. Methods: Uterine smooth muscle preparations were obtained from non-pregnant female rats. The uterus of the rat was dissected and cut into 10 mm strips. The muscle strips were bathed in Krebs solution. After spontaneous uterine contractile activity had been accomplished, propofol, ketamine, thiopental, and etomidate in various concentrations were added cumulatively to the baths and resting tension, active tension, and frequency of contration were recorded at each concentration of agents. EC5, EC25, EC50, EC75, and EC95 of each drug on active tension and frequency of contraction were calculated using a probit model. Results: Propofol, thiopental, and etomidate reduced uterine contractions in a concentration-dependent manner. Ketamine concentrations of 10(-7) to 10(-5) M augmented uterine contractions but ketamine concentrations of 10(-4) to 10(-3) M attenuated uterine contractions. The EC50`s of propofol, thiopental, ketamine, and etomidate on active tension were 1.56 × 10(-5) M, 4.97 × 10(-5) M, 3.52 × 10(-4) M, and 2.73 × 10(-5) M, respectively. Conclusions: All four intravenous hypnotics relaxed the uterine smooth muscle of rats except for ketamine in low concentrations (10(-7) to 10(-5) M). Propofol had the greatest relaxant effects on isolated rat uterine smooth muscle among these hypnotics. It seems that ketamine is a suitable obstetric hypnotic agent for hypovolemic parturients and propofol is a useful hypnotic agent for uterine relaxation during pregnancy. 일본 사회복지시설의 건축경향 및 편의시설 설치특성에 관한 연구

The purpose of this study is seeking plan and maintenance direction of the rehabilitation and day care center for the disabled in domestic that is paid much social attention these days. To achieve the result, this study is shown plans analysis, spatial composition, main concept and convenience facilities's form of the Japan's social welfare organization. The results of the study are as bellow : First, convenience facilities for the elderly and disabled should be considered of installation courtyard for the active introduction external environment into the welfare facilities. Second, construction interaction space where can socialize with not only for the elderly and disabled but also local residences. Finally, making system that users can participate planning stage of welfare facilities. 본 연구는 최근, 국내에서 장애인 및 고령자의 자립생활과 사회통합의 중요성이 부각되면서 사회적으로 많은 관심을 받고 있는 복지관과 자립생활센터, 주간보호시설 등, 장애인과 고령자가 주로 이용하는 복지시설의 계획 및 정비방향을 살펴보고자 한다. 이를 위하여 ‘고령자 거주안정법’과 ‘고령자, 장애인 등의 이동 등의 원활화 촉진에 관한 법률’(배리어 프리(Barrier-free) 신법)과 같은 법적 기준에 의하여 건축된 일본의 사회복지시설의 공간구성 및 편의시설의 실태를 살펴봄으로써 앞으로의 국내 복지시설의 계획과제와 방향성을 제안하고자 한다. 연구대상은 동경도와 요코하마시, 오오사카시에 건축된 8개의 복지시설을 대상으로 주민이용 실태와 공간구성, 설계의 주요개념, 편의시설의 설치실태를 분석하였다. 연구결과, 복지시설이 주이용자인 장애인과 고령자만을 위한 시설이 아닌 지역주민과 어울릴 수 있는 교류의 장소이며 이를 위한 시설의 복합화와 교류공간의 확보, 시설내에 자연환경을 적극적으로 도입하기 위한 중정의 설치, 시설내부의 편의시설 정비뿐만 아니라 교통수단, 보행로 등의 외부공간과 연계한 안전하고 편리한 보행공간의 확보로 요약할 수 있다.
