Interleukin-21 (IL-21)+CD4+ T cells are involved in the immune response against hepatitis B virus (HBV) by secreting IL-21. However, the role of IL-21+CD4+ T cells in the immune response against chronic hepatitis C (CHC) virus infection is poorly understood. This study aimed to investigate the role of IL-21+CD4+ T cells in CHC patients and the potential mechanisms. The study subjects in-cluded nineteen CHC patients who were grouped by viral load (low, < 106 RNA copies/ml, n = 8; high, > 106 RNA copies/ml, n = 11). The peripheral frequency of HCV-specific IL-21+CD4+ T cells was higher in the low viral load group and was negatively correlated with the serum HCV RNA viral load in all CHC patients. Meanwhile, IL-21+ cells accumulated in the liver in the low viral load group. In vitro, IL-21 treatment increased the expression of proliferation markers and cytolytic molecules on HCV-specific CD8+ T cells. In summary, these findings suggest that HCV-specific IL-21+CD4+ T cells might contribute to HCV control by rescuing HCV-specific CD8+ T cells in CHC patients.

Background Antibiotics bear an inherent risk of mitochondrial toxicity due to the structural similarities between mitochondria and bacteria. Drug-induced mitochondrial dysfunction can contribute to organ toxicity and has resulted in a considerable number of market withdrawals. Fluoroquinolones were alerted for hepatotoxicity liability and were recently given black box warnings by FDA for the potential disabling side eff ects including tendonitis, pain in extremities and joints, neuropathies associated with paraesthesia, depression, impairment of memory, hearing, vision, etc. Objective In this study, the potential involvement of mitochondrial impairment in the toxicity of four fl uoroquinolones was investigated with a panel of mechanistic endpoints including ATP, mitochondrial respiration, as well as mitochondrial DNA and protein levels. Results Data revealed mitochondrial toxicity to diff erent extents, induced by the tested fl uoroquinolones and suggested mitochondrial protein synthesis as potential mechanism of action. Conclusion This study exemplifi es an integrated/holistic approach with a selected battery/panel of in vitro assays/endpoints serving to identify mitochondrial toxicity early on in the development of drugs, in particular, antibiotics.
EFFECT OF INITIAL CONTACT LOCATION ON MULTIASPERITY NANOCONTACT: QUASICONTINUUM SIMULATION

Two nanocontact models with di®erent initial contact locations are built to simulate the processof the multiasperity nanocontact for investigating the e®ect of initial contact location on thenanocontact process by using the quasicontinuum method. The indenter is initially located on thetop of the middle wave crest (MWC) of the substrate and the top of the wave trough on the leftside (LWT) of the substrate, respectively. The microscopic deformation mechanism, the load –displacement curve and the nanohardness – displacement curve are examined. It is found that thedeformation mechanisms in the two multiasperity contact models are di®erent. During the initialcontact stage, in the MWC model, the twinning deformation dominates the whole contact pro-cess, while in the LMT model many Lomer-Cottrell locks are generated in the copper substrate,which inhibits the occurrence of twinning deformation.

A simplified approach for the fabrication of CuO/TiO2@C is demonstrated. In this study, low cost CuO/TiO2@C photocatalysts (CSs loading 0.25–2 wt.%) were prepared, characterized and evaluated for gasphaseHg0 oxidation. The obtained CuO/TiO2@C photocatalysts were used to remove gaseous elementalmercury under UV light and LED respectively, and the optimum doping of content of CSs was determined. The results showed that the CuO/TiO2@C photocatalysts were highly efficient in oxidizing Hg0, reachingabove 64% when the CSs doping reached 0.5 wt.%. We have proposed that the probable mechanism bywhich CuO/TiO2@C photocatalysts remove gaseous elemental mercury: with CSs doping, CSs can act assensitizer and transfer electrons to the semiconductors, triggering the formation of very reactive radicalsto improve photocatalytic efficiency.

