Quiescin Sulfhydryl Oxidase 1 (QSOX1) Secreted by Lung Cancer Cells Promotes Cancer Metastasis

<P>As lung cancer shows the highest mortality in cancer-related death, serum biomarkers are demanded for lung cancer diagnosis and its treatment. To discover lung cancer protein biomarkers, secreted proteins from primary cultured lung cancer and adjacent normal tissues from patients were subjected to LC/MS–MS proteomic analysis. Quiescin sulfhydryl oxidase (QSOX1) was selected as a biomarker candidate from the enriched proteins in the secretion of lung cancer cells. QSOX1 levels were higher in 82% (51 of 62 tissues) of lung cancer tissues compared to adjacent normal tissues. Importantly, QSOX1 serum levels were significantly higher in cancer patients (<I>p</I> < 0.05, Area Under curve (AUC) = 0.89) when measured by multiple reaction monitoring (MRM). Higher levels of QSOX1 were also uniquely detected in lung cancer tissues, among several other solid cancers, by immunohistochemistry. QSOX1-knock-downed Lewis lung cancer (LLC) cells were less viable from oxidative stress and reduced migration and invasion. In addition, LLC mouse models with QSOX1 knock-down also proved that QSOX1 functions in promoting cancer metastasis. In conclusion, QSOX1 might be a lung cancer tissue-derived biomarker and be involved in the promotion of lung cancers, and thus can be a therapeutic target for lung cancers.</P>
Occult Breast Cancers Manifesting as Axillary Lymph Node Metastasis in Men: A Two-Case Report

Occult breast cancer is a type of breast cancer without any symptoms on the breasts or any abnormalities upon radiologic examination such as mammography. In males, there are few cases of breast cancer, the rate of diagnosis of occult breast cancer is very low, and little is known about this disease. We experienced two cases of occult breast cancers manifesting as axillary lymph node metastasis in men. They had a palpable lesion on axillary area several years ago and had not seen a doctor about it. As such there was no abnormality on evaluations for cancer except for axillary lymph node showing signs of carcinoma (primary or metastatic) on biopsy and estrogen receptor-positive and progesterone receptor-positive on immunohistochemistry. The patients were diagnosed with occult breast cancer, and treatments were performed. Herein, we report the rare cases of occult breast cancers in men.
Characterization of an inclusion complex of, 7-dehydrocholesterol and cyclodextrin

7-Dehydrocholesterol (7-DHC) plays an important role in the biosynthesis of vitamin D. It is virtually insoluble in water, in which its manipulation is difficult, but the problems may be alleviated by cyclodextrin inclusion. Cyclodextrins are known to form inclusion complex with many kinds of hydrophobic compounds because theirmolecular structure generates a hydrophilic exterior surface and a hydrophobic cavity interior. The purpose of this study was to characterize inclusion complex of 7-dehydrocholesterol with cyclodextrin (hydroxypropyl-b-cyclodextrin, HPbCD). The inclusion complex showed good solubility in contrast to physical mixture. The inclusion efficiency determined by HPLC analysis was 83.72%. Additionally, the inclusion complex was characterized by UV–vis spectrophotometry, Fourier transform infrared spectroscopy (FT-IR) and differential scanning calorimetry (DSC). These results demonstrated that 7-dehydrocholesterol with hydroxypropyl-b-cyclodextrin formed an inclusion complex.
Triacetate cellulose gate dielectric organic thin-film transistors

<P><B>Abstract</B></P> <P>Here, we report on the performance and the characterization of all solution-processable top-contact organic thin-film transistors (OTFTs) consisting of a natural-resourced triacetate cellulose gate dielectric and a representative hole-transport poly[2,5-bis(3-dodecylthiophen-2-yl)thieno[3,2-<I>b</I>]thiophene] (pBTTT) semiconductor layer on rigid or flexible substrates. The bio-based triacetate cellulose layer has an important role in the OTFT fabrication because it provides the pBTTT semiconducting polymer with highly suitable gate dielectric properties including a low surface roughness, hydrophobic surface, appropriate dielectric constant, and low leakage current. The triacetate cellulose gate dielectric-based pBTTT OTFTs exhibit an average filed-effect mobility of 0.031 cm<SUP>2</SUP>/Vs similar to that obtained from a SiO<SUB>2</SUB> gate dielectric-based OTFT device in ambient conditions. Even after a bending stimulation of 100 times and in an outward bending state, the flexible triacetate cellulose gate pBTTT OTFT device still showed excellent electrical device performance without any hysteresis.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We introduced natural-resourced triacetate cellulose as a gate dielectric in bio-based OTFT (BiOTFT) devices. </LI> <LI> The environmentally-friendly TAC showed good surface properties and suitable gate dielectric characteristics. </LI> <LI> The TAC pBTTT BiOTFT devices exhibited high mobility values and reliably stable device performance. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

