Effects of quercetin derivatives from mulberry leaves

        Xufeng Sun,Masayuki Yamasaki,Takuya Katsube,Kuninori Shiwaku 한국영양학회 2015 Nutrition Research and Practice Vol.9 No.2

BACKGROUND/OBJECTIVES: Mulberry leaves contain quercetin derivatives, which have the effects of reducing obesity and improving lipid and glucose metabolism in mice with obesity. It is not clear whether or not mulberry leaves can directly affect metabolic disorders, in the presence of obesity, because of the interaction between obesity and metabolic disorders. The aim of the current study was to assess the direct action of quercetin derivatives on metabolic disorders in non-obese conditions in short-term high-fat diet fed mice. MATERIALS/METHODS: C57BL/6N mice were fed a high-fat diet, supplemented with either 0% (control), 1%, or 3% mulberry leaf powder (Mul) or 1% catechin powder for five days. Anthropometric parameters and blood biochemistry were determined, and hepatic gene expression associated with lipid and glucose metabolism was analyzed. RESULTS: Body and white fat weights did not differ among the four groups. Plasma triglycerides, total cholesterol, and free fatty acids in the 1%, 3% Mul and catechin groups did not differ significantly from those of the controls, however, plasma glucose and 8-isoprostane levels were significantly reduced. Liver gene expression of gp91phox, a main component of NADPH oxidase, was significantly down-regulated, and PPAR-α, related to β-oxidation, was significantly up-regulated. FAS and GPAT, involved in lipid metabolism, were significantly down-regulated, and Ehhadh was significantly up-regulated. Glucose-metabolism related genes, L-PK and G6Pase, were significantly down-regulated, while GK was significantly up-regulated in the two Mul groups compared to the control group. CONCLUSIONS: Our results suggest that the Mul quercetin derivatives can directly improve lipid and glucose metabolism by reducing oxidative stress and enhancing β-oxidation. The 1% Mul and 1% catechin groups had similar levels of polyphenol compound intake (0.4 × 10-5 vs 0.4 × 10-5 mole/5 days) and exhibited similar effects, but neither showed dose-dependent effects on lipid and glucose metabolism or oxidative stress.

Protective effect of cilostazol on vascular injury in rats with acute ischemic stroke complicated with chronic renal failure

        Sun Ru,Gu Qun,Zhang Xufeng,Zeng Ruiqi,Chen Dan,Yao Jingjing,Min Jingjing 한국독성학회 2024 Toxicological Research Vol.40 No.2

Chronic renal failure (CRF) resulting in vascular calcification, which does damage to blood vessels and endothelium, is an independent risk factor for stroke. It has been reported that cilostazol has a protective effect on the focal cerebral ischemic infarct. However, its impact on vascular injury in CRF combined stroke and its molecular protection mechanism have not been investigated. In this study, we carried out the effect of cilostazol on CRF combined stroke rats, and the results confirmed that it improved the neurobehavior, renal function as well as pathologic changes in both the kidney and brain. In addition, the inflammation and oxidative stress factors in the kidney and brain were suppressed. Moreover, the rates of brain edema and infarction were decreased. The injured brain-blood barrier (BBB) was recovered with less Evans blue extravasation and more expressions of zonula occludens-1(ZO-1) and occludin. More cerebral blood flow (CBF) in the ipsilateral hemisphere and more expression of CD31 and vascular endothelial growth factor (VEGF) in brain and kidney were found in the cilostazol group. Furthermore, cell apoptosis and cell autophagy became less, on the contrary, proteins of vascular endothelial growth factor receptor 2 (VEGFR2) after the cilostazol treatment were increased. More importantly, this protective effect is related to the pathway of Janus Kinase (JAK)/signal transducer and activator of transcription 3 (STAT3), mammalian target of rapamycin (mTOR), and the hypoxia inducible factor-1α (HIF-1α). In conclusion, our results confirmed that cilostazol exerted a protective effect on the brain and kidney function, specifically in vascular injury, oxidative stress, cell apoptosis, cell autophagy, and inflammation response in CRF combined with stroke rats which were related to the upregulation of JAK/STAT3/mTOR signal pathway.

