      • Cordycepin diminishes thymic stromal lymphopoietin-induced interleukin-13 production

        <P>Atopic dermatitis (AD) is known to aggravate by thymic stromal lymphopoietin (TSLP) and TSLP is also known to up-regulate mast cell proliferation via production of interleukin (IL)-13. Thus, we investigated whether cordycepin could regulate mast cell proliferation induced by TSLP in human mast cell line, HMC-1 cell. Cordycepin significantly diminished the production and mRNA of IL-13 through the down-regulation of phosphorylated-signal transducer and activation of transcription 6 in the TSLP-stimulated HMC-1 cells. Cordycepin also significantly diminished the cell proliferation via down-regulating MDM2 and Bc12 levels and up-regulating p53, caspase-3, and cleaved poly ADP-ribose polymerase levels in the TSLP-stimulated HMC-1 cells. Moreover, cordycepin significantly diminished the production of IL-6, tumor necrosis factor-alpha, and IL-1 beta in the TSLP-stimulated HMC-1 cells. In conclusion, our study shows that cordycepin has potential effect for the treatment of allergic inflammatory diseases through the blockade of IL-13 and MDM2 exacerbated by TSLP.</P>

        Conjugation Process in Spirogyra varians Monitored with FITC-lectins(Zygnemataceae, Chlorophyta)

        The conjugation processes of a filamentous freshwater green alga Spirogyra varians were examined using FITC-lectins. Conjugation comprised five steps: 1) aligning with adjacent filaments, 2) formation of conjugation protru-sion (papilla), 3) fusion of the protrusions, 4) formation of conjugation tube,and 5) formation of zygotes. Three lectins, ConA, RCA and UEA, showed considerable labeling during the progression of conjuation. FITC-ConA labeled the surfaces of filaments throughout the whole conjugation processes. FITC-RCA labeling was observed at the conjugation protrusions only after the papilla formation. Strong labeling continued until formationg of zygotes at the contacting area where the conjugation tube developed, but no labeling was detected on the surface of vegetative filaments. The labeling decreased gradually over time and disappeared when zygotes were formed. FITC-UEA showed similar labeling pattern with FITC-RCA except that weak labeling remained after zygote formation. Inhibition experiments using RCA, UEA which are complementary to sugars L-fucose and D-galactose, showed considerable decrease of conjugation (<32% vs. 70% in control). These results suggested that the lectin-carbohydrate recognition system might be involved in the conjugation of spirogyra varians.

      • Inhibition of MDM2 expression by rosmarinic acid in TSLP-stimulated mast cell

        <P>Rosmarinic acid (RA) has an anti-inflammatory property while thymic stromal lymphopoietin (TSLP) has an important role in mast cell-mediated inflammatory responses. Thus, the aim of this study was to determine the regulatory effect of RA in TSLP-stimulated human mast cell line, HMC-1 cells, and short ragweed pollen-induced allergic conjunctivitis mouse model. As a result, we found that RA significantly decreased the TSLP-induced mast cell proliferation and murine double minute (MDM) 2 expression. RA significantly decreased the levels of interleukin (IL)-13 and phosphorylated the signal transducer and activation of transcription 6 in the TSLP-stimulated HMC-1 cells. RA induced the increment of p53 levels, caspase-3 activation, and poly-ADP-ribose polymerase cleavage and the reduction of the procaspase-3 and BcI2. RA significantly reduced the production of tumor necrosis factor-alpha, IL-1 beta, and IL-6 on the TSLP-stimulated HMC-1 cells. In addition, RA significantly reduced the levels of IgE, IL-4, and TSLP in the short ragweed pollen-induced allergic conjunctivitis mouse model. In conclusion, the results of the study suggest that RA has a significant anti-inflammatory effect on TSLP-induced inflammatory reactions.         Ameliorative effect of atractylenolide III in the mast cell proliferation induced by TSLP