<P>Asthma is a chronic inflammatory disease characterized by T-lymphocyte and eosinophil infiltration, mucus overproduction and airway hyper-responsiveness. The present study examined the therapeutic effects and action mechanism of a saponin-enriched extract of <I>Asparagus cochinchinensis</I> (SEAC) on airway inflammation and remodeling in an ovalbumin (OVA)-induced asthma model. To accomplish this, alterations of the nitric oxide (NO) level, inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression levels, as well as variations in immune cell numbers, immunoglobulin E (IgE) concentration, histopathological structure and inflammatory cytokine levels were measured in lipopolysaccharide (LPS)-activated RAW264.7 cells or an OVA-induced mouse model of asthma treated with SEAC. The concentration of NO and mRNA levels of COX-2 and iNOS were significantly decreased in the SEAC + LPS-treated RAW264.7 cells compared with the vehicle + LPS-treated RAW264.7 cells. Additionally, in the OVA-induced asthma model, the number of immune cells in the bronchoalveolar lavage fluid, the concentration of OVA-specific IgE, the infiltration of inflammatory cells, the bronchial thickness and the levels of the inflammatory mediators interleukin-4 (IL-4), IL-13 and COX-2 were significantly lower in the OVA + SEAC-treated group compared with the OVA + vehicle-treated group. In addition, a significant reduction in goblet cell hyperplasia, peribronchiolar collagen layer thickness and VEGF expression for airway remodeling was detected in the OVA + SEAC-treated group compared with the OVA + vehicle-treated group. These findings indicate that SEAC is a suppressor of airway inflammation and remodeling, and may therefore be useful as an anti-inflammatory drug for the treatment of asthma.</P>
Experience of treatment of patients with granulomatous lobular mastitis

Purpose: To present the author’s experience with various treatment methods of granulomatous lobular mastitis (GLM) and to determine effective treatment methods of GLM. Methods: Fifty patients who were diagnosed with GLM were classified into five groups based on the initial treatment methods they underwent, which included observation (n = 8), antibiotics (n = 3), steroid (n = 13), drainage (n = 14), and surgical excision (n = 12). The treatment processes in each group were examined and their clinical characteristics, treatment processes, and results were analyzed respectively. Results: Success rates with each initial treatment were observation, 87.5%; antibiotics, 33.3%; steroids, 30.8%; drainage, 28.6%; and surgical excision, 91.7%. In most cases of observation, the lesions were small and the symptoms were mild. A total of 23 patients underwent surgical excision during treatment. Surgical excision showed particularly fast recovery, high success rate (90.3%) and low recurrence rate (8.7%). Conclusion: The clinical course of GLM is complex and the outcome of each treatment type are variable. Surgery may play an important role when a lesion is determined to be mass-forming or appears localized as an abscess pocket during breast examination or imaging study.

Point mutations in the human cardiac homeobox gene NKX2.5 are associated with familial atrial septal defect (ASD), atrioventricular (AV) conduction disturbance, as well as sudden cardiac death. To date, more than 60 NKX2.5 mutations have been documented, but there are no reports in Korea. We are reporting the first Korean family with ASD and AV block associated with a novel mutation in the NKX2.5 coding region. A 9-year-old boy presented with a slow and irregular pulse, and was diagnosed with secundum ASD and first degree AV block. The boy’s father, who had a history of ASD correction surgery, presented with second degree AV block and atrial fibrillation. The boy’s brother was also found to have secundum ASD and first degree AV block. There were two sudden deaths in the family. Genetic testing revealed a novel mutation of NKX2.5 in all affected members of the family.
