S6K1 Phosphorylation of H2B Mediates EZH2 Trimethylation of H3: A Determinant of Early Adipogenesis

S6K1 has been implicated in a number of key metabolic responses, which contribute to obesity. Critical among these is the control of a transcriptional program required for the commitment of mesenchymal stem cells to the adipocytic lineage. However, in contrast to its role in the cytosol, the functions and targets of nuclear S6K1 are unknown. Here, we show that adipogenic stimuli trigger nuclear translocation of S6K1, leading to H2BS36 phosphorylation and recruitment of EZH2 to H3, which mediates H3K27 trimethylation. This blocks Wnt gene expression, inducing the upregulation of PPARγ and Cebpa and driving increased adipogenesis. Consistent with this finding, white adipose tissue from S6K1-deficient mice exhibits no detectable H2BS36 phosphorylation or H3K27 trimethylation, whereas both responses are highly elevated in obese humans or in mice fed a high-fat diet. These findings define an S6K1-dependent mechanism in early adipogenesis, contributing to the promotion of obesity.

<P>Shikonin has been reported to induce apoptosis and inhibit angiogenesis in vivo and in vitro. 6-(1-propoxyiminoalkyl)-5,8-dimethoxyoxy 1,4-naphtoquinone S-64 (DMNQ S-64) was synthesized as a shikonin derivative. In this article, the underlying apoptotic mechanism of DMNQ S-64 was examined. DMNQ S-64 exerted cytotoxicity against A549 lung carcinoma cells with IC(50) of 27.3 microM. Apoptotic bodies were observed in DMNQ S-64-treated A549 cells by 4'-6-diamidino-2-phenylindole (DAPI) staining assay. DMNQ S-64 also increased sub-G1 DNA portion in a concentration-dependent manner by flow cytometric analysis. Western blotting has revealed that DMNQ S-64 effectively activates the expression of caspase 8, 9, and 3, cleaves poly (ADP-ribose) polymerase, and increases the ratio of Bax/Bcl-2. Furthermore, cytochrome c was released in a concentration-dependent manner by DMNQ S-64. Similarly, DMNQ S-64 significantly increased caspase 3 activity by enzyme-linked immunosorbent assay (ELISA). It also significantly inhibited the level of prostaglandin E2 (PGE(2)) by ELISA and downregulated the expression of cyclooxygenase-2 (COX-2) in a concentration-dependent manner. Taken together, DMNQ S-64 may exhibit cytotoxicity against A549 cells via caspase activation and COX-2 inhibition.</P>
벼 육묘트레이를 이용한 어린 잎 채소 생육에 미치는 광 조절 영향

벼 육묘 트레이를 활용한 어린잎 채소 재배 가능성을 탐색하고자 재배 기간 중 차광 처리하여 다채, 청경채, 알파인배추, 청치마 상추 등 4종을 17일간 재배하였다. 차광 처리는 무차광 17일(NS), 차광 8일+무차광 9일 (S8NS9), 무차광 8일+차광 9일(NS8S9), 차광 17일(S17) 하였다. 처리에 따른 누적 광량은 NS>S8NS9>NS8S9>S17처리구 순으로 높았으나, 누적 온도는 처리에 따른 차이가 크지 않았다 차광 8일 째 어린잎채소 4종의 생육과 엽록소함량은 감소하였다. 광 조절 17일 째 어린잎채소 4종의 초장, 엽장, 엽폭, 경경, 생체중, 엽록소 함량 등의 생육은 무차광 NS 처리구에서 가장 높았다. 생육 초기 광조절에 따른 생육 후기 초장과 생체중 증가율이 어린잎채소 4종 모두 NS 또는 S8NS9 처리구에서 높았다. 따라서 벼 육묘장 다층 시스템에서의 공간 활용을 고려한 어린잎채소 재배시 차광이 많은 하부 공간은 생육초기에 활용하고, 수확기에는 광 확보가 이루어지는 상부에서 재배가 이루어짐이 적합하리라 본다. This study was conducted to investigate the effect of light control on growth of baby leaf vegetables (tatsoi, pak-choi, chinese cabbage ‘alpine’ and leaf lettuce ‘cheongchima’) in rice seedling tray. Light treatment controlled in non-shading (NS), shading for 8 days+non-shading for 9 days (S8NS9), non-shading for 8 days+shading for 9 days (NS8S9), and shading for 17 days (S17). Integrated solar radiation in NS was highest, and in S8NS9>NS8S9>S17 order. Integrated temperature did not shown difference among the treatments for growing periods. Growth and chlorophyll content of baby leaf vegetables were lowest in shading treatment for 8days. The highest growth characteristics (plant height, leaf length, leaf width, stem diameters, and fresh weight) and chlorophyll content of baby leaf vegetables was shown in NS. Increment rate of plant height and fresh weight of baby leaf vegetables were high in NS or S8NS9 as affected by light control for 8 days in early growth periods. These results suggest that we can utilize in low layer for early period and in high layer for harvest periods for baby vegetable production in rice seedling factory.

