      • Effect of Heat Treatment Condition on Quality Characteristics of Croceine Croaker Gulbi

        In a previous study, we selected salting and drying conditions using fermented product (mixed soybean and anchovy) to develop croceine croaker Gulbi with high quality. This study was carried out to analyze the quality characteristics of croceine croaker Gulbi according to heat treatment conditions. The quality characteristics of croceine croaker Gulbi by steam (100℃, 10 min) and oven (150℃, 10 min) treatment were compared. The croceine croaker Gulbi treated with steam condition showed lower histamine, VBN, and TBARS and higher antioxidant activity and overall acceptability than oven treated Gulbi. We further investigated the effect of steaming time (8, 10, 12, 15 min) on quality characteristics of croceine croaker Gulbi. Total nitrogen and amino-type nitrogen contents increased with increasing steaming time. Histamine, acid value, VBN, TBARS, and antioxidant activity were no significant difference in the steaming treatment for more than 10 min. In the sensory evaluation, the highest overall acceptability was observed in the steam treated croceine croaker Gulbi for 10 min. This work was supported by the Jeollannam-do.

      • Effect of Feed Additives with Barley by-product on the Antioxidant Activity and Immunoreactivity in the Swine

        This experiment was conducted to investigate the effects of dietary feed additives with barley by-product on antioxidant effect and Immunoreactivity in swine. The 36 heads of commercial Landrace×Yorkshire×Duroc (LYD) crossbreed pigs were divided into 3 groups: basal dietary only (control), basal dietary+2% barley by-product (T1) and basal dietary+4% barley by-product (T2). The antioxidant activity of pork loin were evaluated by DPPH and ABTS radical scavenging activity. ABTS radical scavenging activity was not significantly different among the control and treatment groups. DPPH radical scavenging activity revealed significantly higher in T2 group than in others. Next, the effect of the feed with barley by-product on the immunity of swine was investigated. The immunoglobulins (IgA, IgG and IgM) in the serum, T2 group was significantly higher than in others. As a result, dietary supplementation with 4% barley by-product could be recommended for the effective of antioxidant activity and immunity-related.

      • Effect of Ovarian Extract on Oocyte Maturation and Early Embryonic Development in Pigs

        Various factors in the ovary are known to regulate oocyte maturation and hormone secretory functions; however, the effect of ovarian extract (OE) on oocyte maturation and embryonic development in pigs remains unknown. In this study, we first evaluated whether OE supplementation in the in vitro maturation (IVM) medium alters the oocyte maturation capacity by affecting glucose/amino acid metabolites, meiotic maturation, cumulus cell (CC) expansion, and antioxidants. Various OE concentrations (50, 100, 200, 500, and 5000 μg/mL) were included in the IVM medium. Only the oocytes treated with 100 μg/mL OE exhibited an improved meiotic maturation rate when compared with that of the other groups (non-treated group, 78.6 ± 3.0% vs. 100 μg/mL OE-treated group, 81.6 ± 4.3%); however, the difference was not significant. To observe the changes in glucose and amino acid metabolism in the OE-treated oocytes, we measured the amounts of diverse constituents (glucose, lactate, glutamine, and ammonia) in the IVM medium containing OE. Lactate and ammonia levels in the OE-treated group after 44 h of IVM were higher (p < 0.01) than those in the non-treated group. In addition, the expression of the CC expansion factors (Has2 and Tnfaip6) significantly increased (p < 0.05), whereas the mRNA expression levels of antioxidative enzymes (Sod1, Cat, and Gpx1) significantly diminished (p < 0.05) in the OE-treated group. Moreover, mature oocytes treated with 100 μg/mL OE demonstrated increased subsequent embryonic development rates after 144 h of IVM. Thus, the addition of OE in IVM mediums may improve oocyte maturation capacity which could enhance antioxidative enzyme activation, energy metabolism, and expression of the CC expansion factors in porcine oocytes.

