        Improvement of Anti-CD36 Antibody Detection via Monoclonal Antibody Immobilization of Platelet Antigens Assay by Using Selected Monoclonal Antibodies

        Antibodies against human CD36 are responsible for several immune-mediated disorders. The detection of anti-CD36 antibodies using the standard monoclonal antibody (mAb) immobilization of platelet antigens (MAIPA) assay is hampered by a high frequency of false-negative results, most likely due to competitive inhibition of the mAb used as the capture antibody. We generated a panel of mouse mAbs against CD36 and seven hybridomas (GZ-3, GZ-13, GZ-70, GZ-143, GZ-413, GZ-507, and GZ-608), which were selected for MAIPA assays, as they reacted with mouse and human CD36. Fourteen anti-CD36 sera were assayed; all of which showed a positive reaction in a PakPlus (Immucor GTI Diagnostics, Inc., Waukesha, WI, USA) ELISA-based screening (optical density: 0.257–2.292). When the reference anti-CD36 mAb FA6-152 was used in the MAIPA assay, only 6/14 (42.9%) sera displayed a positive reaction. In contrast, anti-CD36 antibodies were detected in 13/14 (92.9%) sera when GZ-70 and GZ-608 mAbs were used. This significant improvement resulted in the identification of anti-CD36 antibodies by an antigen capture assay. Since patient’s platelets possibly carrying rare native antigens are used, this method will facilitate the identification of new platelet antibodies against CD36 that are involved in immune-mediated thrombocytopenia and other diseases, such as transfusion-related acute lung injury.

        Bacterial Community Structure and Function Shift in Rhizosphere Soil of Tobacco Plants Infected by Meloidogyne incognita

        Root-knot nematode disease is a widespread and catastrophic disease of tobacco. However, little is known about the relationship between rhizosphere bacterial community and root-knot nematode disease. This study used 16S rRNA gene sequencing and PICRUSt to assess bacterial community structure and function changes in rhizosphere soil from Meloidogyne incognita-infected tobacco plants. We studied the rhizosphere bacterial community structure of M. incognita-infected and uninfected tobacco plants through a paired comparison design in two regions of tobacco planting area, Yuxi and Jiuxiang of Yunnan Province, southwest China. According to the findings, M. incognita infection can alter the bacterial population in the soil. Uninfested soil has more operational taxonomic unit numbers and richness than infested soil. Principal Coordinate Analysis revealed clear separations between bacterial communities from infested and uninfested soil, indicating that different infection conditions resulted in significantly different bacterial community structures in soils. Firmicutes was prevalent in infested soil, but Chloroflexi and Acidobacteria were prevalent in uninfested soil. Sphingomonas, Streptomyces, and Bradyrhizobium were the dominant bacteria genera, and their abundance were higher in infested soil. By PICRUSt analysis, some metabolism-related functions and signal transduction functions of the rhizosphere bacterial community in the M. incognita infection-tobacco plants had a higher relative abundance than those uninfected. As a result, rhizosphere soils from tobacco plants infected with M. incognita showed considerable bacterial community structure and function alterations.

        In recent years, the power supply of the data center is increasing day by day. The new power supply architecture uses 48V bus to supply power to the subsequent VRM, so as to reduce I2R loss. Because of its soft switching characteristics, LLC resonant converter can meet the requirements of high power density and high efficiency, and is used in isolated DC-DC converter. In this paper, a magnetic integrated transformer is proposed to reduce winding loss and core loss. The windings and magnetic cores are optimized in local detail. A 1500W module was made in the experiment, achieving 97.8% peak efficiency and 97.6% efficiency at full load.

        A Novel Interleaving Control Scheme for Boost Converters Operating in Critical Conduction Mode

        Interleaving techniques are widely used to reduce input/output ripples and to increase the power capacity of boost converters operating in critical conduction mode. Two types of phase-shift control schemes are studied in this paper, the turn-on time shifting method and the turn-off time shifting method. It is found that although the turn-off time shifting method exhibits better performance, it suffers from sub-harmonic oscillations at high input voltages. To solve this problem, an intensive quantitative analysis of the sub-harmonic oscillation phenomenon is made in this paper. Based upon that, a novel modified turn off time shifting control scheme for interleaved boost converters operating in critical conduction mode is proposed. An important advantage of this scheme is that both the master phase and the slave phase can operate stably in critical conduction mode without any oscillations in the full input voltage range. This method is implemented with a FPGA based digital PWM control platform, and tests were carried out on a two-phase interleaved boost PFC converter prototype. Experimental results demonstrated the feasibility and performance of the proposed phase-shift control scheme.