In recent years, there has been significant research progress on in situ articular cartilage (AC) tissue engineering with endogenous stem cells, which uses biological materials or bioactive factors to improve the regeneration microenvironment and recruit more endogenous stem cells from the joint cavity to the defect area to promote cartilage regeneration.In this study, we used ECM alone as a bioink in low-temperature deposition manufacturing (LDM) 3D printing and then successfully fabricated a hierarchical porous ECM scaffold incorporating GDF-5.Comparative in vitro experiments showed that the 7% ECM scaffolds had the best biocompatibility. After the addition of GDF-5 protein, the ECM scaffolds significantly improved bone marrow mesenchymal stem cell (BMSC) migration and chondrogenic differentiation. Most importantly, the in vivo results showed that the ECM/GDF-5 scaffold significantly enhanced in situ cartilage repair.In conclusion, this study reports the construction of a new scaffold based on the concept of in situ regeneration, and we believe that our findings will provide a new treatment strategy for AC defect repair.

A study was conducted to evaluate effects of alfalfa meal on growth performance and gastrointestinal tract development of growing layer ducks to provide evidence for application of alfalfa meal in the duck industry. Two hundred and fifty-six healthy Shaoxing 7-wk old growing layer ducks were selected and randomly allocated to 1 of 4 dietary treatments based on corn and soybean meal and containing 0, 3, 6, and 9% of alfalfa meal for 8 wks. Each treatment consisted of 4 replicates of 16 ducks each. Briefly, birds were raised in separate compartments, and each compartment consisted of three parts: indoor floor house, adjacent open area and a connecting water area. The results showed: i) Growing ducks fed alfalfa meal diet were not significantly different in average daily gain, feed intake and gain-to-feed ratio from those fed no alfalfa diet (p>0.05). ii) Alfalfa meal increased the ratio crop, gizzard to live weight, caecum to live weight, the caecum index of growing ducks (p<0.05). iii) Villus height in duodenum and jejunum of growing ducks increased significantly with the increase of alfalfa meal levels (p<0.05). Crypt depth in duodenum and jejunum of growing ducks decreased significantly with the increase of alfalfa meal levels (p<0.05). This experiment showed that feeding of alfalfa meal to growing layer ducks could improve gastrointestinal tract growth and small intestinal morphology without effect on performance. This experiment provides evidence that alfalfa meal is a very valuable feedstuff for growing layer ducks.
Analysis of signal cable noise currents in nuclear reactors under high neutron flux irradiation

Cables are indispensable in nuclear power plants for transmitting data measured by various types of detectors, such as self-powered neutron detectors (SPNDs). These cables will generate disturbing signals that must be accurately distinguished and eliminated. Given that the cable current is not very significant, previous research has focused on SPND, with little attention paid to cable evaluation and validation. This paper specifically focuses on the quantitative analysis of cables and proposes a theoretical model to predict cable noise. In this model, the reaction characteristics between irradiated neutrons and cables were discussed thoroughly. Based on the Monte Carlo method, a comprehensive simulation approach of neutron sensitivity was introduced and long-term irradiation experiments in a heavy water reactor (HWR) were designed to verify this model. The theoretical results of this method agree quite well with the experimental measurements, proving that the model is reliable and exhibits excellent accuracy. The experimental data also show that the cable current accounts for approximately 0.2% of the total current at the initial moment, but as the detector gradually depletes, it will contribute more than 2%, making it a non-negligible proportion of the total signal current

We combined real-time RT-PCR and real-time PCR (R/P) assays using a hydrolysis probe to detect Mycobacterium tuberculosis complex (MTBC)-specific 16S rRNA and its rRNA gene (rDNA). The assay was applied to 28 nonrespiratory and 207 respiratory specimens from 218 patients. Total nucleic acids (including RNA and DNA) were extracted from samples, and results were considered positive if the repeat RT-PCR threshold cycle was ≤35 and the ratio of real-time RT-PCR and real-time PCR load was ≥1.51. The results were compared with those from existing methods, including smear, culture, and real-time PCR. Following resolution of the discrepant results between R/P assay and culture, the overall sensitivity, specificity, positive predictive values (PPV), and negative predictive values (NPV) of all samples (including nonrespiratory and respiratory specimens) were 98.2%, 97.2%, 91.7%, and 99.4%, respectively, for R/P assay, and 83.9%, 89.9%, 72.3%, and 94.7%, respectively, for real-time PCR. Furthermore, the R/P assay of four patient samples showed a higher ratio before treatment than after several days of treatment. We conclude that the R/P assay is a rapid and accurate method for direct detection of MTBC, which can distinguish viable and nonviable MTBC, and thus may guide patient therapy and public health decisions.