        대두와 멸치 발효소재를 이용한 염지 처리와 건조 시간이 부세 굴비의 품질 특성에 미치는 영향

        본 연구에서는 지방 산화를 억제하고 품질이 우수한 부세 굴비의 가공법을 개발하기 위해 대두와 멸치의 복합 발효소재를 첨가한 염지제 처리 및 건조 시간에 따른 부세 굴비를 제조하여 품질 특성을 비교하였다. 발효소재 첨가에 따른 염지제와 부세 굴비의 DPPH 및 ABTS 라디칼 소거능은 발효소재의 함량에 따라 증가하였으나, 발효소재 1% 이상에서 부세 굴비의 라디칼 소거능은 유의적인 차이를 보이지 않았다. 발효소재의 함량이 증가할수록 부세 굴비의 아미노태질소 함량은 증가하고 VBN, TBARS, 산가는 감소하는 결과를 나타내나, 발효소재 1% 이상에서 품질의 유의적인 차이가 크지 않으므로 염지제의 발효소재 함량은 1%로 선정하였다. 염지제의 염도에 따른 부세 굴비의 품질을 분석한 결과 염도가 증가할수록 아미노태질소 함량은 증가하고 히스타민, VBN, TBARS, 산가는 감소하는 결과는 나타내며, 염지제의 염도는 품질 개선 효과가 가장 우수한 7% 조건을 선정하였다. 염지 처리 후 건조 시간에 따른 부세 굴비의 품질을 분석한 결과 건조 시간이 증가할수록 수분은 감소하고 염도, pH, 아미노태질소, 히스타민, VBN, TBARS, 산가는 증가하였다. 그러나 건조 48시간 이하는 수분이 많아 저장성이 낮으며, 건조 96시간에서 히스타민과 산패도의 증가율이 높아지므로 부세 굴비의 건조 시간은 72시간이 적합한 것으로 판단되었다. This study was carried out to analyze the quality characteristics of croceine croaker Gulbi according to brine treatment and drying time. To develop a brine agent for croceine croaker, the effect of fermented product (mixed soybean and anchovy) content (0, 0.5, 1, 3, 5%), and salinity (3, 5, 7, 9%) in brine agent on quality characteristics of croceine croaker Gulbi were investigated. The DPPH and ABTS radical scavenging activities of the brine agent and croceine croaker Gulbi increased according to the content of the fermented product. Also, the content of amino-type nitrogen increased, and VBN, TBARS, and acid values decreased according to the content of the fermented product. Because there was no significant difference in quality characteristics at contents above 1%, the content of fermented product for bine agent was selected as 1%. As the salinity of the brine agent increased, the content of amino-type nitrogen increased, while histamine, VBN, TBARS, and acid values decreased. As a result of salinity evaluation, 7% salinity with the best quality improvement effect was selected. This study further investigated the effects of drying time on the quality characteristics of croceine croaker Gulbi. Results indicated that the drying for 72 h have a significant quality advantage in croceine croaker Gulbi processing.

        항효모 활성 까마중의 추출 조건 최적화 및 추출물의 안정성 조사

        24종의 약용 식물의 열수 추출물을 대상으로 항효모 활성을 평가한 결과 까마중 추출물에서 유일하게 항효모 활성을 나타내었다. 항효모 활성 천연 보존 소재의 개발을 위해 추출수율과 항효모 활성이 우수한 까마중의 추출 조건을 평가한 결과 50% 에탄올, 용매 비율 1:25, 70°C, 4시간 추출 조건이 선정되었다. 까마중 추출물의 산업적 활용도를 평가하기 위해 항균 스펙트럼과 온도, pH, NaCl 처리에 대한 항효모 활성 안정성을 조사하였다. 세균, 효모, 곰팡이에 대한 까마중 추출물의 생육 저해 활성을 조사한 결과 항세균 활성은 나타나지 않았지만 효모 5종과 곰팡이 3종에 대한 저해 활성을 나타내어 항진균 활성이 우수함을 확인하였다. 까마중 추출물의 항효모 활성 안정성을 평가한 결과, -20~121°C의 온도와 pH 3~9의 처리에 모두 안정하였다. NaCl에 대한 영향은 10% 처리까지 안정하였으나, 15%와 20% NaCl 처리에 항효모 활성이 2배 정도 감소하였다. 식품 부패 효모에 대한 생육 저해 활성과 항효모 활성 안정성이 우수한 까마중 추출물은 천연 식품 보존제로써 활용 가능성이 높을 것으로 기대된다. Hot water extracts of 24 medicinal plants were screened for their anti-yeast activities against Zygosaccharomyces rouxii. Solanum nigrum extract showed only anti-yeast activity. To develop a natural anti-yeast preservative from S. nigrum, extraction yield and anti-yeast activity of S. nigrum extract were investigated using various extraction solvents, solid-solvent ratios, temperatures, and times. The results showed that the optimum extraction conditions for anti-yeast S. nigrum were as follows: 50% ethanol, solid-solvent ratio of 1:25, extraction temperature of 70°C, and extraction time of 4 h. The antimicrobial activity of S. nigrum extract was tested against food-spoilage bacteria, yeasts, and molds. S. nigrum extract exhibited strong antifungal activity with minimum inhibitory concentrations of 39∼625 μg/mL against five yeasts and three molds. The stability of anti-yeast activity of S. nigrum extract was examined under different conditions of temperature, pH, and NaCl concentration. The anti-yeast activity of S. nigrum extract was stable at -20∼121°C, pH 3∼9, and 0∼10% NaCl. However, the anti-yeast activity of S. nigrum extract was reduced by 2-fold under 15% and 20% NaCl conditions.

