Galectin-9 Acts as a Prognostic Factor with Antimetastatic Potential in Hepatocellular Carcinoma

Considerable research has been conducted concerning galectin-9 and carcinomas, but little information is available about any relation with the hepatocellular carcinoma. In this study, we employed a small interfering RNA (siRNA) targeting galectin-9 to down-regulate the expression in HepG2 cells. As a result, after galectin-9 expression was reduced, cell aggregation was suppressed, while other behaviour such as the proliferation, adhesion and invasion to ECM, cell-endothelial adhesion and transendothelial invasion of the cells were markedly enhanced. When tumors of 200 patients with hepatocellular carcinoma were tested for galectin-9 expression by immunohistochemistry, binding levels demonstrated intimate correlations with the histopathologic grade, lymph node metastasis, vascular invasion and intrahepatic metastasis (P<0.05). Moreover, survival analysis indicated that patients with galectin-9 expression had much longer survival time than those with negative lesions, and the Log-rank test indicated that this difference was statistical significant (P<0.0001). The Cox proportional hazards model suggested that negative galectin-9 expression in hepatocellular carcinoma represented a significant risk factor for patient survival. We propose that galectin-9 might be a new prognostic factor with antimetastatic potential in patients with hepatocellular carcinoma.
Behaviour of micropiles in collapsible loess under tension or compression load

This study examines the behaviour of single micropiles subjected to axial tension or compression load in collapsible loess under in-situ moisture content and saturated condition. Five tension loading tests and five compression loading tests on single micropiles were carried out at a typical loess site of the Loess Plateau in Northwest China. A series of laboratory tests, including grain size distribution, specific gravity, moisture content, Atterberg limits, density, granular components, shear strength, and collapse index, were carried out during the micropile loading tests to determine the values of soil parameters. The loess at the test site poses a severe collapse risk upon wetting. The tension or compression load-displacement curves of the micropiles in loess, under in-situ moisture content or saturated condition, can generally be simplified into three distinct regions: an initial linear, a curvilinear transition, and a final linear region, and the bearing capacity or failure load can be interpreted by the L1-L2 method as done in other studies. Micropiles in loess should be considered as frictional pile foundations though the tip resistances are about 10%-15% of the applied loads. Both the tension and compression capacities increase linearly with the ratio of the pile length to the shaft diameter, L/d. For micropiles in loess under in-situ moisture content, the interpreted failure loads or capacities under tension are 66%-87% of those under compression. However, the prewetting of the loess can lead to the reductions of 50% in the tensile bearing capacity and 70% in the compressive bearing capacity.

This study elucidates the uplift behaviors of the straight-sided and belled shafts. The field uplift load tests were carried out on 18 straight-sided and 15 belled shafts at the three collapsible loess sites under an arid environment on the Loess Plateau in Northwest China. Both the site conditions and the load tests were documented comprehensively. In general, the uplift load-displacement curves of the straight-sided and belled shafts approximately exhibited an initial linear, a curvilinear transition, and a final linear region, but did not provide a well defined peak or asymptotic value of the load, and therefore their uplift resistances should be interpreted from the load test results using an appropriate criterion. Nine representative uplift resistance interpretation criteria were used to define the "interpreted failure load" for each of the load tests, and all of these interpreted uplift resistances were normalized by the failure threshold, $T_{L2}$, obtained using the $L_1-L_2$ method. These load test data were compared statistically and graphically. For the straight-sided and belled shafts, the normalized uplift load-displacement curves were respectively established by the plots that related the mean interpreted uplift resistance ratio against the mean displacement at the corresponding interpreted criteria, and the comparisons of the normalized load-displacement curves were made. Specific recommendations for the designs of uplift belled and straight-sided shafts in the loess were given, in terms of both capacity and displacement.

Three groups of spontaneously hypertensive rats were administered by gavage with distilled water (control group), low-dose green tea (LGT, 0.2 g/kg BW·day) and high-dose green tea (HGT, 1.0 g/kg BW·day) for 4 weeks,during which systolic blood pressure (SBP) was measured weekly by non-invasive tail-cuff method. At the end of experiment, left ventricular hypertrophy index (LHVI) and plasma biochemical indicators were determined, and ultrastructures of myocardium and aortic vascular smooth muscle cells were observed by transmission electron microscopy (TEM). The results show that green tea gavage suppressed the increase in SBP, along with decline in levels of plasma nitric oxide, aldosterone, malondialdehyde, and LHVI, but increased levels of plasma creatinine, calcitonin gene-related peptide, and glutathione peroxidase in a dose dependant manner. Green tea had no effect on plasma total cholesterol. TEM shows that green tea gavage protected mitochondria of left ventricular myocardium and aortic vascular smooth muscle cells from damage.

Twenty novel tetramic acid derivatives (E)-3-(1-(alkyloxyamino)ethylidene)-1-alkylpyrrolidine-2,4-diones were synthesized by the reaction of 3-(1-hydroxyethylidene)pyrrolidine-2,4-diones with O-alkyl hydroxylamines. The title compounds were confirmed by IR, 1H NMR, MS and elemental analysis. The structure of compound 6r was further verified by X-ray diffraction crystallography. The bioassays showed that most of the title compounds exhibited noticeable herbicidal and fungicidal activities.

Roles of Fibroblast Growth Factor-inducible 14 in Hepatocellular Carcinoma

The prognostic value of the fibroblast growth factor-inducible 14 (Fn14) expression in hepatocellular carcinoma (HCC) is unknown. Real-time PCR (RT-PCR), western blot assays and immunohistochemistry analysis were here performed in order to compare Fn14 expressios in paired liver samples of HCC and normal liver tissue. Most of the tumor tissues expressed significantly higher levels of Fn14 compared to adjacent non-tumor tissues, with Fn14High accounting for 54.6% (142/260) of all patients. The Pearson ${\chi}^2$ test indicated that Fn14 expression was closely associated with serum alpha fetal protein (AFP) (P=0.002) and tumor number (p=0.019). Univariate and multivariate analyses revealed that along with tumor diameter and portal vein tumor thrombosis (PVTT ) type, Fn14 was an independent prognostic factor for both overall survival (OS) (HR=1.398, p=0.008) and recurrence (HR=1.541, p=0.001) rates. Fn14 overexpression HCC correlated with poor surgical outcome, and this molecule may be a candidate biomarker for prognosis as well as a target for therapy.
Isolation of L-theanine from Tea Solution by Cation Exchange Resin in Batch and Fixed Bed Column

L-Theanine, a bioactive compound in tea, was isolated from tea solution using cation exchange resin no.732. The adsorption of L-theanine by cation exchange resin no.732 fit the Langmuir isotherm model and was a monolayer molecular interaction process. Thermodynamic studies revealed that the adsorption of L-theanine by resin no.732 was an exothermic and spontaneous physically driven process. The adsorption capacity was influenced by temperature, initial concentration, and pH. The L-theanine adsorption capacity under conditions at room temperature,pH 4.73, and initial L-theanine concentration 18 g/L was 241.731 ± 3.679 mg/g. The Thomas model was fit to describe the column adsorption data at different flow rates and initial concentrations. The L-theanine adsorbed by resin no.732 could be desorbed by 0.134 mol/L Na2HPO4aqueous solution with a recovery rate of 84.96%. These findings indicate that resin no.732 was a promising material for isolating L-theanine from tea solution.