질환성과 관련된 세균의 분포 및 유전자형을 탐색하고자 구강농양 및 골수염의 급성감염 혼자와 진료실 및 실험실의 정상인을 대상으로 시료를 채취하여 포도상구균을 분리 및 동정을 시행하고, 특성을 규명하였으며, plasmid 및 염색유전자를 분리하여 제한효소를 처리후 전기영동을 실시하고 분리된 plasmid로 탐색자를 제작하여 dot blot을 시행하였다. 대부분의 급성환자에서 분리된 포도상구균을 S. lugdunensis와 S. aureus이었으나, 진료실 및 실험실에서는 coagulase 음성 staphylococci가 분리되었다. 급성환자에서 분리된 포도상구균은 ampicillin과 penicillin에 내성을 보였다. 분리된 S. lugdunensis균주중 네 균주는 δ형의 용혈소를 생산하였다. Plasmid를 분리한 결과 S. lugdunensis균주중 세 균주는 약 6.5 kilobases이었으나 S. aureus는 약 4.3 kilobases 정도 크기의 band를 보였다. S. lugdunensis에서 분리된 plasmid로 제작한 탐색자로 dot blot를 시행한 결과 치과 영역에서 분리한 plasmid를 갖는 균주는 양성반응을 보였다. 염색체유전자의 유전자형을 분석한 결과 δ형의 용혈소를 생산한 네 균주의 S. lugdunensis는 유사한 유전자형을 보였다. 이러한 연구결과 질환의 진행에 S. lugdunensis가 중요한 역할을 하는 것으로 생각되고, 치과영역에 존재하는 plasmid는 공통적인 유전자 서열을 갖는 것으로 추정된다.

<P>In South Korea, nine million tons of fly ash (FA) are annually produced and approximately 70 % is reutilized for industrial demand. For the prompt reuse and insufficient reclamation site, quality control of FA which is main productive construction material from coal ash is very important. Assessed Pozzolanic-activity Index (API) test which needs only 2 days for evaluation of pozzolanic reaction is currently considered as an alternative of activity index measurement. This paper aims for an applicability of API test for prompt quality control and investigation of domestic FA properties. For the work, FAs from two different power plant types are prepared, and quality tests are carried out based on Korean Standards (KS) methods and API method. Lots of test results are compared with those from API and K-value test for FA with age of 7 days-1 year. From the test results for FA aged 1 year, API results are evaluated to be closely related with those from activity index and K-value, and the correlations are improved with increasing ages regardless of plant types. The applicability of API test is verified and the reduced period of FA quality evaluation can accelerate prompt use and the related process of FA.</P>

A rapidly spreading unidentified type of pneumonia was initially reported in China in December 2019. On March 11, 2020, the World Health Organization declared the coronavirus disease 2019 (COVID-19) a global pandemic. The world is making efforts to overcome COVID-19 using the experience of each country. From the beginning, Korea has been relatively successful in managing COVID-19 according to the "3T principle," that is, test-trace-treat. To share these experiences with the international community, in June 2020, Korea launched 18 projects in an attempt to standardize the COVID-19 response technology. This paper introduces the standardization trend of Korea's response to COVID-19.

The effects of B-site non-stoichiometry in [(K0.5Na0.5)] [(Nb0.9+xTa0.1)O3] ceramics on microstructure and piezoelectric properties were investigated. The density, electromechanical coupling factor (kp), mechanical quality factor (Qm), piezoelectric constant (d33), TC and TOeT of NKNT ceramics with x = 0.0065 showed the optimum values of 4.58 g/cm3,0.427, 1554, 109 pC/N, 373 ℃ and 226 ℃, respectively, suitable for piezoelectric motor, and transformer applications. <P>For large scale batteries, sulfur is an attractive cathode active material for lithium batteries because of high theoretical capacity of 1675 mA hg(-1). However, severe capacity fading and low conductivity of sulfur are significant challenges for its practical application. Here, we report a facile approach to prepare reduced graphene oxide nanoscroll embedded with well-dispersed sulfur (S/GNSC) through a one-pot oxidation of sulfide and reduction of graphene oxide followed by freeze-casting process to generate graphene nanoscroll. The S/GNSC takes the novel shape of reduced graphene oxide sheet spirally wrapped into one-dimensional tubular structure, which enhances electrochemical performance by acting as physical barrier to prevent polysulfide from dissolving and an effective network to promote electron and Li+ transport during reaction. Therefore, the S/GNSC composite with high sulfur loading of 76 wt% exhibits high initial capacity of 1295 mA h g(-1) and reversible capacity of 744 mA h g(-1) after the 100th cycle at 0.2 degrees C when it is used as a cathode material for lithium-sulfur battery, while the capacity of sulfur loaded on reduced graphene oxide sheet (S/rGO) is continuously fading to 510 mA h g(-1) after the 100th cycle due to the irreversible loss of polysulfide.
