Clinical Application of Serum Tumor Abnormal Protein (TAP) in Colorectal Cancer Patients

Objective: To explore the association of serum tumor abnormal protein (TAP) with other serological biomarkers e.g. carcinoembryonic antigen (CEA), carbohydrate antigen 125 (CA125), carbohydrate antigen 19-9 (CA19-9) and its clinical application in colorectal cancer (CRC) patients. Methods: Patients (N=98) were enrolled into this study with histologically or cytologically confirmed CRC. Using a test kit, the level of TAP was determined, while chemiluminescence was used to measure the levels of some other common serological biomarkers e.g. CEA, CA125 and CA19-9. Results: The area of TAP condensed particulate matter decreased after chemotherapy compared with before chemotherapy when CT or MRI scans showed disease control. In contrast, it increased with disease progression (P<0.05). Furthermore, a statistically significant difference was confirmed in monitoring of TAP and common serological biomarkers e.g. CEA and CA19-9 (p<0.05). Conclusions: Detecting TAP in CRC patients has high sensitivity and specificity and can be used as a new independent indicator for clinically monitoring CRC patients in the course of chemotherapy.

A new antagonistic strain of actinomycete, designated AP19-2, was isolated from the feces of giant pandas inhabiting the Foping National Nature Reserve in China. Cultural characteristic studies strongly suggested that this strain is a member of the genus Streptomyces. The nucleotide sequence of the 16S rRNA gene of strain AP19-2 evidenced profound similarity (97-99 %) with other Streptomyces strains. Two pure active molecules were isolated from a fermentation broth of Streptomyces sp. strain AP19-2 via extraction, concentration, silica gel G column chromatography, and HPLC. The chemical structures of the two related compounds (referred to as chromomycin $A_2$ and chromomycin $A_3$) were established on the basis of their Infrared spectra (IR), High Resolution Electrospray Ionization Mass Spectrometry (HR-ESI-MS), and Nuclear Magnetic Resonance (NMR) data, and by comparison with published data.

In order to get optimal extraction performance as well as ultra-low solvent residual, three fluorinated hexane, perfluorohexane (C6F14), 1H-perfluorohexane (C6F13H) and 1H,6H-perfluorohexane (C6F12H2), were evaluated as diluents for synergistic effect on extraction capability of 1H,1H,2H,2H-Perfluorooctan-1-ol by the way of experimental and computational investigation. Based on extraction experiments, C6F14 showed obvious antagonistic effect because of hydrophobic nature, while the synergistic effect of C6F12H2 was better than that of C6F13H. According to DFT calculation, supported by FT-IR and 1H NMR spectra, the synergistic effect was illustrated as that fluorinated hexane could interact with salicylic acid-fluorinated octanol complex through hydrogen bond.

Purpose: To highlight the potential factors that could predict the response rate of patients with metastatic colorectal cancer (mCRC) treated with pemetrexed combined chemotherapy after first- or second-line chemotherapy using the FOLFOX regimen. Materials and Methods: Between January 2007 and July 2014, 54 patients diagnosed and pathologically-confirmed with advanced colorectal cancer in Jiangsu Cancer Hospital and Research Institute, were enrolled. They received pemetrexed at a dose of $500mg/m^2$ by 10 minute infusion on day 1, repeated every 3 weeks. Doses were modified depending on nadir counts of blood cells. Combined chemotherapeutic agents included irinotecan, lobaplatin, carboplatin, oxaliplatin, gemcitabine, cis-platinum or bevacizumab. Multiple variables (age, sex, hemoglobin, platinum drugs combined, metastasis sites, LDH, ALP, CEA>40 ug/ml) reported earlier were selected. We used logistic regression analysis to evaluate relationships between these and tumor response. Results: On multivariable analysis, we found that age was significant in predicting the responsiveness to pemetrexed (p<0.05) combined with oxaliplatin. We did not find any other factors which were significantly associated with the response rate to chemotherapy with pemetrexed and irinotecan. Conclusions: By multivariate analysis, we found that age had significant impact on the responsiveness of pemetrexed when combined with oxaliplatin. Additional research based on genomic properties of host and tumors are needed to clarify markers for better selection of patients who could benefit from pemetrexed combined chemotherapy.
Legumain Protein as a Potential Predictive Biomarker for Asian Patients with Breast Carcinoma

Background: Treatment for breast cancer is mainly performed by surgical resection of primary tumors and chemotherapy. However, after tumor invasion and metastases, breast cancer is hard to control. Clarification of the pathogenic mechanisms would be helpful to the prognosis or therapy for the breast cancer. The aim of this study is to investigate the clinical and prognostic implications of legumain protein Materials and Methods: In this study, we examined mastectomy specimens from 114 breast cancer and matching, 26 adjacent non-cancerous tissues using immunohistochemistry. Results: The results indicated that positive expression of legumain protein in breast cancer was 51.8 % (59/114) and the positive expression of legumain protein in adjacent non-cancerous tissue was 11.5% (3/26). It appeared to be related with lymph node metastasis of breast cancer (p=0.02) and correlation analysis indicated that legumain expression was correlated positively with the estrogen receptor (ER) and mutant-type p53 expression (both p<0.05). Positive legumain expression was significantly associated with shorter overall survival time in breast cancer patients (log-rank p<0.01). Multivariate survival analysis suggested that the positive legumain expression was an independent predictor of poorer overall survival in patients with breast cancer (HR=0.24; 95%CI 0.11-0.65, p=0.03). Conclusions: Legumain might be a new potential biomarker for breast cancer, which may reflect the prognosis and overall survival.

The objectives of this study were to study the effects of different levels of dietary copper on organ indexes, tissular Cu, Zn and Fe deposition and fur quality of mink in the growing-furring periods. One hundred and five standard dark male mink were randomly assigned to seven groups with the following dietary treatments: basal diet with no supplemental Cu (Control); basal diet supplemented with either 6, 12, 24, 48, 96 and 192 mg/kg Cu from copper sulphate, respectively. The colour intensity scores displayed a linear trend (P = 0.057). The spleen Cu concentrations responded in a linear (P < 0.05) fashion with increasing level of Cu, but copper supplementation did not affect speen concentrations of Fe or Zn. Supplemental dose of Cu linearly increased (P < 0.05) liver Cu and Fe concentrations but did not alter (P > 0.10) liver Zn. Our results indicate that Cu plays an important role in the pigmentation in growing-furring mink, and supplemental dietary Cu in growing-furring mink improve hair colour, and copper has limited effects on liver mineral deposition.

In order to get optimal extraction performance as well as ultra-low solvent residual, the extraction behavior of tris(2,2,3,3,4,4,4-heptafluorobutyl) phosphate (FTBP) dissolved in 2,3,4,5,6-Pentafluorotoluene (FB) was evaluated to investigate the influence of the aggregation state on the extraction of salicylic acid (SA). The effect of SA distribution and water content on aggregation structure in organic phase was assessed through analysis of SAXS data. Based on molecular dynamic simulation, FTBP tended to aggregate with the existence of FB to accommdate more SA molcules. Water in this case was not the critical factor for aggregation formation due to the hydrophobicity of fluorinated solvents.
