Molecular characterization of chemosensory protein genes in Bactrocera minax (Diptera: Tephritidae)

Chemoreception is of great importance for survival of insects. Chemosensory proteins (CSPs) are believed to be involved in the perireceptor events of chemosensory system in many insect species, but this has not been clarified in the citrus fruit fly, Bactrocera minax, a serious dipteran pest of citrus crops. Previous studies indicated that four CSP genes were identified in many fly speices in Diptera. In the present study, we also identified four CSP genes in B. minax,namely BminCSP1–4, from a transcriptome database. All CSP proteins encoded by these genes bear the typical hallmarks of the CSP family: an N-terminal signal peptide and the four highly conserved cysteine residues. Phylogenetic analysis comparing with other dipteran CSPs indicated that dipteran CSPs may evolved from three ancestral CSP genes, and revealed the sequence diversities of BminCSPs and showed that BminCSP1–4 are clustered in separate groups, indicating the possibility of their contrasting function in B. minax. Furthermore, the tissue distribution of the four CSP genes in adult B. minax was analyzed by real-time quantitiative PCR. The results demonstrated that BminCSP3 are significantly transcriptionally enriched in antennae; BminCSP4 expressed primarily in heads; BminCSP1 and BminCSP2 showed high expressions in different tissues, such as antennae, abdomens and wings. Based on these findings, the different implications for the functions of BminCSPs are discussed. This study will offer a significant indication for further functional studies of the CSPs in B. minax

In older to raise the availability and meet the urgent needs of microcomputer principle teaching, we have developed on MASSCOMP or Massachusetts Computer, a large mainframe multi-user system with up to 64 terminals, Z80 Emulator (Z80SS V1.0) and Advanced Z80 Emulator(Z80SS V2.0) in C language. This paper addresses the struchure, function, effect and technical features of these two systems.
Mechanisms of Extracellular NO and Ca2+ Regulating the Growth of Wheat Seedling Roots

Our previous studies suggested the cross talk of nitric oxide (NO) with Ca2+ in regulating stomatal movement. However, its mechanism of action is not well defined in plant roots. In this study, sodium nitroprusside (SNP, a NO donor) showed an inhibitory effect on the growth of wheat seedling roots in a dose-dependent manner, which was alleviated through reducing extracellular Ca2+ concentration. Analyzing the content of Ca2+ and K+ in wheat seedling roots showed that SNP significantly promoted Ca2+ accumulation and inhibited K+ accumulation at a higher concentration of extracellular Ca2+, but SNP promoted K+ accumulation in the absence of extracellular Ca2+. To gain further insights into Ca2+ function in the NOregulated growth of wheat seedling roots, we conducted the patch-clamped protoplasts of wheat seedling roots in a whole cell configuration. In the absence of extracellular Ca2+,NO activated inward-rectifying K+ channels, but had little effects on outward-rectifying K+ channels. In the presence of 2 mmol L−1 CaCl2 in the bath solution, NO significantly activated outward-rectifying K+ channels, which was partially alleviated by LaCl3 (a Ca2+ channel inhibitor). In contrast,2 mmol L−1 CaCl2 alone had little effect on inward or outward-rectifying K+ channels. Thus, NO inhibits the growth of wheat seedling roots likely by promoting extracellular Ca2+ influx excessively. The increase in cytosolic Ca2+ appears to inhibit K+ influx, promotes K+outflux across the plasma membrane, and finally reduces the content of K+ in root cells.
Theoretical Limits Analysis of Indoor Positioning System Using Visible Light and Image Sensor

To solve the problem of parameter optimization in image sensor-based visible light positioning systems, theoretical limits for both the location and the azimuth angle of the image sensor receiver (ISR) are calculated. In the case of a typical indoor scenario, maximum likelihood estimations for both the location and the azimuth angle of the ISR are first deduced. The Cramer-Rao Lower Bound (CRLB) is then derived, under the condition that the observation values of the image points are affected by white Gaussian noise. For typical parameters of LEDs and image sensors, simulation results show that accurate estimates for both the location and azimuth angle can be achieved, with positioning errors usually on the order of centimeters and azimuth angle errors being less than 1 degree. The estimation accuracy depends on the focal length of the lens and on the pixel size and frame rate of the ISR, as well as on the number of transmitters used.

