Background and Purpose Intravenous tenecteplase (TNK) efficacy has not been well demonstrated in acute ischemic stroke (AIS) beyond 4.5 hours after onset. This study aimed to determine the effect of intravenous TNK for AIS within 4.5 to 24 hours of onset. Methods In this pilot trial, eligible AIS patients with diffusion-weighted imaging (DWI)-fluid attenuated inversion recovery (FLAIR) mismatch were randomly allocated to intravenous TNK (0.25 mg/kg) or standard care within 4.5–24 hours of onset. The primary endpoint was excellent functional outcome at 90 days (modified Rankin Scale [mRS] score of 0–1). The primary safety endpoint was symptomatic intracranial hemorrhage (sICH). Results Of the randomly assigned 80 patients, the primary endpoint occurred in 52.5% (21/40) of TNK group and 50.0% (20/40) of control group, with no significant difference (unadjusted odds ratio, 1.11; 95% confidence interval 0.46–2.66; <i>P</i>=0.82). More early neurological improvement occurred in TNK group than in control group (11 vs. 3, <i>P</i>=0.03), but no significant differences were found in other secondary endpoints, such as mRS 0–2 at 90 days, shift analysis of mRS at 90 days, and change in National Institutes of Health Stroke Scale score at 24 hours and 7 days. There were no cases of sICH in this trial; however, asymptomatic intracranial hemorrhage occurred in 3 of the 40 patients (7.5%) in the TNK group. Conclusion This phase 2, randomized, multicenter study suggests that intravenous TNK within 4.5–24 hours of onset may be safe and feasible in AIS patients with a DWI-FLAIR mismatch.

Purpose: Metastatic castration-resistant prostate cancer (mCRPC) has a poor prognosis. Abiraterone acetate (AA), enzalutamide, and chemotherapy are first-line treatments for patients with mCRPC. This study examined prognostic factors for AA response in the form of prostate-specific antigen (PSA) kinetics throughout androgen-deprivation therapy (ADT) in chemonaïve patients with mCRPC. Materials and Methods: We retrospectively included data from 34 chemonaïve patients with mCRPC who had received AA at some point between January 2017 and December 2018. We separated patients into two study arms according to the decrease in PSA percentages after use of AA for 3 months. We correlated PSA kinetics parameters with response and compared the two study groups with respect to PSA kinetics. Results: The patients’ median age was 77 years. In the total group of patients, 64% had a response to AA, whereas 35% did not. The ratio of the PSA level at nadir to the level during ADT was significantly higher in the AA-sensitive group (19.78 vs. 1.03, p=0.019). Conclusions: Patients who experienced a dramatic change in PSA level during ADT were more likely to be resistant to AA after progression to mCRPC. Chemotherapy rather than AA might be more suitable as a first-line treatment for these patients.

Ligand-dependent or independent oligomerization of receptor protein tyrosine kinase (RPTK) is often an essential step for receptor activation and intracellular signaling. The novel oncogene with kinase-domain (NOK) is a unique RPTK that almost completely lacks an ectodomain, expresses intracellularly and activates constitutively. However, it is unknown whether NOK can form oligomer or what function oligomerization would have. In this study, two NOK deletion mutants were generated by either removing the ectodomain (NOKECD) or including the endodomain (NOK-ICD). Co-immunoprecipitation demonstrated that the transmembrane (TM) domain of NOK was essential for its intermolecular interaction. The results further showed that NOK aggregated more closely as lower order oligomers (the dimer- and trimer-sized) than either deletion mutant did since NOK could be cross-linked by both Sulfo-EGS and formaldehyde, whereas either deletion mutant was only sensitive to Sulfo-EGS. Removing the NOK TM domain (NOK-ICD) not only markedly promoted higher order oligomerization, but also altered the subcellular localization of NOK and dramatically elevated the NOK-mediated constitutive activation of extracellular signal-regulated kinase (ERK). Moreover, NOK-ICD but not NOK or NOKECD was co-localized with the upstream signaling molecule RAS on cell membrane. Thus, TM-mediated intermolecular contacting may be mainly responsible for the constitutive activation of NOK and contribute to the autoinhibitory effect on RAS/MAPK signaling.

In [1], they build two populations' cellular automata model with predation based on the Penna model. In this paper, uncertain aspects and problems of imprecise and vague data are considered in this model. A fuzzy cellular automata model containing movable wolves and sheep has been built. The results show that the fuzzy cellular automata can simulate the classical CA model and can deal with imprecise and vague data.