        녹차씨에서 분리한 물질의 Candida albicans에 대한 저해 활성

        녹차씨에 함유된 항효모 물질의 분리를 위하여 녹차씨의 물 추출물을 여러 용매를 사용하여 순차 분획하고 각 분획층의 항효모 활성을 측정한 결과 n-butanol 층이 가장 높은 활성을 나타내었다. n-Butanol 분획층을 MPLC와 HPLC를 수행하여 F4-3 항효모 물질을 분리하였으며, LC-MS/MS 분석 결과 분자량 1,230의 theasaponin E와 유사한 물질로 확인되었다. F4-3 항효모 물질에 의한 효모 세포의 손상도를 neutral red 염색법으로 조사한 결과 항효모 물질의 농도가 증가할수록 Candida albicans 세포의 vacuole 손상으로 세포질이 붉게 염색되었으며, 세포 용해에 의한 cell debris도 관찰되었다. 항효모 물질에 의한 효모 세포 투과성 변화를 crystal violet 흡수율로 관찰한 결과 항효모 물질의 농도가 증가할수록 세포 손상에 따른 crystal violet의 흡수율이 증가하였다. 항효모 물질을 처리한 C. albicans 세포의 형태를 주사전자현미경으로 관찰한 결과 세포벽 또는 세포막의 손상으로 세포 표면이 불규칙한 형태를 나타내었다. 본 연구의 결과 녹차씨 유래의 항효모 활성 사포닌은 천연보존제 및 항진균제 개발을 위한 물질로 활용될 수 있을 것이다. Our previous study showed that green tea seed extract has strong anti-yeast activity. In this study, we isolated an anti-yeast compound from water extract of green tea seeds and investigated its inhibitory effect on Candida albicans cells. Green tea seeds were extracted with water, and water extract was partitioned with chloroform, ethyl acetate, n-butanol, and H2O. The n-butanol fraction showing the highest anti-yeast activity on C. albicans was further fractionated by medium pressure liquid chromatography, after which anti-yeast compound (F4-3) was obtained using high pressure liquid chromatography. Anti-yeast compound in liquid chromatography-tandem mass spectrometry analysis presumed theasaponin E as having a molecular mass of 1,230 kDa. Anti-yeast saponin damaged cell vacuoles of C. albicans, which was analyzed by neutral red staining. Membrane permeability was assessed by crystal violet assay, and C. albicans cells treated with anti-yeast compound exhibited significant uptake of crystal violet in a concentration-dependent manner. The morphological changes of C. albicans cells were observed with a scanning electron microscope. As a result, anti-yeast saponin from green tea seeds is expected to be used as a natural preservative and anti-fungal agent.

        Young-onset type 2 diabetes in South Korea: a review of the current status and unmet need

        The prevalence of young-onset (diagnosis at age < 40 years) type 2 diabetes mellitus (T2DM) is increasing globally. Young-onset T2DM has a common pathophysiology of glucose dysregulation as in late-onset T2DM. However, it presents a greater association with obesity and a more rapid decline in β-cell function than late-onset T2DM. Accumulating evidence indicates that disease progression in young-onset T2DM is rapid, resulting in early and frequent development of microvascular and macrovascular complications, as well as premature death. Improper management and low adherence to medical therapy are important issues in young-onset T2DM. This review discusses the epidemiology, disease entity, and clinical issues associated with young-onset T2DM. We also present the prevalence and clinical characteristics of patients with young-onset T2DM in South Korea.