        Soil Layer Disturbance Caused by Pipe Jacking: Measurement and Simulation of a Case Study

        In this paper, areas of soil disturbed by pipe jack construction are categorized and analyzed in detail. Mechanisms of soil disturbance are discussed and patterns of soil deformation are studied using random medium theory. Lateral deformations in deep soil, pore water pressures, stratified settlement, and earth pressures are investigated using measurements from an electrical transmission pipeline project in China. The measurements show that soil layer movement can be predicted by monitoring changes in pore water pressure, and the amplitude of soil disturbance transverse to the pipeline is larger than that above the pipeline in this project. The distributions of stress components in the direction of, and vertical to, the pipeline axis are studied by numerical simulation. It shows that the closer to the pipeline axis, the greater the additional stress, the faster the attenuation speed, and the smaller the range of influence. Relationships of positive propulsion, friction, and stratum loss with longitudinal and transverse surface deformations are also discussed. The ground displacement is a coupling of the three factors to the action of the soil. The surface transverse subsidence with a main influence range of about ±4 D (D is outer diameter of pipe jacking). The findings from this study can be used as technical foundation and reference for the construction of similar projects.

        An Adaptive Fault-tolerant Control Method for Robot Manipulators

        This paper presents a new method for adaptive continuous nonsingular fast terminal sliding mode control (ACNFTSMC) based on a novel structure-improved extended state observer (SIESO) for fault-tolerant control (FTC). In response to the initial peaking phenomenon in traditional ESO (TESO), which can severely degrade the accuracy and stability of the control system, The SIESO is designed to replace TESO to estimate the lump disturbances/faults. Besides, to address the problem of the unknown estimation error of ESO, an adaptive technique is applied to compensate for the observation error in real-time. To guarantee fast convergence and chattering-free, the CNFTSMC method is employed. Afterward, the stability and rapid convergence of the control system is demonstrated using Lyapunov theory. Finally, the simulation results verify the superiority of the proposed control strategy compared to the other existing advanced control techniques.

        Application of different surrogate models on the optimization of centrifugal pump

        An optimization process for impellers was carried out based on numerical simulation, Latin hypercube sampling (LHS), surrogate model and Genetic algorithm (GA) to improve the efficiency of residual heat removal pump. The commercial software ANSYS CFX 14.5 was utilized to solve the Reynolds-averaged Navier-Stokes equations by using the Shear stress transport turbulence model. The impeller blade parameters, which contain the blade inlet incidence angle ∆β, blade wrap angle φ, and blade outlet angle β 2 , were designed by random sample points according to the LHS method. The efficiency predicted under the design flow rate was selected as the objective function. The best combination of parameters was obtained by calculating the surrogate model with the GA. Meanwhile, the prediction accuracies of three surrogate models, namely, Response surface model (RSM), Kriging model, and Radial basis neural network (RBNN), were compared. Results showed that the calculated findings agree with the experimental performance results of the original pump. The RSF model predicted the highest efficiency, while the RBNN had the highest prediction accuracy. Compared with the simulated efficiency of the original pump, the optimization increased efficiency by 8.34% under the design point. Finally, the internal flow fields were analyzed to understand the mechanism of efficiency improvement. The optimization process, including the comparison of the surrogate models, can provide reference for the optimization design of other pumps.

        Pressure Contact Interconnection for High Reliability Medium Power Integrated Power Electronic Modules

        This paper presents a novel spring pressure contact interconnect technique for medium power integrated power electronics modules (IPEMs). The key technology of this interconnection is a spring which is made from Be-Cu alloy. By means of the string pressure contact, sufficient press-contact force and good electrical interconnection can be achieved. Another important advantage is that the spring exhibits excellent performance in enduring thermo-mechanical stress. In terms of manufacture procedure, it is also comparatively simple. A 4 ㎾ half-bridge power inverter module is fabricated to demonstrate the performance of the proposed pressure contact technique. Electrical, thermal and mechanical test results of the packaged device are reported. The results of both the simulation and experiment have proven that a good performance can be achieved by the proposed pressure contact technique for the medium power IPEMs.
