Synergistic growth in bacteria depends on substrate complexity

Both positive and negative interactions among bacteria take place in the environment. We hypothesize that the complexity of the substrate affects the way bacteria interact with greater cooperation in the presence of recalcitrant substrate. We isolated lignocellulolytic bacteria from salt marsh detritus and compared the growth, metabolic activity and enzyme production of pure cultures to those of three-species mixed cultures in lignocellulose and glucose media. Synergistic growth was common in lignocellulose medium containing carboxyl methyl cellulose, xylan and lignin but absent in glucose medium. Bacterial synergism promoted metabolic activity in synergistic mixed cultures but not the maximal growth rate (μ). Bacterial synergism also promoted the production of β- 1,4-glucosidase but not the production of cellobiohydrolase or β-1,4-xylosidase. Our results suggest that the chemical complexity of the substrate affects the way bacteria interact. While a complex substrate such as lignocellulose promotes positive interactions and synergistic growth, a labile substrate such as glucose promotes negative interactions and competition. Synergistic interactions among indigenous bacteria are suggested to be important in promoting lignocellulose degradation in the environment.

Purpose: Multidrug resistance (MDR) remains a major obstacle in the treatment of triple-negative breast cancer (TNBC) with conventional chemotherapeutic agents. A previous study demonstrated that hsa-miRNA-143-3p plays a vital role in drug resistance of TNBC. Downregulation of hsa-miRNA-143-3p upregulated the expression of its target protein cytokine-induced apoptosis inhibitor 1 (CIAPIN1) in order to activate MDR, while upregulation of hsa-miRNA-143-3p effectively enhances the sensitivity of drug-resistant TNBC cells to chemotherapeutics. The present study aimed to further verify these findings in vivo. Methods: We established a hypodermic tumor nude mice model using paclitaxel- resistant TNBC cells. We expressed ectopic hsa-miRNA- 143-3p under the control of a breast cancer-specific human mammaglobin promoter that guided the efficient expression of exogenous hsa-miRNA-143-3p only in breast cancer cells. Thereafter, we overexpressed hsa-miRNA-143-3p in xenografts using a recombinant virus system and quantified the expression of hsa-miRNA-143-3p, CIAPIN1 protein, and proteins encoded by related functional genes by western blot. Results: We successfully completed the prospective exploration of the intravenous virus injection pattern from extensive expression to targeted expression. The overexpression of hsa-miRNA-143-3p significantly alleviated chemoresistance of TNBC by inhibiting viability. In addition, we observed that the expression of CIAPIN1 as a hsa-miRNA-143-3p target protein was remarkably decreased. Conclusion: We partly illustrated the mechanism underlying the hsa-miRNA-143-3p/CIAPIN1 drug resistance pathway. HsamiRNA- 143-3p as a tumor suppressive microRNA may be a novel target to effectively reverse MDR of TNBC in vivo.
MiR-449a attenuates autophagy of T-cell lymphoma cells by downregulating ATG4B expression

Increasing evidence suggests the role of miR-449a in the regulation of tumorigenesis and autophagy. Autophagy plays an important role in the malignancy of T-cell lymphoma. However, it is still unknown whether miR-449a is associated with autophagy to regulate the malignancy of T-cell lymp homa. In this study, we for the first time demonstrated that miR-449a enhanced apoptosis of T-cell lymphoma cells by decreasing the degree of autophagy. Further, miR-449a downregulated autophagy- associated 4B (ATG4B) expression, which subsequently reduced the autophagy of T-cell lymphoma cells. Mechanistically, miR-449a decreased ATG4B protein level by binding to its mRNA 3’UTR, thus reducing the mRNA stability. In addition, studies with nude mice showed that miR-449a significantly inhibited lymphoma characteristics in vivo. In conclusion, our results demonstrated that the “miR-449a/ATG4B/autophagy” pathway played a vital role in the malignancy of T-cell lymphoma, suggesting a novel therapeutic target.
Roles of E-Cadherin (CDH1) Genetic Variations in Cancer Risk: a Meta-analysis