Objectives: To assess knowledge of HPV and attitudes towards HPV vaccination among the general female population, government officials, and healthcare providers in China to assist the development of an effective national HPV vaccination program. Methods: A cross-sectional epidemiologic survey was conducted across 21 urban and rural sites in China using a short questionnaire. 763 government officials, 760 healthcare providers, and 11,681 women aged 15-59 years were included in the final analysis. Data were analyzed using standard descriptive statistics and logistic regression. Results: Knowledge of HPV among the general female population was low; only 24% had heard of HPV. Less than 20% of healthcare providers recognized sexually na$\ddot{i}$ve women as the most appropriate population for HPV vaccination. There was high acceptance of the HPV vaccine for all categories of respondents. Only 6% of women were willing to pay more than US $300 for the vaccine. Conclusions: Aggressive education is necessary to increase knowledge of HPV and its vaccine. Further proof of vaccine safety and efficacy and government subsidies combined with increased awareness could facilitate development and implementation of HPV vaccination in China.
Overexpression of ENA1 from Yeast Increases Salt Tolerance in Arabidopsis

In yeast, the plasma membrane Na+/H+ antiporter and Na+ -ATPase are key enzymes for salt tolerance. Saccharomyces cerevisiae Na+ -ATPase (Ena1p ATPase) is encoded by the ENA1/PMR2A gene; expression of ENA1 is tightly regulated by Na+ and depends on ambient pH. Although Ena1p is active mainly at alkaline pH values in S. cerevisiae, no Na+ -ATPase has been found in flowering plants. To test whether this yeast enzyme would improve salt tolerance in plants, we introduced ENA1 into Arabidopsis (cv. Columbia) under the control of the cauliflower mosaic virus 35S promoter. Transformants were selected for their ability to grow on a medium containing kanamyin. Southern blot analyses confirmed that ENA1 was transferred into the Arabidopsis genome and northern blot analyses showed that ENA1 was expressed in the transformants. Several transgenic homozygous lines and wild-type (WT) plants were evaluated for salt tolerance. No obvious morphological or developmental differences existed between the transgenic and WT plants in the absence of stress. However, overexpression of ENA1 in Arabidopsis improved seed germination rates and salt tolerance in seedlings. Under saline conditions, transgenic plants accumulated a lower amount of Na+ than did the wild type, and fresh and dry weights of the former were higher. Other experiments revealed that expression of ENA1 promoted salt tolerance in transgenic Arabidopsis under both acidic and alkaline conditions.

D-ribose, a five-carbon sugar with diverse applications, is mainly produced by transketolase (tkt)-deficient Bacillus subtilis (B. Subtilis). We constructed B.subtilis SFR-3A by replacing the corresponding sites of B. subtilis 168 with the mutation site of tkt in the “wild” D-ribose high-producing strain B. subtilis SFR-4, resulting in 5.29 g/L of Dribose. In the meantime, B.subtilis SFR-159 was constructed by completely removing the tkt gene of B. subtilis 168 with a higher production of 6.21 g/L. Through medium optimization, batch fermentation of SFR-3A and SFR-159 gave the best result of 27.56 g/L and 29.89 g/L, which corresponds to productivity of 0.51 g/L/h and 0.41 g/L/h, respectively. In this work, SFR-3A showed more latent capacity over SFR-159 as to productivity and had greater potential to serve as a platform for D-ribose production.
Study on hydrodynamic performance of Heavier-than-water AUV with overlapping grid method

Hydrodynamic coefficients strongly affect the dynamic performance of autonomous underwater vehicles (AUVs). A novel kind of underwater vehicle (Heavier-than-water AUV) with higher density than water is presented, which is different from conventional ones. RANS method and overlapping grids are used to simulate the flow field around the vehicle. Lifts, drags and moments of different attack and drift angles in steady state are calculated. The hydrodynamic performances and how the forces change with the attitude are analyzed according to the flow field structure. The steady-state results using overlapping grid method are compared with those of software FLUENT and wind tunnel tests. The calculation results show that the overlapping grid method can well simulate the viscous flow field around the underwater vehicle. Overlapping grid skills have also been used to figure out the planar-motion-mechanism (PMM) problem of Heavier-than-water AUV and forecast its hydrodynamic performance, verifying its effectiveness in dealing with the dynamic problems, which would be quite helpful for design and control of Heavier-than-water AUV and other underwater vehicles.