<P>A marine bacterium, designated SCSIO 03483<SUP>T</SUP>, was isolated from a marine sediment sample collected from the Nansha Islands in the South China Sea. The strain produced roundish colonies with diffusible yellow-coloured pigment on nutrient agar medium or marine agar 2216. Optimal growth occurred in the presence of 0–4 % (w/v) NaCl, at pH 7.0 and a temperature range of 28–37 °C. 16S rRNA gene sequence analysis indicated that the isolate belonged to the family <I>Flavobacteriaceae</I> and showed relatively high sequence similarity with <I>Imtechella halotolerans</I> K1<SUP>T</SUP> (92.7 %). Phylogenetic analysis based on nearly complete 16S rRNA gene sequences revealed that the isolate shared a lineage with members of the genera <I>Imtechella</I>, <I>Joostella</I> and <I>Zhouia</I>. Phospholipids were phosphatidylethanolamine, two unidentified aminolipids and three unknown polar lipids. The major respiratory quinone was MK-6 and the major fatty acids were iso-C<SUB>15 : 0</SUB>, iso-C<SUB>17 : 0</SUB> 3-OH and summed feature 3 (C<SUB>16 : 1</SUB>ω6<I>c</I>/C<SUB>16 : 1</SUB>ω7<I>c</I>). The DNA G+C content of strain SCSIO 03483<SUP>T</SUP> was 38.4 mol%. On the basis of phenotypic, chemotaxonomic and molecular data, strain SCSIO 03483<SUP>T</SUP> represents a novel species in a new genus in the family <I>Flavobacteriaceae</I>, for which the name <I>Sinomicrobium oceani</I> gen. nov., sp. nov. is proposed. The type strain of <I>Sinobacterium oceani</I> is SCSIO 03483<SUP>T</SUP> ( = KCTC 23994<SUP>T</SUP> = CGMCC 1.12145<SUP>T</SUP>).</P>

This study investigated the effects of fucosylated chondroitin sulfate (CHS) isolated from sea cucumber on glucose metabolism and insulin signaling in the liver of insulin-resistant C57BL/6 mice fed a high-fat, high-sucrose diet (HFSD). Male C57BL/6J mice were randomly assigned into six groups: control; HFSD; 1mg RSG/kg$body weight (RSG); 80 mg CHS/kg$body weight (CHS); 20 mg CHS + 1mg RSG/kg$body weight (20 CHS + RSG); and 80 mg CHS + 1mg RSG/kg$body weight (80 CHS + RSG). Blood glucose, insulin parameters, glucose metabolism-related enzymes activities and insulin-signaling transducers in the liver were analyzed at 19 weeks. Results showed that CHS significantly decreased body weight gain, adipose tissue weight, and fasting blood glucose and serum insulin levels in insulin-resistant mice. Rosiglitazone (RSG) is an effective thiazolidinedione hypoglycemic agent, and CHS synergistically enhanced the effect of RSG. CHS feeding normalized the activities of hexokinase, pyruvate kinase, glycogen phosphorylase, glucose-6-phosphatase, and increased glycogen reserves in the liver. Quantitative real-time polymerase chain reaction (qRT-PCR) analysis showed that CHS promoted the mRNA expression of insulin receptors (IR), insulin receptor substrate 2 (IRS-2), phosphatidylinositol 3 kinase (PI3K), protein kinase B (PKB), and glycogen synthase (GS) in the liver of insulin resistant mice, and inhibited glycogen synthase kinase-3 (GSK-3b) mRNA expression. The results suggested that CHS treatment improved glucose metabolism by modulating metabolic enzymes and promoting the PI3K/PKB/GSK-3b signaling pathway mediated by insulin at the transcriptional level. These results provided strong justification for the development of CHS as a functional food.

In this work, reduced graphene oxide (RGO)/ferroferric oxide (RGO/Fe3O4) hybrid composite was successfully fabricated by a facile one-step solvothermal method. The structure, chemical composition, morphology and magnetic properties of the samples were investigated in detail. Fe3O4 microspheres with an average diameter of 250 nm were uniformly anchored on the surface of the RGO sheets without large aggregation. Moreover, the results demonstrated that the hybrid composite exhibited obviously enhanced microwave absorption properties compared with the pure Fe3O4 microspheres. The minimum reflection loss (RL) of the hybrid composite reached -36.8 dB at 17.2 GHz with a thickness of 5.0mm and its effective absorption bandwidth (lower than - 10 dB) was 3.9 GHz with a thickness of 2.5 mm. Significantly, the hybrid composite exhibited a dual-waveband absorption characteristic covering the C and Ku bands. Besides, the relationship between the matching thickness and peak frequency was reasonably explained according to the quarter-wavelength matching theory. Therefore, the obtained composite was a promising candidate for application in microwave absorption.