E-Cadherin (CDH1) genetic variations may be involved in invasion and metastasis of various cancers by altering gene transcriptional activity of epithelial cells. However, published studies on the association of CDH1 gene polymorphisms and cancer risk remain contradictory, owing to differences in living habits and genetic backgrounds. To derive a more better and comprehensive conclusion, the present meta-analysis was performed including 57 eligible studies of the association between polymorphisms of CDH1 gene promoter -160 C>A, -347 G>GA and 3'-UTR +54 C>T and cancer risk. Results showed that these three polymorphisms of CDH1 were significantly associated with cancer risk. For -160 C>A polymorphism, -160A allele carriers (CA and CA+AA) had an increased risk of cancer compared with the homozygotes (CC), and the similar result was discovered for the -160A allele in the overall analyses. In the subgroup analyses, obvious elevated risk was found with -160A allele carriers (AA, CA, CA+AA and A allele) for prostate cancer, while a decreased colorectal cancer risk was shown with the AA genotype. For the -347 G>GA polymorphism, the GAGA genotype was associated with increased cancer risk in the overall analysis with homozygous and recessive models. In addition, results of subgroup analysis indicated that the elevated risks were observed in colorectal cancer and Asian descendants. For +54 C>T polymorphism, a decreased risk of cancer was found in heterozygous, dominant and allele models. Moreover, +54T allele carriers (CT, CT+TT genotype and T allele) showed a potential protective factor in gastric cancer and Asian descendants.

The stiffness and fatigue life of deep groove ball bearings are greatly affected by interference fits, preloads and tilting moments. Up to now, most of current works failed to focus on the integrated analysis of stiffness and fatigue life, especially the off-diagonal stiffness items in the bearing stiffness matrix. This paper proposed an improved mathematical model of deep groove ball bearings with the consideration of interference fits and tilting moments. On this basis, the combined study on stiffness and fatigue life is conducted to determine the optimal match of interference fits, preloads and tilting moments. Moreover, the variations in off-diagonal stiffness items under different interference fits, preloads and tilting moments are also given. These results are beneficial for the assembly of deep groove ball bearings to obtain the best stiffness and fatigue life. Also, the sensitivity of off-diagonal stiffness items to interference fits, preloads and tilting moments is demonstrated, which greatly affects the dynamic characteristic of ball bearings.

Using the clustering analysis and gravity model, this paper investigates the evolution of likely trade-bloc phenomenon for the textile industry and the automatic data processing industry for the last three decades. For the textile industry, a significant two-bloc phenomenon could be identified, one declining bloc mainly composed of the European countries, and the other rising bloc of countries around Pacific Rim. For the automatic data processing industry, only one trade bloc could be identified. However, the core countries have gradually changed from those of Europe and USA to the countries of East Asia over the period.
Instability Prediction of Valve Motion and Internal Flow in Novel Large-scale Reciprocating Pump

To study the transient valve motions and flow pulsation of novel five-cylinder double-acting reciprocating pump, the theoretical analysis and computational fluid dynamics (CFD) method are employed to investigate the full-cycle working process of the pump. The theoretical models describing the motions and flow rates of hydraulic end are established for reciprocating pump under the actions of piston and valves. The open-close behavior of suction and discharge valves, internal turbulent flow and pulsation characteristics of the pump are studied and the effects of crank speed on those are analyzed in detail. The results show that the valves of reciprocating pump open and close quickly and non-linearly with various degrees of striking velocities and lag phenomena. Through the time and frequency domain results of flow pulsation, the single-cavity frequency is the dominant frequency. The most obvious amplitude appears for discharge flow pulsation, which gradually increases with crank speed. The results could provide useful information for the optimization of reciprocating pumping systems.