Purification of naphthalene from eutectic mixture by continuous column crystallization

        Zhengming Yi,Qunsheng Li,Xufeng Sun,Minggao Su 한국화학공학회 2010 Korean Journal of Chemical Engineering Vol.27 No.2

To obtain highly purified naphthalene, a column crystallizer was employed and operated in the continuous multistage countercurrent fractional mode. The principles and characteristics of the process are introduced. A large amount of experimental work has been done on the crystallization process in a D=50 mm×1,350 mm column. The effects of operation conditions including the composition of feed, reflux ratio, stirring speed and the bed height of crystal on crystallization were systematically studied. The experiments showed that over 99.0% of the purity of product can be easily achieved in such conditions: feed composition among w=85.462-95.894%, reflux ratio among 1.42-4.04,the crystal bed height among 400-900 mm. The purity of product even reaches 99.996% in experiment.

The intrinsic relationship between color variation and performances of the deteriorated aviation lubrication oil

        Jun Ma,Shaojun Ruan,Jianqiang Hu,Yuanbao Sun,Yiwei Fei,Xufeng Jiang,Shibao Dong,Teng Chen,Nan Wu 한국공업화학회 2020 Journal of Industrial and Engineering Chemistry Vol.92 No.-

Dose the blackened lubrication oil needs to be replaced forflying safety? Since there is very limited roomfor inaccurate assessment of oil quality due to strictflying safety standards, much aviation lubrication oilis disposed in advance long before their service life. More important, aircrafts still face the fatalchallenges from unpredictable lubrication oil deterioration. Color change is surely a significant signal to diagnose the quality of lubrication oils. Based on thecolorimeter, the color numbers of the testing lubrication oils are measured in this paper to identify thecolor variation degree of the oxidized lubrication oils. Then the molecular structures of the oxidizedlubrication oils are investigated tofind the intrinsic relationship between the oxidation products andcolor numbers. Based on key performances of the oxidized oil samples, the oil quality is undoubtfullydeteriorated which is coincident with the variation of color number. The color numbers of the usedaviation lubrication oils could provide an alternative way to readily and quickly measure the declinedegree and residual lifetime.

Anti-obesity effects of hot water extract from Wasabi (Wasabia japonica Matsum.) leaves in mice fed high-fat diets

        Masayuki Yamasaki,Tetsuro Ogawa,Li Wang,Takuya Katsube,Yukikazu Yamasaki,Xufeng Sun,Kuninori Shiwaku 한국영양학회 2013 Nutrition Research and Practice Vol.7 No.4

The anti-obesity effects of a hot water extract from wasabi (Wasabia japonica Matsum.) leaves (WLE), without its specific pungent constituents, such as allyl-isothiocyanate, were investigated in high fat-diet induced mice. C57J/BL mice were fed a high-fat diet (control group) or a high-fat diet supplemented with 5% WLE (WLE group). Physical parameters and blood profiles were determined. Gene expression associated with lipid metabolism in liver and white adipose tissue were analyzed. After 120 days of feeding, significantly lower body weight gain, liver weight and epididymal white adipose tissue weight was observed in the WLE group compared to the control group. In liver gene expression within the WLE group, PPARα was significantly enhanced and SREBP-1c was significantly suppressed. Subsequent downstream genes controlled by these regulators were significantly suppressed. In epididymal white adipose tissue of the WLE group, expression of leptin, PPARγ, and C/EBPα were significantly suppressed and adiponectin was significantly enhanced. Acox, related to fatty acid oxidization in adipocytes, was also enhanced. Our results demonstrate that the WLE dietary supplement induces mild suppression of obesity in a high-fat diet induced mice, possibly due to suppression of lipid accumulation in liver and white adipose tissue.