        <P><B>Abstract</B></P> <P>Atractylenolide III (ATL-III) is an active compound of <I>Atractylodes lancea</I>, which has been widely used for the treatment of cancer. Cancer is closely connected with inflammation, and many anti-inflammatory agents are also used to treat cancer. We investigated the influence of ATL-III on thymic stromal lymphopoietin (TSLP)-induced inflammatory reactions. Pretreatment with ATL-III suppressed murine double minute 2 levels and promoted p53 levels in TSLP-treated human mast cell, HMC-1 cells. Mast cell proliferation increased by TSLP or IL-3 stimulation was significantly decreased by ATL-III pretreatment. Interleukin (IL)-13 and phosphorylated signal transducer and activator of transcription 3, 5, and 6 levels in TSLP-treated HMC-1 cells were also decreased by ATL-III pretreatment. In addition, ATL-III decreased the TSLP-induced production of proinflammatory cytokines (IL-6, IL-1β, tumor necrosis factor-α, and IL-8). ATL-III decreased the levels of Bcl2 and procaspase-3 and increased caspase-3 activation and cleaved PARP levels. Furthermore, ATL-III decreased TSLP-induced mast cell proliferation and the production of inflammatory cytokine by LAD2 cells.         Cordycepin Suppresses Thymic Stromal Lymphopoietin Expression via Blocking Caspase-1 and Receptor-Interacting Protein 2 Signaling Pathways in Mast Cells

        <P>Cordycepin (3'-deoxyadenosine) is one of the active components isolated from Cordyceps militaris, and has been shown to have anti-inflammatory, anti-oxidant, anti-aging, and anti-cancer effects. Mast cell-derived thymic stromal lymphopoietin (TSLP) plays an important role in the pathogenesis of allergic inflammatory reactions. Here, we investigated the regulatory effect and mechanisms of cordycepin on the expression of TSLP in the human mast cell line, HMC-1 cells, and in the human keratinocyte cell line, HaCaT cells. Cordycepin significantly decreased the production and mRNA expression of TSLP through the inhibition of caspase-1 and nuclear factor-kappa B activation. Cordycepin also significantly reduced the phosphorylation of receptor-interacting protein 2 and inhibitory kappa B (I kappa B) kinase beta. Cordycepin significantly decreased the production and mRNA expression of interleukin (IL)-8, IL-1 beta, IL-6, and tumor necrosis factor-alpha in activated HMC-1 cells. Moreover, cordycepin significantly decreased the levels of TSLP in activated HaCaT cells. Our studies suggest that cordycepin can be applied to the treatment of allergic inflammatory diseases exacerbated by TSLP.</P>

        본 연구는 공공기관에 있어서 가족친화적 조직분위기가 조직시민행동에 미치는 영향과 이들 간의 관계에서 협력적 노사관계의 매개효과를 검증하였다. 공공기관에 근무하고 있는 292명을 대상으로 살펴본 결과 첫째, 가족친화적 조직분위기는 협력적 노사관계에 긍정적인 영향을 주었다. 둘째, 가족친화적 조직분위기는 구성원들의 조직시민행동에도 긍정적인 영향을 주었다. 셋째, 조직구성원들이 지각하는 협력적 노사관계는 조직시민행동에 긍정적인 영향을 주었다. 마지막으로 협력적 노사관계는 가족친화적 조직분위기와 조직시민행동의 관계에서 부분 매개역할을 보였다. 이상의 연구결과를 토대로 본 연구의 시사점과 한계점, 향후 연구방향을 제시하였다. This study examined the effect on family-friendly organization culture on organized citizen actions and the intermediary relationship between these factors with cooperative labor-management relations. Based on research of 292 people working in public office, such results were observed: firstly, family-friendly organization culture gave positive effects on cooperative labor-management relations. Secondly, family-friendly organization culture also gave positive effects on organized citizen actions. Lastly, cooperative labor-management relations partly served as a intermediary factor in case of the relationship between family-friendly organization culture and organized citizen actions. To conclude, the implications, limitations and later study guides are discussed based on these research results.

        Molecular Docking-assisted Protein Chip Screening of Inhibitors for Bcl-2 Family Protein-protein Interaction to Discover Anticancer Agents by Fragment-based Approach

        For fragment-based cancer drug discovery, we introduced a molecular docking simulation combined with a protein chip assay. Protein chip technology was used to find fragment-hits that had inhibitory activity against Bcl-2 protein from 131 pre-selected fragment chemicals. Molecular docking simulation was performed for the 12 identified fragment-hits to establish the binding mode of these compounds in the Bcl-2 site. Using the molecular docking-assisted protein chip screening system, we derived a virtual compound structure with an important scaffold feature for interaction with the Bcl-2 protein. We then tested the anticancer activity of 26 compounds that were similar to the scaffold structure. The anticancer activity was confirmed by MTT-assay in A549 lung cancer cells. Finally, three chemicals showed dose-dependent inhibitory activity against cancer cell proliferation. We suggest that the present molecular docking-assisted protein chip assay can be used as a platform technology in the fragment-based drug development process to discover inhibitory agents of protein-protein interactions.
